Chemogenetic studies using Designer Receptors Exclusively Activated by Designer Drugs (DREADDs) enable the precise manipulation of neuronal activity in specific brain regions and cell types. DREADDs are widely used to dissect neural circuits underlying animal behavior, including learning and memory. Clozapine-N-Oxide (CNO), a metabolite of clozapine and one of the earliest-developed ligands for muscarinic DREADDs, was initially considered pharmacologically inert. However, CNO is now known to undergo back-metabolism to clozapine, leading to undesired off-target behavioral effects, including alt

BACKGROUND: Chronic pain is a prevalent condition often associated with cognitive impairments, including memory deficits. The dorsal raphe nucleus (DRN) is implicated in both pain processing and memory regulation, yet the role of DRN dopaminergic (DAergic) neurons in chronic pain-induced memory impairment remains unclear. This study aimed to investigate whether DRN-DA neurons and their projections to the bed nucleus of the stria terminalis (BNST) contribute to memory deficits in a mouse model of chronic neuropathic pain.
METHODS: Chronic constrictive injury (CCI) was induced in male C57BL/6J

OBJECTIVES: Type 2 diabetes mellitus (T2DM) is a multisystem disorder characterized by metabolic dysregulation and cognitive decline. While hippocampal dysfunction is a recognized feature of cognitive impairment, upstream neural circuits that couple peripheral metabolic signals to central cognition remain poorly defined. This study aimed to determine whether the lateral posterior thalamic nucleus-hippocampal glutamatergic (LP-HPCGlu) circuit mediates the intersection of metabolic and cognitive deficits in T2DM.
METHODS: A T2DM mouse model was established using a high-fat diet (HFD) and low-do

BACKGROUND: Depression is frequently accompanied by gastrointestinal disturbances, reflecting the close interplay between emotional regulation and gut physiology. The central amygdala (CeA), particularly its GABAergic neurons, has been identified as a critical hub within the brain-gut axis. However, the mechanisms by which CeA inhibitory neurons mediate comorbid depressive and gastrointestinal symptoms remain poorly understood, and their potential as therapeutic targets has not been fully explored.
AIM: To investigate CeA GABAergic mechanisms linking mood and gastric function and to appraise

In PTSD, microglia are more engaged in fear-related circuits, where they participate in fear memory processing responses. The reconsolidation-impairing effect of Δ⁹-tetrahydrocannabinol (THC), the primary psychoactive constituent of Cannabis, is typically linked to cannabinoid type-1 receptor (CB1) signaling. THC also engages peroxisome proliferator-activated receptor gamma (PPARγ), and both CB1 and PPARγ modulate neuroimmune processes, including microglial activity. This dual mechanism suggests that THC's impact on fear memory reconsolidation may extend beyond classical cannabinoid signaling.

Previous studies have shown that dorsal vagal complex (DVC) astrocytes play important roles in homeostatic regulation of food intake and caloric balance, specifically upregulation of NMDA receptor-mediated glutamatergic signaling to brainstem dorsal motor nucleus of the vagus (DMV) motoneurons restores caloric balance following exposure to caloric dense diets. DMV neurons are critical to the regulation of gastric functions, including motility, tone, and emptying; hence food intake and energy homeostasis. Prior studies have also shown that caloric intake in female rats fluctuates across the est

OBJECTIVE: Previous studies have indicated that ferulic acid possesses sedative and hypnotic functions. As a derivative of ferulic acid, ferulic acid methylester (FAM) exhibits stronger activity and lower toxicity than ferulic acid. This study is intended to establish a rat model of insomnia induced by PCPA, with the aim of exploring the improvement effect of FAM on the comorbidity of anxiety and insomnia in PCPA-induced insomnia model rats and its underlying mechanisms.
METHODS: Insomnia models were established by intraperitoneal injection of 400 mg/kg p-chlorophenylalanine (PCPA) in SD rats

AIMS: Heavy alcohol use during adolescence significantly impacts cognitive function in adulthood. However, the neural mechanisms underlying adolescent alcohol exposure-induced adult cognitive deficits remain unclear. This study aimed to elucidate the effects of adolescent intermittent ethanol (AIE) exposure on cholinergic neurons in the medial septum (MS) and the resultant cognitive deficits in adult mice.
MATERIALS AND METHODS: From postnatal day (PND) 28 to PND 41, mice received ethanol through intragastric gavage on alternate days to establish the model of AIE. To assess the electrophysiol

Cocaine- and amphetamine-regulated transcript (CART) peptide has been implicated in stress-related behaviors that are regulated by central serotonergic (5-HT) systems in the dorsal raphe nucleus (DRN). Here, we aimed to investigate the interaction between CART and DRN 5-HTergic systems after initially observing CART axonal terminals in the DRN. We found that microinfusion of CART peptide (55-102) into the DRN-induced anxiogenic effects in male C57BL/6J mice, while central administration of CART reduced c-Fos in 5-HTDRN neurons. This inhibitory effect of exogenous CART on 5-HTDRN activity and l

BACKGROUND: Microglia-neuronal communication is crucial for the development and maintenance of pain. However, the exact mechanisms underlying this interaction and its role in anterior cingulate cortex (ACC) circuitry in pain regulation are under exploration.
METHODS: We explored the role of P2X4R-brain-derived neurotrophic factor (BDNF)-TrkB signalling of ACC in regulating muscle pain (MP). Mechanical and thermal pain thresholds along with open field tests were used to assess pain and anxiety-like behaviours. Golgi staining, transmission electron microscopy, and patch-clamp recordings were pe

Cocaine use disorder (CUD) is highly comorbid with HIV infection and worsens HIV outcomes. Preclinical research on the outcomes of HIV infection may yield crucial information on neurobehavioral changes resulting from chronic drug exposure in people living with HIV (PLWH). Repeated exposure to cocaine alters behavioral responses to cocaine. This includes development of cocaine locomotor sensitization - or increased locomotor responses to the same doses of cocaine - which depends on nucleus accumbens (NAc) neural plasticity. NAc astrocytes are key regulators of neural activity and plasticity, an

Drug-induced neuroadaptations in the prefrontal cortex (PFC) have been implicated in drug-associated memories that motivate continued drug use. Chronic cocaine exposure increases pyramidal neuron excitability in the prelimbic subregion of the PFC (PL), an adaptation that has been attributed in part to a suppression of inhibitory signalling mediated by the GABAB receptor (GABAB R) and G protein-gated inwardly rectifying K+ (GIRK/Kir3) channels. Although reduced GIRK channel activity in PL pyramidal neurons enhances the motor-stimulatory effect of cocaine in mice, the impact on cocaine reward an

BACKGROUND: Attentional deficits are among the most common pain-induced cognitive disorders. Pain disrupts attention and may excessively occupy attentional resources in pathological states, leading to daily function impairment and increased disability. However, the neural circuit mechanisms by which pain disrupts attention are incompletely understood.
METHODS: We used a three-choice serial reaction time task (3CSRTT) to construct a sustained-attention task model in male C57BL/6J mice. Formalin or complete Freund's adjuvant was injected into a paw to establish an inflammatory pain model. We me

States of physiological need motivate individuals to seek and consume stimuli that restore homeostatic balance. This goal-directed behavior is driven, in part, by pathways that process reward and are sensitive to changes in physiological state, including the mesolimbic dopamine system. The effects of hunger and its physiological markers have been more widely studied for their role in modulating reward signaling pathways. However, fluid need produces robust goal-directed behavior and has also been shown to affect neural substrates of reward processing. To test how acute and chronic states of th

AIMS: Itch is an unpleasant sensation that severely impacts the patient's quality of life. Recent studies revealed that the G protein-coupled estrogen receptor (GPER) may play a crucial role in the regulation of pain and itch perception. However, the contribution of the GPER in primary sensory neurons to the regulation of itch perception remains elusive. This study aimed to investigate whether and how the GPER participates in the regulation of itch perception in the trigeminal ganglion (TG).
METHODS AND RESULTS: Immunofluorescence staining results showed that GPER-positive (GPER+ ) neurons of

AIMS: Sepsis-associated encephalopathy (SAE) often leads to cognitive impairments. However, the pathophysiology of SAE is complex and unclear. Here, we investigated the role of hippocampus (HPC)-prefrontal cortex (PFC) in cognitive dysfunction in sepsis induced by cecal ligation puncture (CLP) in mice.
METHODS: The neural projections from the HPC to PFC were first identified via retrograde tracing and viral expression. Chemogenetic activation of the HPC-PFC pathway was shown via immunofluorescent staining of c-Fos-positive neurons in PFC. Morris Water Maze (MWM) and Barnes maze (BM) were used

BACKGROUND: Breathing disorders (BD) (apnoeas/hypopneas, periodic breathing) are highly prevalent in chronic heart failure (CHF) and are associated with altered central respiratory control. Ample evidence identifies the retrotrapezoid nucleus (RTN) as an important chemosensitivity region for ventilatory control and generation of BD in CHF, however little is known about the cellular mechanisms underlying the RTN/BD relationship. Within the RTN, astrocyte-mediated purinergic signalling modulates respiration, but the potential contribution of RTN astrocytes to BD in CHF has not been explored.

Gastric cancer (GC) is a leading lethal malignancy globally. Gastric precancerous lesion (GPL) represents a critical step in GC, but its neural regulatory mechanisms remain largely unclear. We combined bioinformatics, patch-clamp recording, immunofluorescence, and chemogenetics to investigate serotonergic and cholinergic signaling in GPL. An N-methyl-N-nitrosourea (MNU) -induced mouse model was used for in vivo validation. High 5-HTR1D expression correlated with poor survival in gastric adenocarcinoma patients and was upregulated in AGS and MKN45 cells, as well as in gastric tissues of GPL mic…
