Listeria monocytogenes use multiple mechanisms to disseminate from the intestinal lamina propria to the mesenteric lymph nodes.

UNLABELLED: Listeria monocytogenes are facultative intracellular bacterial pathogens that cause foodborne disease in humans. The bacteria can use the surface protein InlA to invade intestinal epithelial cells or transcytose across M cells in the gut, but it is not well understood how the bacteria traffic from the underlying lamina propria to the draining mesenteric lymph nodes (MLN). Previous studies indicated that L. monocytogenes associated with both monocytes and dendritic cells in the intestinal lamina propria. We show here that CCR2-/- mice had a significant reduction in Ly6Chi monocytes

CNS-Draining Meningeal Lymphatic Vasculature: Roles, Conundrums and Future Challenges.

A genuine and functional lymphatic vascular system is found in the meninges that sheath the central nervous system (CNS). This unexpected (re)discovery led to a reevaluation of CNS fluid and solute drainage mechanisms, neuroimmune interactions and the involvement of meningeal lymphatics in the initiation and progression of neurological disorders. In this manuscript, we provide an overview of the development, morphology and unique functional features of meningeal lymphatics. An outline of the different factors that affect meningeal lymphatic function, such as growth factor signaling and aging,

A model of in-vivo hydrocephalus shunt dynamics for blockage and performance diagnostics.

The accumulation of excess cerebrospinal fluid in the ventricles of the brain results in hydrocephalus, a condition that is fatal if left untreated. The usual remedy is to insert a shunt into the ventricles of the brain, which drains excess fluid away, moderated by a pressure dependent valve. It is important that the system functions properly so that a reasonable intracranial pressure is maintained. Unfortunately, pressure measurements in the ventricles are highly invasive, while pressure measurements in the shunt outside the skull may not detect any blockage in the catheter inside. Here we de

Emerging Roles of Meningeal Lymphatic Vessels in Ischemic Stroke.

This review highlights the emerging relevance of meningeal lymphatic vessels (MLVs) in the context of ischemic stroke, challenging the conventional view of a privileged immunological central nervous system. MLVs facilitate immunological surveillance by modulating the entry of peripheral immune cells into the meningeal compartment, a process not impeded by the blood-brain barrier. In ischemic stroke, these vessels play a crucial role in the neuroinflammatory cascade, contributing to immune responses by draining antigens and signals to cervical lymph nodes. Their involvement extends to potential

"Unusual brain stone": heavily calcified primary neoplasm with some features suggestive of angiocentric glioma.

This 40-year-old man presented with a 5-month history of progressive right-sided headache associated with visual blurring. He also had a history of epilepsy but had been seizure free with medication for the past 10 years. An initial CT scan of his brain performed 16 years previously had revealed a small area of calcification in the right parietal region. In the current presentation, he had a left-sided homonymous hemianopia but no other neurological deficits. A CT scan of his brain showed a much larger calcified, partly cystic lesion in the right parietal region. Because he was symptomatic, th

[Brain abscess caused by Haemophilus influenzae type E in a pediatric patient suffering from Apert syndrome].

We report a case of a brain abscess caused by Haemophilus influenzae type e in a 12 year-old patient suffering from Apert syndrome. Apert syndrome is characterized by the premature closure of cranial sutures. In 2010 the patient suffered head trauma in the frontal area with cranial fracture and a cerebrospinal fluid fistula. In February 2013 he was admitted to hospital with fever, vomiting and generalized tonic-clonic seizure with deteriorating mental status/progressive sensory impairment. The computerized axial tomographic scan showed a right frontal lesion, perilesional edema, mild ventricul

CCN1 Is a Therapeutic Target for Reperfused Ischemic Brain Injury.

Ischemic stroke can lead to systemic inflammation, which can activate peripheral immune cells, causing neuroinflammation and brain injury. Meningeal lymphatics play a crucial role in transporting solutes and immune cells out of the brain and draining them into cervical lymph nodes (CLNs). However, the role of meningeal lymphatics in regulating systemic inflammation during the reperfusion stage after ischemia is not well understood. In this study, we demonstrated that brain infarct size, neuronal loss, and the effector function of inflammatory macrophage subsets were reduced after ischemia-repe

The effects of ventricle geometries and boundary conditions on computational modeling of ventriculoperitoneal catheters.

Hydrocephalus is a condition where an excess amount of cerebrospinal fluid (CSF) accumulates within the ventricles of the brain, leading to elevated intracranial pressure. The most common treatment is the surgical placement of a brain shunt to drain CSF from the ventricles. However, brain shunts have extremely high failure rates and improved devices are needed to minimize device obstruction and failure. To help establish modeling standards for this complex scenario, this work computationally investigates geometric and fluid dynamic variables to determine their effects on shunt performance, in

Incidence, predictors, and impact of cerebral edema during the early brain injury phase of aneurysmal subarachnoid hemorrhage: A retrospective cohort study.

BACKGROUND: Early brain injury (EBI) is a common and serious complication of aneurysmal subarachnoid hemorrhage (aSAH) that occurs within the first 72 h after the event. Burden of cerebral edema during EBI (early cerebral edema [ECE]) and its impact on patient outcomes remains to be completely explored. The primary objective of our study was to determine the incidence and identify the predictors of ECE after aSAH using the Subarachnoid Hemorrhage Early Brain Edema Score (SEBES). METHODS: This retrospective study included adult patients with aSAH of any severity grade presenting within 72 h of

Decompressive Craniectomy and Shunt-Amenable Post-Traumatic Hydrocephalus: A Single-Center Experience.

BACKGROUND: Prior studies have shown that decompressive craniectomy may be an independent risk factor for the development of post-traumatic hydrocephalus (PTH). It is upon this background that we chose to conduct our single-center retrospective study to establish the possibility of an association between decompressive craniectomy and PTH. METHODS: A retrospective review involving a database of all patients with traumatic brain injury was undertaken. All referrals and admissions with traumatic brain injury, as defined by the Mayo Classification, from January 2012 to May 2022, were included in

CNS: Not an immunoprivilaged site anymore but a virtual secondary lymphoid organ.

The cardinal dogma of central nervous system (CNS) immunology believed brain is an immune privileged site, but scientific evidences gathered so far have overturned this notion proving that CNS is no longer an immune privileged site, but rather an actively regulated site of immune surveillance. Landmark discovery of lymphatic system surrounding the duramater of the brain, made possible by high resolution live imaging technology has given new dimension to neuro-immunology. Here, we discuss the immune privilege status of CNS in light of the previous and current findings, taking into account the d

AlGaN/GaN heterojunction field-effect transistors for suppressing optical artifacts from integrated light-emitting diodes.

The integrated photoelectrode probe capable of simultaneously stimulating neurons with light and collecting electrophysiological signals is the core device of optogenetics. In this paper, AlGaN/GaN heterojunction field-effect transistors (HFETs) are used instead of the traditional metal microelectrode to prepare brain neural probe, and then bonded with the LED photoelectrode probe to form an integrated photoelectrode probe. The AlGaN/GaN HFET structure brain neural probe has high sensitivity and biocompatibility, which can effectively improve the quality of neuronal signal acquisition. While t

Cerebral Vessels: An Overview of Anatomy, Physiology, and Role in the Drainage of Fluids and Solutes.

The cerebral vasculature is made up of highly specialized structures that assure constant brain perfusion necessary to meet the very high demand for oxygen and glucose by neurons and glial cells. A dense, redundant network of arteries is spread over the entire pial surface from which penetrating arteries dive into the cortex to reach the neurovascular units. Besides providing blood to the brain parenchyma, cerebral arteries are key in the drainage of interstitial fluid (ISF) and solutes such as amyloid-beta. This occurs along the basement membranes surrounding vascular smooth muscle cells, tow

Intra-operative cerebrospinal fluid sampling versus post-operative lumbar puncture for detection of leptomeningeal disease in malignant paediatric brain tumours.

INTRODUCTION: Leptomeningeal disease is a feared sequelae of malignant paediatric brain tumours. Current methods for its detection is the combined use of cranio-spinal MRI, and CSF cytology from a post-operative lumbar puncture. In this study, the authors hypothesize that CSF taken at the start of surgery, either from an external ventricular drain or neuroendoscope will have equal sensitivity for positive tumour cells, in comparison to lumbar puncture. Secondary hypotheses include positive correlation between CSF cytology and MRI findings of LMD. From a clinical perspective, the key aim of the

Pediatric Cerebral Venous Sinus Thrombosis: A Case Report.

Cerebral venous sinus thrombosis (CVST) in infants is a rare vascular disorder that presents with nonspecific symptoms leading to a delay in diagnosis and treatment. Thrombus formation in the cerebral sinuses prevents blood from draining out of the brain leading to local and systemic complications. Here, we present an 11-week-old patient who presented to the emergency department (ED) with three days of lethargy, multiple episodes of projectile emesis, increased fussiness, and downward gaze. A CT scan demonstrated intraventricular hemorrhage (IVH) with acute hydrocephalus. A CT venogram of the

Treatment of hydrocephalus with high-pressure valve ventriculoperitoneal shunt in a dog.

A 5-month-old male Maltese with right-sided circling, deafness, and blindness was presented. A diagnosis of communicating hydrocephalus was made. A ventriculoperitoneal shunt was implanted and the cerebrospinal fluid was drained by using an adjustable valve type (Medtronic Strata). The valve was set at 2.5 (135-155 mmH2O). This was done to prevent the possibility of an overdrainage-induced collapse of the brain parenchyma, which can occur rarely when canine hydrocephalus is treated by using a low-pressure valve. Computed tomography performed 6 weeks and 1 year after surgery revealed the ventri

SEM study of the choroid plexus of the lateral ventricle in the cat.

Corrosion casts of the complete vascular network of the choroid plexus of the lateral ventricle in the cat brain were studied in SEM using the injection-replication method. The villi of this plexus are located only on its supero-anterior and infero-posterior parts, being most densely packed in the former region, close to the interventricular foramen. The capillaries of the villi display small nodular thickenings, which suggest the presence of small, sinusoidal dilatations. The main vessel supplying the plexus with blood is the anterior choroidal artery. The plexus is also characterized by a pa

Current management of traumatic intracranial hypertension: What you need to know.

Intracranial hypertension (ICH) has long been recognized as a major challenge in the management of patients with severe traumatic brain injury. Although it has become widely accepted that patients with unremarkable brain computed tomography scans may not require invasive intracranial pressure (ICP) monitoring, the threshold for using such monitoring in other severe traumatic brain injury patients should be low. Intraparenchymal monitors and external ventricular drains are both effective monitoring techniques. No specific prophylactic interventions are available to prevent ICH. Instead, initial

Management of bacterial central nervous system infections.

Bacterial infections of the central nervous system present as a medical emergency, thus requiring rapid diagnosis and immediate treatment. The most prevalent bacterial infections seen in the intensive care unit can be summarized as acute bacterial meningitis, subdural empyema, intracerebral abscess, and ventriculitis, which all commonly involve the brain parenchyma. The infections can either be community-acquired or hospital-acquired, e.g., after neurosurgical intervention, as a complication of severe neurotrauma or related to indwelling cerebrospinal fluid drains.
