A method for sample preparation and high-performance liquid chromatography detection and quantification has been developed for the routine analysis of psoralen and bergapten, photosensitizing compounds, in topic solutions employed in Brazil for treatment of vitiligo and psoriasis. The relative efficiencies of four enzymic digestion procedures and two conventional methods in releasing barbiturates from spiked liver tissue have been compared. Carbonic anhydrase-azosulfonamide dissociation constants (Kd) were determined by gel filtration with high-performance liquid chromatography. By measuring the area of the elution peak at two wavelengths, Kd values were derived without having to measure a shallow trough. This report describes the application of high performance liquid chromatography (HPLC) and associated techniques to the isolation of inhibin from ovine follicular fluid (oFF). Two modes of HPLC have been assessed. High speed gel permeation chromatography on a Waters I-125 support showed that at pH 7.0 inhibin bioactivity was associated with an approx. 80000 mol. wt protein fraction which co-eluted with the bulk of the protein in oFF and gave a yield of 30-44% of the applied activity. The quantitative determination of morphine in paregoric by reversed-phase high-pressure liquid chromatography is described. The method is simple, accurate, and fast compared to the USP method. Benzoic acid in paregoric also can be determined quantitatively with the same column. The method was applied to two commercial samples with excellent results.
