A review on charred traditional Chinese herbs: carbonization to yield a haemostatic effect.

Context: Charcoal of Chinese drugs is a kind of special processing product in Chinese medicine and used for treatment of haemoptysis, hematemesis and haemorrhage in the clinic during ancient times. During carbonizing, significant changes occur in chemical constituents and the efficacy of haemostasis will be enhanced. But the quality control standard of 'carbonizing retains characteristics' should be followed. Objective: This review introduces the typical methods of carbonizing, which highlight current research progress on haemostatic substances of charcoal drugs so as to provide a reasonable e

Prediction Model for Antimalarial Activities of Hemozoin Inhibitors by Using Physicochemical Properties.

The rapid spread of strains of malaria parasites that are resistant to several drugs has threatened global malaria control. Hence, the aim of this study was to predict the antimalarial activity of chemical compounds that possess anti-hemozoin-formation activity as a new means of antimalarial drug discovery. After the initial in vitro anti-hemozoin-formation high-throughput screening (HTS) of 9,600 compounds, a total of 224 hit compounds were identified as hemozoin inhibitors. These 224 compounds were tested for in vitro erythrocytic antimalarial activity at 10 μM by using chloroquine-mefloquin

Discovery of a New Scaffold RSK2 Inhibitor With Virtual and Phenotypical Screening.

Ribosomal S6 kinase (RSK2) is regarded as an important target for the treatment of breast and ovarian cancers. However, most reported RSK2 inhibitors suffer from insufficient selectivity and unfavorable pharmacokinetic properties due to their similar chemical scaffolds. Therefore, the discovery of novel RSK2 inhibitors with high potency, selectivity, and desirable drug-like properties remains highly desirable. In this work, we established a virtual screening workflow combining pharmacophore modeling, molecular docking, and molecular dynamics simulations. By screening the Chemdiv database (1.6

Development and validation of a novel senescence-associated gene signature for prediction of survival and endocrine-disrupting chemicals in bladder cancer.

Endocrine-disrupting chemicals (EDCs) are prevalent in various aspects of daily life, impacting the homeostasis of physiological processes and potentially contributing to the pathogenesis of cancer. Bladder cancer (BLCA) is the most common malignancy of the urinary system and one of the most prevalent malignancies globally. Cellular senescence is increasingly recognized as a key factor in cancer development. However, research remains limited regarding how EDCs influence BLCA through senescence-related genes (SRGs). In this study, expression data and clinical information were sourced from the C

Nasal hyper-reactivity is a common feature in both allergic and nonallergic rhinitis.

BACKGROUND: Nasal hyper-reactivity is an increased sensitivity of the nasal mucosa to various nonspecific stimuli. Both allergic rhinitis (AR) and nonallergic rhinitis (NAR) patients can elicit nasal hyper-reactivity symptoms. Differences in the prevalence or type of nasal hyper-reactivity in AR and NAR patients are largely unknown. In this study, we quantitatively and qualitatively assessed nasal hyper-reactivity in AR and NAR. METHODS: In the first part, an analysis of a prospectively collected database was performed to reveal patient-reported symptoms of hyper-reactivity. In the second par

Study on Phosphorus Compound/Catechol-Catalyzed Dehydrative Amidation and Its Database Development for Machine Learning.

Herein, we report that the catalytic activity of phosphorus compounds with a P─H bond in dehydrative amidation reactions is substantially enhanced when using catechol derivatives as additives. A systematic investigation into the catalytic activity of four phosphorus compounds and 17 catechol derivatives allowed identifying several effective combinations, for example, dimethyl phosphite and 2,3-dihydroxynaphthalene, for the catalytic amidation of carboxylic acids with amines. The developed catalytic system enables the synthesis of pharmaceuticals and facilitates the amidation of anilines, which

Feline ocular tumors following ciliary body ablation with intravitreal gentamicin.

Practitioners approach chemical ciliary body ablation (CBA) in cats with caution. In 1994, an academic letter proposed a potential link between intraocular gentamicin injections for glaucoma and the appearance of ocular tumors in cats (Veterinary and Comparative Ophthalmology, 4, 1994, 166). There is an historic perceived risk for the development of feline ocular post-traumatic sarcoma following gentamicin ciliary body ablation, and many clinicians refrain from chemical ablation in cats for this reason. A recent study discussed the possibility of a correlation between intravitreal gentamicin a

The Good, The Bad, and The Perplexing: Structural Alerts and Read-Across for Predicting Skin Sensitization Using Human Data.

In our earlier work (Golden et al., 2021), we showed 70-80% accuracies for several skin sensitization computational tools using human data. Here, we expanded the data set using the NICEATM human skin sensitization database to create a final data set of 1355 discrete chemicals (largely negative, ∼70%). Using this expanded data set, we analyzed model performance and evaluated mispredictions using Toxtree (v 3.1.0), OECD QSAR Toolbox (v 4.5), VEGA's (1.2.0 BETA) CAESAR (v 2.1.7), and a k-nearest-neighbor (kNN) classification approach. We show that the accuracy on this data set was lower than prev
