Chlamydia trachomatis Hijacks Host Cell Programs to Establish Persistence.

Chlamydia trachomatis (C. trachomatis) is a major global pathogen responsible for bacterial sexually transmitted infections. The majority of infections are asymptomatic and may persist for prolonged periods; consequently, clinically silent transmission and upper genital tract complications represent a persistent public health burden. The biological basis of this clinical phenotype resides in the bacterium's capacity to sustain infection despite host immune pressure. However, the molecular mechanisms underpinning this process remain unclear. Accumulating evidence indicates that, upon invasion, …

Iron limitation-induced modulation of transcription in Chlamydia trachomatis.

Chlamydia trachomatis (C. trachomatis) is an obligate intracellular bacterial pathogen responsible for trachoma and sexually transmitted infections. Like other Chlamydiota members, such as Waddlia chondrophila, C. trachomatis undergoes a biphasic developmental cycle alternating between infectious elementary bodies (EBs) and replicative reticulate bodies (RBs). Under stressful conditions, RBs differentiate into non-dividing aberrant bodies (ABs), a reversible state associated with persistence. Here, we investigate how early iron deprivation affects transcriptional regulation in C. trachomatis. …

Detection of Chlamydia trachomatis Pgp3 Antibody in Human Sera.

OBJECTIVE: Chlamydia trachomatis is the most common bacterial sexually transmitted infection (STI) worldwide. Genital infection with C. trachomatis in women can cause ascending infection leading to pelvic inflammatory disease. Studies have suggested that the Pgp3 protein encoded by C. trachomatis plasmid may contribute to long term inflammatory complications of C. trachomatis infection (tubal factor infertility). This study assessed C. trachomatis-specific Pgp3 IgG antibody levels in sera from adolescents and young adults in an inner-city population in New York. METHODS: Blood was obtained fr…

Chlamydia trachomatis infection induces ferroptosis and enhances chlamydial replication.

Chlamydia trachomatis (C. trachomatis) has been shown to activate multiple programmed cell death pathways, which contribute significantly to host immune responses. Nevertheless, the precise molecular mechanisms by which C. trachomatis induces cell death remain poorly characterized. Ferroptosis, a recently identified form of iron-dependent, lipid peroxidation-driven regulated cell death, may represent a previously unrecognized pathway in chlamydial pathogenesis. To investigate the mechanisms underlying C. trachomatis-induced cell death, we first performed transcriptomic analysis to identify dif…

Metabolic dormancy in Chlamydia trachomatis treated with different antibiotics.

Diseases caused by Chlamydia spp. are often associated with persistent infections. Chlamydial persistence is commonly associated with a uniq … lementary bodies were not detected during treatment with each antibiotic, but viable and culturable Chlamydia trachomatis emerged after the drug was removed. Time-lapse imaging of live, antibiotic-treated infected cells identified metabolically dormant developmental forms within cells that emerged to form typical productive inclusions. Chlamydia trachomatis Subverts Alpha-Actinins To Stabilize Its Inclusion.

Chlamydia trachomatis is the leading cause of sexually transmitted bacterial disease and a global health burden. As an obligate intracellular pathogen, Chlamydia has evolved many strategies to manipulate its host and establish its intracellular niche called the inclusion. C. trachomatis reorganizes the host actin cytoskeleton to form scaffolds around the inclusion and reinforce the growing inclusion membrane. To control the kinetics and formation of actin scaffolds, Chlamydia expresses the effector InaC/CT813, which activates the host GTPase RhoA. Here, we have discovered that InaC stabilizes …

ATG9A supports Chlamydia trachomatis infection via autophagy-independent mechanisms.

ATG9A is an autophagy-related gene that functions during the isolation membrane expansion process to form autophagosomes, but it also has other functions independent of autophagy. In this study, we employed ATG9A-deficient HeLa cells and found that the absence of ATG9A negatively impacted proliferation of Chlamydia trachomatis in inclusions. Furthermore, rescue experiments using ATG9A mutants revealed that this action was mediated not by its autophagic function but by its binding ability to clathrin adapter proteins. These findings suggest that the proper trafficking of ATG9A assists C. tracho…

Interferon-ε as potential inhibitor of Chlamydia trachomatis infection.

Chlamydia trachomatis, the main cause of bacterial sexually transmitted diseases, is responsible for severe reproductive sequelae. Amongst all the cytokines involved in host immunity towards this pathogen, IFN-ε has recently acquired importance for its potential contribution to the female reproductive tract innate defenses. Herein, our study aimed to explore, for the first time, the activity of IFN-ε toward C. trachomatis in an in vitro infection model, by testing its effects on the different phases of chlamydial developmental cycle, as well as on the ultrastructural characteristics of chlamyd…

Mycoplasma decontamination in Chlamydia trachomatis culture: a curative approach.

Mycoplasma contamination of cell culture represents a serious problem in research and decontamination from cell-propagated obligate intracellular bacteria has proven challenging. Here, we presented an optimized protocol to remove Mycoplasma from contaminated Chlamydia trachomatis culture. A stepwise procedure of Mycoplasma removal entails (i) incubation in nonionic detergent-containing solution and (ii) separation of viable chlamydial organisms by fluorescence-activated cell sorting (FACS), followed by subcloning using a focus-forming assay. We also adapted a polymerase chain reaction (PCR) as…

Chlamydia trachomatis Cell-to-Cell Spread through Tunneling Nanotubes.

Tunneling nanotubes (TNTs) are transient cellular connections that consist of dynamic membrane protrusions. They play an important role in c … djacent cells to mediate efficient communication and the exchange of material between them. We used Chlamydia trachomatis and immortalized cells to analyze whether TNTs mediate bacterial transmission from an infected donor to uninfected acceptor cells. We show that chlamydia-infected cells build TNTs through which the intracellular reticulate bodies (RBs) of the chlamy …

Chlamydia trachomatis: quest for an eye-opening vaccine breakthrough.

INTRODUCTION: Chlamydia trachomatis, commonly referred to as chlamydia (a bacterium), is a common sexually transmitted infection, and if attended to early, it can be treatable. However, if left untreated it can lead to serious consequences. C. trachomatis infects both females and males although its occurrence in females is more common, and it can spread to the eyes causing disease and in some case blindness. AREA COVERED: With ongoing attempts in the most impoverished regions of the country, trachoma will be eradicated as a blinding disease by the year 2022. A prophylactic vaccine candidate w…

Incident Chlamydia trachomatis Infection in a High School Population.

Prospective cohort studies of sexually transmitted infections (STIs) are logistically impractical owing to time and expenses. In schools, students are readily available for school-related follow-ups and monitoring. Capitalizing on the logistics that society already commits to ensure regular attendance of adolescents in school, a school-based STI screening in New Orleans made it possible to naturally observe the occurrence of chlamydia and to determine its incidence among 14-19-year-old adolescents. Among participants screened repeatedly, we calculated incidence rates, cumulative incidence, and

HIV, HPV and Chlamydia trachomatis: impacts on male fertility.

Sexually transmitted diseases (STDs) are pathologies that have viruses, bacteria, protozoa and fungi as infectious agents, affecting millions of people worldwide and causing physical and psychological consequences for the carrier. Some of these infections such as HIV, HPV and Chlamydia trachomatis may present an asymptomatic phase, making the diagnosis difficult, which is often only performed when the couple looks for infertility treatment after not achieving spontaneous pregnancy. Infertility affects 15% of couples, 50% of cases are male-related, and it is estimated that STDs, which interfere…

The Cervicovaginal Microbiota-Host Interaction Modulates Chlamydia trachomatis Infection.

The mechanism(s) by which Lactobacillus-dominated cervicovaginal microbiota provide a barrier to Chlamydia trachomatis infection remain(s) unknown. Here we evaluate the impact of different Lactobacillus spp. identified via culture-independent metataxonomic analysis of C. trachomatis-infected women on C. trachomatis infection in a three-dimensional (3D) cervical epithelium model. Lactobacillus spp. that specifically produce d(-) lactic acid were associated with long-term protection against C. trachomatis infection, consistent with reduced protection associated with Lactobacillus iners, which do…
