Increased vascular permeability was studied in the synovium of the rat's stifle joint following mechanical trauma and haemarthrosis. The abnormal permeability was detected by injecting colloidal carbon intravenously and examining the synovial vessels for intra-mural deposits of carbon. The effects of rotational and stretch injuries were minimal except when accompanied by intra-synovial haemorrhage. Injection of autologous whole blood into the joint evoked a marked venular permeability response which persisted for 24 hours. These findings indicate that the major factor in the formation of traum

BACKGROUND: Inflammation is the response of the immune system against harmful stimuli in tissues. Leukocyte extravasation or TransEndothelial Migration (TEM) is a crucial step during inflammation, in which leukocytes migrate over the endothelial barrier toward the damaged tissue.
OBJECTIVE: Historically, it was believed that leukocyte TEM directly causes excessive vascular leakage, resulting in tissue edema. However, it is now clear that leukocyte TEM and vascular leakage are uncoupled events with different spatiotemporal regulation. Moreover, several mechanisms have been identified that prev

BACKGROUND: Endothelial cell (EC) activation, endotheliitis, vascular permeability, and thrombosis have been observed in patients with severe coronavirus disease 2019 (COVID-19), indicating that the vasculature is affected during the acute stages of SARS-CoV-2 infection. It remains unknown whether circulating vascular markers are sufficient to predict clinical outcomes, are unique to COVID-19, and if vascular permeability can be therapeutically targeted.
METHODS: Prospectively evaluating the prevalence of circulating inflammatory, cardiac, and EC activation markers as well as developing a mic

BACKGROUND: The atrial natriuretic peptide (ANP) released from the heart regulates intravascular volume and is suspected to increase capillary permeability. Contradictory results regarding ANP and glycocalyx degradation have been reported. The aim of this study was to investigate if an infusion of ANP causes degradation of the endothelial glycocalyx.
METHODS: Twenty pigs, pretreated with 250 mg methylprednisolone, were randomized to receive an infusion of either ANP (50 ng/kg/min) (n = 10) or 0.9% NaCl (n = 10) during 60 min. Endothelial glycocalyx components (heparan sulphate proteoglycan an…

OBJECTIVE: Fluid and protein continuously transude from the surface of the liver. Despite a common understanding that transudation plays a critical role in hepatic interstitial and peritoneal fluid balance, transudation from the entire liver has not been studied. Therefore, the goal of the present work was to provide the first direct measurement of the hepatic transudation rate and transudation barrier properties.
METHODS: Transudation rates were determined by collecting transudate from the entire liver. Hydraulic conductivity, and fluid transudation and protein reflection coefficients of the

Oral, intestinal and genital mucosal epithelia have a barrier function to prevent paracellular penetration by viral, bacterial and other pathogens, including human immunodeficiency virus (HIV). HIV can overcome these barriers by disrupting the tight and adherens junctions of mucosal epithelia. HIV-associated disruption of epithelial junctions may also facilitate paracellular penetration and dissemination of other viral pathogens. This review focuses on possible molecular mechanisms of HIV-associated disruption of mucosal epithelial junctions and its role in HIV transmission and pathogenesis of

The role of platelets in hemostasis is to produce a plug to arrest bleeding. During thrombocytopenia, spontaneous bleeding is seen in some patients but not in others; the reason for this is unknown. Here, we subjected thrombocytopenic mice to models of dermatitis, stroke, and lung inflammation. The mice showed massive hemorrhage that was limited to the area of inflammation and was not observed in uninflamed thrombocytopenic mice. Endotoxin-induced lung inflammation during thrombocytopenia triggered substantial intra-alveolar hemorrhage leading to profound anemia and respiratory distress. By im

Isocapnic dry gas hyperpnea causes bronchoconstriction in guinea pigs that is mediated by release of tachykinins from airway sensory nerves. Exogenous neuropeptides can induce microvascular leak. Therefore we tested whether dry gas hyperpnea also elicits bronchovascular hyperpermeability by measuring Evans blue-labeled albumin extravasation along the airways of mechanically ventilated guinea pigs. We found that 1) room temperature dry gas hyperpnea increased Evans blue extravasation in extrapulmonary and intrapulmonary airways as a specific consequence of local airway heat/water losses, 2) cap

Intestinal hypersensitivity reactions during parasitic helminth infections are T-cell-dependent. Little is known about the characteristics of T-cells that initiate and regulate intestinal inflammation or about how they do this. In this paper data are presented that indicate that during Trichinella spiralis infections in mice T-cells regulate intestinal inflammation in the same way that they regulate delayed-type hypersensitivity reactions in the skin and cellular immune responses against tumors.
