Binding of androgens including 5 alpha-dihydrotestosterone, testosterone and androstenedione stabilized the androgen receptor from rat submandibular gland with respect to its [3H]methyltrienolone binding activity. Nonandrogenic steroids, 0.5 microM of progesterone and estradiol-17 beta, or 5 microM of triamcinolone acetonide also satisfactorily stabilized the androgen receptor. These results suggest that the association of ligands, even if those ligands were not stereospecific for the androgen receptor, stabilizes the androgen receptor and prevents the loss of its binding capacity.

Elevated concentrations of peptide hormones have been described previously in human breast fluid. In the current study, the levels of cortisol, progesterone, testosterone, dihydrotestosterone, androsterone, androsterone sulfate, dehydroisoandrosterone, dehydroisoandrosterone sulfate, estradiol, estrone, estradiol sulfate, and estrone sulfate were measured. The levels of the four 17-ketosteroids and the two estrogen sulfates were markedly elevated over the plasma level, while that of the other compounds was the same or only slightly higher than the plasma levels of the same compounds.

Testosterone and its metabolite dihydrotestosterone are the libido hormones for the male, vital to sex drive and sexual function. Fear of loss of libido and orgasm is the main reason to retain at least one testis in male-to-female transsexuals during vaginoplasty. We report on four South American transsexual patients in whom we resected a remaining testis to illustrate the superfluity of retaining it. Because there are multiple reasons for castration, we advise that bilateral orchidectomy be performed in the course of sex reassignment surgery for male-to-female transsexuals.

Intracellular metabolism of testosterone was studied in in vitro cultured skin fibroblasts obtained from women with idiopathic hirsutism. A significantly higher rate of conversion of testosterone into 5-alpha-dihydrotestosterone was found in these women (4.78 +/- 2.08 fmol/microgram DNA/hour) as compared to that obtained for skin cultures of women without symptoms of androgenization (0.98 +/- 0.37 fmol/microgram DNA/hour). These results demonstrate that fibroblast culture may be useful in diagnosing the causes of hyperandrogenization in women with normal androgen secretion.
