Mild brain ischemia produces bladder hyperactivity without brain damage in rats.

INTRODUCTION: The influence of brain ischemia without cerebral infarction on voiding function is unknown. To investigate the effects of a reduction in cerebral blood flow on voiding function, the influence of chronic cerebral hypoperfusion (CH) on bladder activity was examined in rats. MATERIALS AND METHODS: CH was induced in each of 11 female Sprague-Dawley rats by anastomosis between the right external jugular vein and the right common carotid artery with partial obstruction of the left common carotid artery. Twelve intact animals comprised a control group. Voided volume per micturition was…

Abnormal brain activity in prefrontal brain regions in abstinent marijuana users.

We used PET (15)O and a modified version of the Stroop task to determine if 25-day abstinent heavy marijuana (MJ) users have persistent deficits in executive cognitive functioning (ECF) and brain activity. Performance on a modified version of the Stroop task and brain activity was compared between 25-day abstinent, heavy marijuana users (n = 11), and a matched comparison group (n = 11). The 25-day abstinent marijuana users showed no deficits in performance on the modified version of the Stroop task when compared to the comparison group. Despite the lack of performance differences, the marijuan…

Blood-brain barrier abnormalities in vulnerable brain regions during thiamine deficiency.

Experimental thiamine deficiency (TD) is a classical model of metabolic encephalopathy and selective cell loss in the brain resulting from a generalized, low-grade oxidative deficit. Late stages of TD are characterized by hemorrhages in the brain indicating a disruption of the blood-brain barrier (BBB). However, the relation of the breakdown of the BBB to selective cell loss in TD is not understood. The current studies examined the BBB at different stages of TD using immunoglobulin G (IgG) as an indicator of BBB integrity. Adult rats received thiamine-deficient diet ad libitum and daily inject…

[Brain stem AEP in toxic comas. Correlation with brain stem reflexes].

24 deep toxic comas with respiratory assistance were studied by brain-stem reflex and BAEP. Laboratory analysis showed several simultaneous toxics (phenobarbital, benzodiazepines, tricyclic antidepressants...). Three groups of patients were defined: Twelve patients with normal BAEP and with relatively preserved brain-stem reflex (oculocephalic and oculovestibular are often disturbed but photomotor is present). Eleven patients with delayed BAEP and with more disturbed brain-stem reflex (photomotor is missing 3 times). In 2 cases (one of which is mentioned above in group B) brain-stem reflex and…

Brain age gap as a diffusion MRI-based marker of traumatic brain injury-related brain changes and associated outcomes.

Traumatic brain injury is a common neurological disorder and a leading cause of long-term disability, presenting with heterogeneous cognitive, emotional, and functional impairments. A critical clinical challenge is the early identification of patients at risk for persistent symptoms. The Brain Age Gap (BAG), the difference between an individual's predicted brain age from imaging data and their chronological age, has emerged as a potential marker of injury-related brain changes. Here, we aim to evaluate diffusion-MRI-derived BAG for identifying traumatic brain injury patients (Glasgow Coma Scal…

Brain Health and Traumatic Brain Injury (TBI): Relationship Between Brain Care Score and Disability in Subacute-Long Term TBI.

Traumatic brain injury (TBI) is a prevalent, chronic disease with heterogenous recovery and dementia risk. Poor brain health is associated with increased dementia risk, but brain health remains unexplored in TBI. Brain Care Score (BCS), a tool measuring brain health with physical, lifestyle, and social factors, has not been used in TBI. Retrospective cohort study of TBI-clinic visits in the Neurology Traumatic Brain Injury and Concussion Clinic (9/2022-10/2025). Inclusion criteria: ≥18yo, self-reported TBI, BCS, functional outcomes. Disability defined as Glasgow Outcome Scale-Extended (GOSE) <…

Nose-to-Brain Delivery of mRNA-Loaded Lipid Nanoparticles Bypasses the Blood-Brain Barrier for Effective Brain Disease Therapy.

mRNA-loaded lipid nanoparticles (mRNA-LNPs) show great therapeutic potential, but their use in central nervous system (CNS) disorders is limited by poor blood-brain barrier (BBB) penetration. Intranasal (IN) administration can bypass the BBB via olfactory/trigeminal pathways, enabling direct brain targeting and rapid screening of brain-specific lipid nanoparticles (LNPs). Using a peptide-based ionizable lipid platform, we systematically evaluated how LNP surface charge affects IN brain delivery and found that positively charged mRNA-LNPs produced superior brain transfection. Iterative in vivo …

Clinically altered brain activity may not look like aged brain activity: Implications for brain-age modeling and biomarker strategies.

INTRODUCTION: Brain-age gap (BAG), the difference between predicted age and chronological age, is studied as a biomarker for the natural progression of neurodegeneration. The BAG captures brain atrophy as measured with structural magnetic resonance imaging (MRI). Electroencephalography (EEG) has also been explored for estimating the BAG (EEG-BAG). However, studies showed mixed results for the EEG-BAG including counterintuitive findings of younger predicted age in clinical populations, raising doubts about its utility as a clinical tool for mild cognitive impairment (MCI) and Alzheimer's diseas…

Association between mild traumatic brain injury, brain structure, and mental health outcomes in the Adolescent Brain Cognitive Development Study.

BACKGROUND: Children that experience a mild traumatic brain injury (mTBI) are at an increased risk of neural alterations that can deteriorate mental health. We test the hypothesis that mTBI is associated with psychopathology and that structural brain metrics (e.g., volume, area) meaningfully mediate the relation in an adolescent population. METHODS: We analyzed behavioral and brain MRI data from 11,876 children who participated in the Adolescent Brain Cognitive Development (ABCD) Study. Mixed-effects models were used to examine the longitudinal association between mTBI and mental health outco…

Efficient brain delivery and sustained retention of antibodies via dual targeting of blood-brain barrier and intra-brain proteins.

Efficient delivery of therapeutic antibodies to the brain has been increasingly achieved using receptor-mediated transcytosis (RMT) approaches such as transferrin receptor (TfR)-mediated shuttle systems. Moreover, a strategy for enhancing brain retention has recently emerged as a focus of attention to overcome the rapid clearance of TfR shuttles from the brain. Here, we propose a novel approach that combines binding to intra-brain proteins, such as myelin oligodendrocyte glycoprotein (MOG), cell adhesion molecule 3, and chondroitin sulfate proteoglycan 5, for sustained retention in the brain w…

Life history of a brain autoreactive T cell: From thymus through intestine to blood-brain barrier and brain lesion.

Brain antigen-specific autoreactive T cells seem to play a key role in inducing inflammation in the central nervous system (CNS), a characteristic feature of human multiple sclerosis (MS). These T cells are generated within the thymus, where they escape negative selection and become integrated into the peripheral immune repertoire of immune cells. Typically, these autoreactive T cells rest in the periphery without attacking the CNS. When autoimmune T cells enter gut-associated lymphatic tissue (GALT), they may be stimulated by the microbiota and its metabolites. After activation, the cells mig…

Traumatic brain injury and delayed sequelae: a review--traumatic brain injury and mild traumatic brain injury (concussion) are precursors to later-onset brain disorders, including early-onset dementia.

Brain injuries are too common. Most people are unaware of the incidence of and horrendous consequences of traumatic brain injury (TBI) and mild traumatic brain injury (MTBI). Research and the advent of sophisticated imaging have led to progression in the understanding of brain pathophysiology following TBI. Seminal evidence from animal and human experiments demonstrate links between TBI and the subsequent onset of premature, psychiatric syndromes and neurodegenerative diseases, including Alzheimer's disease (AD) and Parkinson's disease (PD). Objectives of this summary are, therefore, to instil…

Hepatic encephalopathy in cirrhotic and portacaval shunted dogs: lack of changes in brain GABA uptake, brain GABA levels, brain glutamic acid decarboxylase activity and brain postsynaptic GABA receptors.

It has been suggested, from studies of a rabbit model of fulminant hepatic failure, that hepatic encephalopathy might be related to an increase in brain gamma-aminobutyric acid uptake through a more permeable bloodbrain barrier, leading to an overactivity of brain gamma-aminobutyric acid-mediated inhibitory neurotransmission. Five groups of dogs were studied: normal dogs, dogs with secondary biliary cirrhosis without and with hepatic encephalopathy and portacaval shunted dogs without and with hepatic encephalopathy. Brain gamma-aminobutyric acid and sucrose uptake was investigated using the mu…

Brain tissue oxygen monitoring in traumatic brain injury and major trauma: outcome analysis of a brain tissue oxygen-directed therapy.

OBJECT: Cerebral ischemia is the leading cause of preventable death in cases of major trauma with severe traumatic brain injury (TBI). Intracranial pressure (ICP) control and cerebral perfusion pressure (CPP) manipulation have significantly reduced the mortality but not the morbidity rate in these patients. In this study, the authors describe their 5-year experience with brain tissue oxygen (PbtO(2)) monitoring, and the effect of a brain tissue oxygen-directed critical care guide (PbtO(2)-CCG) on the 6-month clinical outcome (based on the 6-month Glasgow Outcome Scale score) in patients with T…

Reduced myelinogenesis and recovery in hyperphenylalaninemic rats. Correlation between brain phenylalanine levels, characteristic brain enzymes for myelination, and brain development.

In a previous paper (Burri et al., 1990), we have shown that experimental hyperphenylalaninemia (hyper-Phe) in 3-17 d-old rats leads to reduced myelinogenesis. Such treated rats recover during a 6 w low phenylalanine (Phe) period between days 17 and 59. In order to get more detailed information about the disturbed myelinogenesis and recovery, we measured in hyper-Phe rats the developmental pattern of two brain enzymes typical for myelination, cerebroside sulfotransferase (CST), and 2',3'-cyclic nucleotide 3'-phosphohydrolase (CNP), and other developmental parameters. Further, we correlated bra…

Extracellular vesicles released from endothelial cells of the blood-brain barrier mediate brain Iron accumulation during LPS-induced brain Inflammation.

INTRODUCTION: Brain inflammation leads to an increase in the amount of iron in brain tissue; however, studies do not address the source of the iron that could lead to the accumulation. Most of the brain iron uptake is mediated through the blood-brain barrier (BBB), but studies have not examined whether inflammation increases or decreases iron flux across the BBB. Our recent in vitro study discovered a novel alternate mechanism that iron transport across the BBB is mediated via the extracellular vesicles (EVs). Herein, we investigated the impact of brain inflammation on iron release and iron tr…

Detection of brain metastases from blood using Brain nanoMET sensor: Extracellular vesicles as a dynamic marker for metastatic brain tumors.

Brain metastases account for a significant number of cancer-related deaths with poor prognosis and limited treatment options. Current diagnostic methods have limitations in resolution, sensitivity, inability to differentiate between primary and metastatic brain tumors, and invasiveness. Liquid biopsy is a promising non-invasive alternative; however, current approaches have shown limited efficacy for diagnosing brain metastases due to biomarker instability and low levels of detectable tumor-specific biomarkers. This study introduces an innovative liquid biopsy technique using extracellular vesi…

[Comparative quantitative study on brains of wild and laboratory rats. I. Comparison of volume of total brain and classical brain parts].

The intraspecific allometric relation between brain weight and body weight of wild Norway rats and laboratory rats was gained from sufficient material. The intraspecific allometry exponent a=0.25, being typical for mammals, also came out as a result for Norway and laboratory rats. But laboratory rats have 8.3% less brain substance at a comparable body weight. This decrease in brain size was interpreted as an adaption to the specific conditions of domestication and discussed accordingly. Futhermore the fresh volumes of the fundamental brain parts telencephalon, diencephalon, mesencephalon, cere…

Serum biomarkers after traumatic and hypoxemic brain injuries: insight into the biochemical response of the pediatric brain to inflicted brain injury.

Inflicted traumatic brain injury (iTBI) involves a combination of mechanical trauma and hypoxemia. Serum biomarker concentrations may provide objective information about their relative importance to the pathophysiology of iTBI. We compared the time course of neuron-specific enolase (NSE), S100B and myelin basic protein after pediatric hypoxic-ischemic brain injury, iTBI and noninflicted TBI (nTBI). The time to reach peak concentrations of all three biomarkers was shorter after nTBI. Initial and peak S100B, initial and peak myelin basic protein and peak NSE concentrations were no different betw…
