Nanoparticle-Based Interventions for Liver Transplantation.

Liver transplantation is the only treatment for hepatic insufficiency as a result of acute and chronic liver injuries/pathologies that fail to recover. Unfortunately, there remains an enormous and growing gap between organ supply and demand. Although recipients on the liver transplantation waitlist have significantly higher mortality, livers are often not allocated because they are (i) classified as extended criteria or marginal livers and (ii) subjected to longer cold preservation time (>6 h) with a direct correlation of poor outcomes with longer cold ischemia. Downregulating the recipient's

A novel affective-motivational-based Overground System for detecting spinal cord injury-associated thermal and mechanical hypersensitivity in rats.

BACKGROUND: Preclinical research for neuropathic pain has depended primarily on the use of behavioural nociceptive testing that is sensory-discriminatory-based and reflexive in nature. This can be particularly problematic in spinal cord injury (SCI)-associated neuropathic pain research where hyperreflexia may develop thus confounding interpretation of reflexive responses as pain symptoms. To address this, we have designed an affective-motivational-based Overground System that has interchangeable floors to allow examination of nociceptive behaviours in response to mechanical and cold stimuli pr

Characterization of a model of cutaneous inflammatory pain produced by an ultraviolet irradiation-evoked sterile injury in the rat.

Neuroimmune interactions are of known importance in the genesis and maintenance of inflammatory pain states. However, the immune response to tissue damage is likely to differ depending on whether or not the injury is accompanied by infection. Many clinically important inflammatory pain states involve a sterile tissue injury. However, existing animal models of cutaneous inflammatory pain use injuries that are likely to involve those components of the immune system that are specialized for combating pathogens (e.g., injections of Complete Freund's Adjuvant, carrageenan, or zymosan). We describe

[The effects of cold exposure on the ileum mechanical barrier in mice and its mechanisms].

Objective: To study the mechanisms of cold exposure mediated ileum mechanical barrier injury in mice. Methods: Twenty mice were randomly divided into the control and cold exposure groups. Both the control and cold exposure groups were placed in the climate room with (24±2)℃ and 40% humidity. The mice in the cold exposure group were moved to the climate room at (4±2)℃ every day for 3 hours for three consecutive weeks. Three weeks later, the ileum tissues of mice were collected. Changes in ileum tissue structure were observed by hematoxylin-eosin staining and Masson staining. The related protein

Long-term Patency of Primary Arterial Repair and the Modified Cold Intolerance Symptom Severity Questionnaire.

BACKGROUND: The goal of this study was to assess the long-term arterial patency of repaired arteries in the upper extremity and any morbidity resulting from the subsequent occlusion of these vessels. Concurrently, a new questionnaire, the modified Cold Intolerance Symptom Severity (mod CISS) questionnaire, was developed to allow for better assessment of cold intolerance. METHODS: Thirteen patients who had undergone repair of the radial (4 patients), ulnar (6 patients), brachial (1 patient), digital (1), and an undefined lower arm artery (1) were examined using questionnaires, physical examina

Chilling Injury to Algal Symbionts Induces Host Starvation and Metabolic Reorganization in a Temperate Cnidarian.

Global temperature anomalies increasingly disrupt the cnidarian-algal symbiosis resulting in a phenomenon termed bleaching. In contrast to heat stress, the mechanisms underlying symbiotic breakdown under cold stress remain largely unknown. Combining physiological and metabolomic measurements, we investigated the response of the photosymbiotic sea anemone holobiont Aiptasia couchii to an experiment mimicking a cold spell in the Mediterranean Sea. Within 4 weeks, we observed the onset of symbiotic breakdown reflected in reduced algal endosymbiont density and chlorophyll a content. While photosyn

Submilligram Level of Beetle Antifreeze Proteins Minimize Cold-Induced Cell Swelling and Promote Cell Survival.

Hypothermic (cold) preservation is a limiting factor for successful cell and tissue transplantation where cell swelling (edema) usually develops, impairing cell function. University of Wisconsin (UW) solution, a standard cold preservation solution, contains effective components to suppress hypothermia-induced cell swelling. Antifreeze proteins (AFPs) found in many cold-adapted organisms can prevent cold injury of the organisms. Here, the effects of a beetle AFP from Dendroides canadensis (DAFP-1) on pancreatic β-cells preservation were first investigated. As low as 500 µg/mL, DAFP-1 significan…

Cold-induced vasodilatation is not homogenous or generalizable across the hand and feet.

Cold-induced vasodilatation (CIVD), a paradoxical cyclical increase in finger temperature upon cold exposure, is assumed to serve against cold injury. Most existing research has focused on hand and finger responses, yet most cold injuries occur in the feet. We investigated if CIVD responses of one finger can predict the responses of other fingers, and also whether the fingers could predict CIVD responses of the feet and toes. Ten subjects immersed their left hand up to the wrist in 8 degrees C water for 30 min. They passively rewarmed for 15 min, followed by immersion of the left foot to the a…

DNA methylation, transcriptomic, and metabolomic analyses provide new mechanistic insights into cold shock-induced chilling tolerance in cucumber.

Cucumber is cold-sensitive; improper storage induces chilling injury. To investigate how 3 h 0 °C cold shock (CS) alleviates chilling injury in 'Baijinyu' cucumbers at 4 °C, we performed integrated DNA methylation, transcriptomic, and metabolomic analyses. CS alleviated chilling injury, delayed quality decline (soluble solids, firmness, color), and altered accumulation of lipids, keto acids/esters, amino acids, and organic acids. CS increased methylation of peroxidase, caffeic acid 3‑O‑methyltransferase, and cinnamyl‑alcohol dehydrogenase, upregulating peroxidase but downregulating the latter

Integrative physiology, transcriptomic, and metabolomic analysis reveals the response mechanism of melon seedlings to cold stress.

Early-spring cultivation enables melons to be marketed ahead of the regular season, yet cold stress during this critical growth stage severely suppresses seedling growth and development, ultimately causing substantial yield losses. To date, the physiological shifts, molecular regulatory cascades and metabolic reprogramming triggered by cold stress in melon seedlings have not been fully characterized. In this work, we combined physiological assays, transcriptome, and metabolome profiling to dissect cold-responsive regulatory networks at the seedling stage using two melon inbred lines with diver

Hypothermia Alleviates Reductive Stress, a Root Cause of Ischemia Reperfusion Injury.

Ischemia reperfusion injury is common in transplantation. Previous studies have shown that cooling can protect against hypoxic injury. To date, the protective effects of hypothermia have been largely associated with metabolic suppression. Since kidney transplantation is one of the most common organ transplant surgeries, we used human-derived renal proximal tubular cells (HKC8 cell line) as a model of normal renal cells. We performed a temperature titration curve from 37 °C to 22 °C and evaluated cellular respiration and molecular mechanisms that can counteract the build-up of reducing equivale

Expression of transient receptor potential ankyrin 1 in human dental pulp.

INTRODUCTION: Transient receptor potential ankyrin 1 (TRPA1) is activated by noxious cold (<17°C) and contributes to cold and mechanical hypersensitivity after inflammation and nerve injury. METHODS: To investigate whether TRPA1 is involved in the mediation of nociception, including noxious cold and cold hypersensitivity in teeth, we examined the expression of TRPA1 and sodium channel Nav1.8 in human dental pulp using fluorescent and electron microscopic immunocytochemistry. RESULTS: TRPA1 was expressed in a large number of axons branching extensively in the peripheral pulp and in a few axon

Excess risk of head and chest colds among teachers and other school workers.

BACKGROUND: Work-related injuries and illnesses in the educational services sector have not been well studied. This analysis examined whether teachers and other school workers are at higher risk of head/chest cold compared to all other workers in the United States. METHODS: Seven years (1998-2004) of National Health Interview Survey data on currently employed workers were combined to provide a basis for estimating the incidence proportion of head/chest cold. RESULTS: The adjusted odds ratio for head/chest cold was significantly elevated for teachers and other workers employed at schools comp

The effect of acute cold ischemia duration on testicular tissue.

PURPOSE: Testicular cancer primarily affects men aged 15–34. Partial orchiectomy has recently emerged as a viable testis-sparing surgical technique. However, prolonged spermatic cord clamping may induce ischemia–reperfusion (I/R) injury due to elevated reactive oxygen species (ROS) production. This study aimed to determine the maximum tolerable duration of testicular cold ischemia under hypothermic conditions and to evaluate its impact on ischemia–reperfusion injury in a rat model. MATERIALS AND METHODS: Thirty sexually mature Wistar albino rats were divided into four groups. Following scrotal

Immediate impacts of low-temperature exposure on cardiac autonomic modulation, neuromuscular efficiency, and postural stability in older adults Yangko dancers - a randomized controlled trial.

INTRODUCTION: To investigate the immediate effects of different cold environments on heart rate variability (HRV), surface electromyography (sEMG), and balance in older adults Yangko dance (a traditional Chinese folk dance) participants, and to elucidate the mechanisms underlying increased injury risks during the early exercise phase (10-20 minutes) in cold conditions. This study aims to provide scientific evidence for targeted protective measures. METHODS: A randomized controlled trial was conducted with 120 regular Yangko dancers (age>60 years) from Changchun, China. Participants were strat

Hypothermic Preconditioning of Human Cortical Neurons Requires Proteostatic Priming.

Hypothermia is potently neuroprotective but poor mechanistic understanding has restricted its clinical use. Rodent studies indicate that hypothermia can elicit preconditioning, wherein a subtoxic cellular stress confers resistance to an otherwise lethal injury. The molecular basis of this preconditioning remains obscure. Here we explore molecular effects of cooling using functional cortical neurons differentiated from human pluripotent stem cells (hCNs). Mild-to-moderate hypothermia (28-32 °C) induces cold-shock protein expression and mild endoplasmic reticulum (ER) stress in hCNs, with full a

Enteral donor pre-treatment with ursodeoxycholic acid protects the liver against ischaemia-reperfusion injury in rats.

Liver donor pre-treatment with ursodeoxycholic acid (UDCA) may protect against injury during transplantation. In the present study we evaluated whether enteral administration of UDCA has an effect on bile flow and protects the liver from injury related to transplantation. Wistar rats were used in liver perfusion (LP) and transplantation (LTx) models. Rats were enterally administered UDCA (800 mg/kg) 3 h before cold perfusion. In LP, bile flow and bile acid composition were analysed. In LTx, serum ALT and liver histology were analysed. LP showed biliary UDCA enrichment up to 36+/-13% in pre-tre

Neuropeptide Y Y2 Receptors in Sensory Neurons Tonically Suppress Nociception and Itch but Facilitate Postsurgical and Neuropathic Pain Hypersensitivity.

BACKGROUND: Neuropeptide Y (NPY) Y2 receptor (Y2) antagonist BIIE0246 can both inhibit and facilitate nociception. The authors hypothesized that Y2 function depends on inflammation or nerve injury status. METHODS: The authors implemented a battery of behavioral tests in mice of both sexes that received (1) no injury; (2) an incision model of postoperative pain; (3) a spared nerve injury model of neuropathic pain; and (4) a latent sensitization model of chronic postsurgical pain. In addition to Y2 gene expression assays, spinal Y2 G-protein coupling was studied with guanosine-5'-O-(3-[35S]thio

Does Cold Water or Ice Slurry Ingestion During Exercise Elicit a Net Body Cooling Effect in the Heat?

Cold water or ice slurry ingestion during exercise seems to be an effective and practical means to improve endurance exercise performance in the heat. However, transient reductions in sweating appear to decrease the potential for evaporative heat loss from the skin by a magnitude that at least negates the additional internal heat loss as a cold ingested fluid warms up to equilibrate with body temperature; thus explaining equivalent core temperatures during exercise at a fixed heat production irrespective of the ingested fluid temperature. Internal heat transfer with cold fluid/ice is always 10
