Aptamer-Functionalized Liposome Delivery System Based on Multifunctional Microreactor Design.

Breast cancer remains the leading cause of cancer-related mortality in women, with conventional chemotherapy often limited by systemic toxicity due to poor drug targeting. Here, we present an integrated microfluidic chip (MMLP) that enables one-step integrated postprocessing, including solvent removal and ligand conjugation. The MMLP chip combines Y-shaped, crescent-shaped, U-shaped, and Tesla-type microchannels, augmented with pillar arrays to enhance mixing efficiency at low flow rates. A dedicated third channel allows simultaneous solvent removal and aptamer conjugation. Through computation

Exploring Indole-Based Molecules as Novel Therapeutics for Parkinson's Disease: A Comprehensive Review.

The primary feature of Parkinson's disease (PD), a progressive neurodegenerative disease that results in both motor and non-motor dysfunctions, is the degeneration of dopaminergic neurons in the substantia nigra. In recent years, indole-based compounds have emerged as promising candidates for developing novel treatments for Parkinson's disease due to their diverse pharmacological properties. Among the significant pathogenic targets against which indole derivatives exhibit potent activity are monoamine oxidase (MAO), NMDA receptors, oxidative stress, and neuroinflammation. This review provides

Topological indices and data analysis techniques modeling to predict the physicochemical properties of tetracycline antibiotics.

The tetracycline family of drugs is one of the most widely used groups of antibiotics in modern medicine. Topological indices act as a bridge between chemistry and mathematics. The Quantitative Structure-Property Relationship (QSPR) models utilize the molecular structure of compounds to predict the physicochemical properties. In this paper, a computational approach was performed using MATLAB coding and decoding to calculate the Sombor-type topological indices of this group of drugs. The linear regression approach has been used in the quantitative model of structure-property relationships to in

Sulfur-arene interactions: the S⋯π and S-H⋯π interactions in the dimers of benzofuran⋯sulfur dioxide and benzofuran⋯hydrogen sulfide.

Non-covalent interactions between sulfur centers and aromatic rings play important roles in biological chemistry. We examined here the sulfur-arene interactions between the fused aromatic heterocycle benzofuran and two prototype sulfur divalent triatomics (sulfur dioxide and hydrogen sulfide). The weakly-bound adducts were generated in a supersonic jet expansion and characterized with broadband (chirped-pulsed) time-domain microwave spectroscopy. The rotational spectrum confirmed the detection of a single isomer for both heterodimers, consistent with the computational predictions for the globa

Efficient Multiconfigurational Quantum Chemistry Approach to Single-Ion Magnets Based on Density Matrix Embedding Theory.

Density matrix embedding theory (DMET) provides a systematic framework to combine low-level (e.g., Hartree-Fock approximation) and high-level correlated quantum chemistry methods to treat strongly correlated systems. In this work, we propose an efficient quantum embedding approach that combines DMET with the complete active space self-consistent field and subsequent state interaction treatment of spin-orbit coupling (CASSI-SO) and apply it to a theoretical description of single-ion magnets (SIMs). We have developed a novel regularized direct inversion of iterative subspace (R-DIIS) technique t

Remote C(sp3 )-H Acylation of Amides and Cascade Cyclization via N-Heterocyclic Carbene Organocatalysis.

The direct functionalization of inert C(sp3 )-H bonds under environmentally benign catalytic conditions remains a challenging task in synthetic chemistry. Here, we report an organocatalytic remote C(sp3 )-H acylation of amides and cascade cyclization through a radical-mediated 1,5-hydrogen atom transfer mechanism using N-heterocyclic carbene as the catalyst. Notably, a diversity of nitrogen-containing substrates, including simple linear aliphatic carbamates and ortho-alkyl benzamides, can be successfully applied to this organocatalytic system. With the established protocol, over 120 examples o

Discovery of a Novel MyD88 Inhibitor M20 and Its Protection Against Sepsis-Mediated Acute Lung Injury.

Myeloid differentiation factor 88 (MyD88) is a hub protein in the Toll-like receptor signaling pathway, which acts as a master switch for nu … inhibitor to efficiently relieve lipopolysaccharide-induced inflammation both in vitro and in vivo. Computational chemistry, surface plasmon resonance detection (SPR) and biological experiments demonstrated that M20 forms an important interaction with the MyD88-Toll/interleukin-1 receptor domain and thereby inhibits the protein dimerization. Taken together, this study fo …

Stimulation Modulates Adhesion and Mechanics of Hydrogel Adhesives.

The ability to modulate the adhesion of soft materials on-demand is desired for broad applications ranging from tissue repair to soft robotics. Research effort has been focused on the chemistry and architecture of interfaces, leaving the mechanics of soft adhesives overlooked. Stimuli-responsive mechanisms of smart hydrogels could be leveraged for achieving stimuli-responsive hydrogel adhesives that respond mechanically to external stimuli. Such stimuli-responsive hydrogel adhesives involve complex chemomechanical coupling and interfacial fracture phenomena, calling for mechanistic understandi

Sampling Potential Energy Surfaces in the Condensed Phase with Many-Body Electronic Structure Methods.

Sampling potential energy surfaces (PES) is pivotal for understanding chemical structure, energetics and reactivity and is of special importance for complex condensed-phase systems. Until recently such simulations based on electronic structure theory have been performed only by density functional theory and semiempirical methods. Many-body electronic structure methods, almost routinely used for molecules, have been practically unavailable for sampling PES in the condensed-phase. This has changed during the last few years, as efficient algorithms and software implementations for the evaluation

Small Molecule Activation by Constrained Phosphorus Compounds: Insights from Theory.

An exciting new development in main group chemistry has been the use of a constrained, "flat", phosphorus-based complex to mediate in reactions such as the dehydrogenation of ammonia borane (AB), and the activation of the N-H bond in primary amines. Its importance is based on the fact that it shows that main group compounds, when properly designed, can be as effective as transition metal complexes for doing significant chemical transformations. What the current computational study, employing density functional theory (DFT), reveals is that a common, general mechanism exists that accounts for t

In-silico analysis of chromone containing sulfonamide derivatives as human carbonic anhydrase inhibitors.

Computational tools of analysis were used on a set of synthetic chromone containing sulfonamide derivatives for evaluation of their enzyme inhibitory activity against Carbonic Anhydrase (CA) isozymes. GOLD docking software was utilized to dock the compounds against two human Carbonic Anhydrase (hCA) proteins; hCAII and hCA-IX. Differences in conformation and orientation of molecules within hCA-II and hCA-IX binding pockets were studied in detail which revealed that compounds with fluorine at R1 position and phenyl sulfonamide substituent at para position served as potent inhibitors against bot

N-heterocyclic phosphenium ligands as sterically and electronically-tunable isolobal analogues of nitrosyls.

The coordination chemistry of an N-heterocyclic phosphenium (NHP)-containing bis(phosphine) pincer ligand has been explored with Pt(0) and Pd(0) precursors. Unlike previous compounds featuring monodentate NHP ligands, the resulting NHP Pt and Pd complexes feature pyramidal geometries about the central phosphorus atom, indicative of a stereochemically active lone pair. Structural, spectroscopic, and computational data suggest that the unusual pyramidal NHP geometry results from two-electron reduction of the phosphenium ligand to generate transition metal complexes in which the Pt or Pd centers

Unimodal vs Multimodal Learning: A Systematic Evaluation of Fusion Strategies and Model Design for Molecular Property Prediction and Uncertainty Quantification.

Accurate prediction of molecular properties is fundamental to environmental chemistry, yet remains challenging when experimental data are limited. Multimodal fusion provides a promising strategy for integrating complementary molecular representations; however, the relative contributions of molecular representation, fusion strategy, and learning algorithm to the predictive accuracy and uncertainty remain poorly understood. Five molecular modalities (RDKit descriptors, Mol2Vec embeddings, graph neural network embeddings, SMILES representations, and MS2 fragmentation spectra) were evaluated by us

Learning to Rank for Selected Configuration Interaction.

Accurate description of electron correlation is a central challenge in computational chemistry, with selected configuration interaction (SCI) emerging as a powerful tool to approach the full CI limit. While recent machine learning (ML) integrations have accelerated determinant selection, existing regression and classification approaches suffer from a fundamental objective-loss mismatch: they evaluate the importance of determinants in isolation without explicitly accounting for their relative importance ranking. Here, we introduce ranking configuration interaction (RCI), a novel ML-supported SC…

Tunable triazole-based cholera toxin inhibitors: A QSAR-guided design and evaluation approach.

Cholera toxin B subunit (CTB) is a validated target for anticholera therapeutics, but current inhibitors often suffer from synthetic complexity and limited tunability. This study aimed to develop a compact, tunable triazole scaffold exhibiting low-micromolar potency combined with high synthetic tractability. We applied a unified, QSAR-driven, multiscale computational-experimental workflow integrating molecular docking, induced fit docking (IFD), molecular dynamics (MD), QM/MM calculations, and descriptor-based QSAR modeling to prioritize a focused library of 44 N-sulfonyl triazole inhibitors.

Capturing Dichotomic Solvent Behavior in Solute-Solvent Reactions with Neural Network Potentials.

Simulations of chemical reactivity in condensed phase systems represent an ongoing challenge in computational chemistry, where traditional quantum chemical approaches typically struggle with both the size of the system and the potential complexity of the reaction. Here, we introduce a workflow aimed at efficiently training neural network potentials (NNPs) to explore energy barriers in solution at the hybrid density functional theory level. The computational burden associated with training at the PBE0-D3(BJ) level is bypassed through the use of active and transfer learning techniques, whereas e…
