A calcium-independent CDPK positively regulates drought resistance in maize by modulating the ABA signaling pathway.

Calcium-dependent protein kinases (CDPKs/CPKs) play crucial roles in plant responses to drought stress. In this study, we reveal that ZmCPK1, an atypical CDPK whose kinase activity is calcium-independent, acts as a positive regulator of drought resistance in maize. ZmCPK1 is a constitutively active kinase interacting with and phosphorylates multiple components of the abscisic acid (ABA) signaling pathway, including ZmSLAC1, ZmSnRK2.10, and ZmPP2C11. In the absence of ABA, ZmCPK1 may phosphorylate ZmSnRK2.10 and the N-terminus of ZmSLAC1. ZmPP2C11 reverses this phosphorylation by directly depho

Exogenous Vitamin D Mitigates Salt Stress in Arabidopsis thaliana via Ion Homeostasis and Calcium-Mediated Signaling.

Understanding the physiological, biochemical, and molecular responses of Arabidopsis thaliana to salt stress and the potential of exogenous vitamin D application to alleviate these responses is critical for developing effective biostimulant strategies under increasing soil salinization. However, knowledge about how vitamin D2 and vitamin D3 mediate salt stress responses at the molecular level remains limited. The physiological, biochemical, and molecular responses of A. thaliana Col-0 seedlings treated with exogenous vitamin D2 (50 ppm) and vitamin D3 (75 ppm) under 100 mM NaCl salt stress wer

TRP-Dependent Calcium Regulation in HCEC-12 Cells: Involvement of Ascorbic Acid and Cannabinoid Receptor Signaling.

The human corneal endothelium (HCE) is critical for maintaining corneal transparency. Dysfunctions due to cell loss are linked to altered intracellular calcium ([Ca2+]i) homeostasis. Transient receptor potential channels (TRPs) are key regulators of [Ca2+]i, and both L-ascorbic acid (Asc) and cannabinoid receptor (CB) agonists have been implicated in modulating TRP activity. This study investigated the effects of 1 mM Asc and the CB agonist WIN 55,212-2 (WIN) (10 µM) on [Ca2+]i regulation in human corneal endothelial cells (HCECs). HCEC-12 was used as the established HCE cell model. [Ca2+]i dy

Ocean acidification disrupts the biomineralization process in the oyster Crassostrea virginica via intracellular calcium signaling dysregulation.

Calcium is a key component in the shell and skeleton structure, serving as a second messenger for regulating biomineralization across many species. Ocean acidification (OA) is well-studied for causing shell dissolution in marine bivalve species by disordering calcium deposition. However, the regulatory pathway of calcification affected by OA remains unclear. This study assessed eastern oyster (Crassostrea virginica) to determine how calcium signaling responds to elevated pCO2 and influences shell formation. Under elevated pCO2, increased calcium influx was found in mantle epithelial cells, fol…

Protocatechualdehyde-functionalized BMSC-derived exosomes orchestrate calcium signaling in the immunoinflammatory microenvironment to promote periodontal regeneration.

The significant global burden of periodontitis has increased over the past few decades. However, the regeneration and reconstruction of periodontal tissue remain major challenges in clinical practice. Successful periodontal treatment relies on the regulation of calcium signaling. Bone mesenchymal stem cell-derived exosomes (BMSC Exo) hold great potential for promoting calcium influx to enhance osteogenic differentiation, modulate the inflammatory microenvironment, and promote angiogenesis, but their therapeutic efficiency is limited by inadequate stability. To overcome this problem, a mild and…

Aquorin Bioluminescence-Based Ca2+ Imaging Reveals Differential Calcium Signaling Responses to Abiotic Stresses in Physcomitrella patens.

Calcium ions (Ca2+) are an important secondary messenger in plant signal transduction networks. The cytosolic free Ca2+ concentration ([Ca2+]i) of plants changes rapidly when they are subjected to different abiotic stresses, which drives calcium signaling. Although this process has been extensively studied in spermatophytes, the details of calcium signaling in bryophytes remains largely unknown. In our study, we reconstituted aequorin in the bryophyte Physcomitrella patens, optimized the percentage of ethanol in the Ca2+ discharging solution, and measured the [Ca2+]i changes induced by differe…

An activating calcium-sensing receptor variant with biased signaling reveals a critical residue for Gα11 coupling.

Autosomal dominant hypocalcemia (ADH) is due to enhanced calcium-dependent signaling caused by heterozygous gain-of-function (GOF) variants in the CASR gene (ADH1) or in the GNA11 gene, encoding Gα11 (ADH2). Both ADH1 and ADH2 are associated with hypocalcemia and normal or inappropriately low levels of circulating PTH. ADH1 patients typically manifest hypercalciuria, while ADH2 is associated with short stature in approximately 42% of cases. We evaluated a 10-yr-old boy with hypoparathyroidism and short stature. Biochemical analyses revealed hypocalcemia, hyperphosphatemia, and inconsistent hyp

Aberrant evoked calcium signaling and nAChR cluster morphology in a SOD1 D90A hiPSC-derived neuromuscular model.

Familial amyotrophic lateral sclerosis (ALS) is a progressive neuromuscular disorder that is due to mutations in one of several target genes, including SOD1. So far, clinical records, rodent studies, and in vitro models have yielded arguments for either a primary motor neuron disease, or a pleiotropic pathogenesis of ALS. While mouse models lack the human origin, in vitro models using human induced pluripotent stem cells (hiPSC) have been recently developed for addressing ALS pathogenesis. In spite of improvements regarding the generation of muscle cells from hiPSC, the degree of maturation of

Intracellular cAMP signaling-induced Ca2+ influx mediated by calcium homeostasis modulator 1 (CALHM1) in human odontoblasts.

In odontoblasts, intracellular Ca2+ signaling plays key roles in reactionary dentin formation and generation of dentinal pain. Odontoblasts also express several Gs protein-coupled receptors that promote production of cyclic AMP (cAMP). However, the crosstalk between intracellular cAMP and Ca2+ signaling, as well as the role of cAMP in the cellular functions of odontoblasts, remains unclear. In this study, we measured intracellular cAMP levels and intracellular free Ca2+ concentration ([Ca2+]i). We also investigated the effect of intracellular cAMP on mineralization by the odontoblasts. Parathyroid Hormone Promotes Human Umbilical Vein Endothelial Cell Migration and Proliferation Through Orai1-Mediated Calcium Signaling.

Parathyroid hormone is the main endocrine regulator of extracellular calcium and phosphorus levels. Secondary hyperparathyroidism-induced endothelial dysfunction may be related to calcium homeostasis disorders. Here, we investigated the effects of parathyroid hormone on human umbilical vein endothelial cells (HUVECs) and characterized the involvement of store-operated Ca2+ entry (SOCE) and the nuclear factor of activated T cells (NFAT) signaling pathway. We used immunoblot experiments to find that parathyroid hormone significantly enhanced the expression of the Orai1 channel, a type of channel

Bitter taste receptors stimulate phagocytosis in human macrophages through calcium, nitric oxide, and cyclic-GMP signaling.

Bitter taste receptors (T2Rs) are GPCRs involved in detection of bitter compounds by type 2 taste cells of the tongue, but are also expressed in other tissues throughout the body, including the airways, gastrointestinal tract, and brain. These T2Rs can be activated by several bacterial products and regulate innate immune responses in several cell types. Expression of T2Rs has been demonstrated in immune cells like neutrophils; however, the molecular details of their signaling are unknown. We examined mechanisms of T2R signaling in primary human monocyte-derived unprimed (M0) macrophages (M[For

Dexamethasone-induced cardiac deterioration is associated with both calcium handling abnormalities and calcineurin signaling pathway activation.

Dexamethasone is a potent and widely used anti-inflammatory and immunosuppressive drug. However, recent evidences suggest that dexamethasone cause pathologic cardiac remodeling, which later impairs cardiac function. The mechanism behind the cardiotoxic effect of dexamethasone is elusive. The present study aimed to verify if dexamethasone-induced cardiotoxicity would be associated with changes in the cardiac net balance of calcium handling protein and calcineurin signaling pathway activation. Wistar rats (~400 g) were treated with dexamethasone (35 µg/g) in drinking water for 15 days. After dex

Dysregulation of mitochondrial calcium signaling and superoxide flashes cause mitochondrial genomic DNA damage in Huntington disease.

Huntington disease (HD) is an inherited, fatal neurodegenerative disorder characterized by the progressive loss of striatal medium spiny neurons. Indications of oxidative stress are apparent in brain tissues from both HD patients and HD mouse models; however, the origin of this oxidant stress remains a mystery. Here, we used a yeast artificial chromosome transgenic mouse model of HD (YAC128) to investigate the potential connections between dysregulation of cytosolic Ca(2+) signaling and mitochondrial oxidative damage in HD cells. We found that YAC128 mouse embryonic fibroblasts exhibit a strik

Semaphorin3A facilitates axonal transport through a local calcium signaling and tetrodotoxin-sensitive voltage-gated sodium channels.

Semaphorin3A (Sema3A), a secreted factor that navigates axons and dendrites of developing neurons, facilitates axonal transport. However, little is known about the mechanism underlying Sema3A-induced facilitation and its functional implications. Here we show that Sema3A induces facilitation of axonal transport via local calcium signaling in growth cone. The facilitation of axonal transport was blocked by inhibitors of voltage-gated sodium channels (tetrodotoxin, TTX), L-type voltage-gated calcium channel, and ryanodine receptor (RyR). Sema3A evoked intracellular Ca(2+) elevation in growth cone…

Chromogranin B regulates calcium signaling, nuclear factor kappaB activity, and brain natriuretic peptide production in cardiomyocytes.

Altered regulation of signaling pathways can lead to pathologies including cardiac hypertrophy and heart failure. We report that neonatal and adult cardiomyocytes express chromogranin B (CGB), a Ca(2+) binding protein that modulates Ca(2+) release by the inositol 1,4,5-trisphosphate receptor (InsP(3)R). Using fluorescent Ca(2+) indicator dyes, we found that CGB regulates InsP(3)-dependent Ca(2+) release in response to angiotensin II, an octapeptide hormone that promotes cardiac hypertrophy. ELISA experiments and luciferase reporter assays identified angiotensin II as a potent inducer of brain

Abscisic acid (ABA) inhibits light-induced stomatal opening through calcium- and nitric oxide-mediated signaling pathways.

Nitric oxide (NO) is an important signaling component of ABA-induced stomatal closure. However, only fragmentary data are available about NO effect on the inhibition of stomatal opening. Here, we present results supporting that, in Vicia faba guard cells, there is a critical Ca2+-dependent NO increase required for the ABA-mediated inhibition of stomatal opening. Light-induced stomatal opening was inhibited by exogenous NO in V. faba epidermal strips. Furthermore, ABA-mediated inhibition of stomatal opening was blocked by the specific NO scavenger cPTIO, supporting the involvement of endogenous

TLR2 signaling in chondrocytes drives calcium pyrophosphate dihydrate and monosodium urate crystal-induced nitric oxide generation.

Microcrystals of calcium pyrophosphate dihydrate (CPPD) and monosodium urate (MSU) deposited in synovium and articular cartilage initiate joint inflammation and cartilage degradation in large part by binding and directly activating resident cells. TLRs trigger innate host defense responses to infectious pathogens, and the expression of certain TLRs by synovial fibroblasts has revealed the potential for innate immune responses to be triggered by mesenchymally derived resident cells in the joint. In this study we tested the hypothesis that chondrocytes also express TLRs and that one or more TLRs
