Detection of Opisthorchis viverrini in infected bithynid snails by real-time fluorescence resonance energy transfer PCR-based method and melting curve analysis.

A real-time fluorescence resonance energy transfer (FRET) PCR combined with melting curve analysis was developed for the detection of Opisthorchis viverrini in experimentally infected bithynid snails, its first intermediate hosts. The test is based on the fluorescence melting curve analysis of a hybrid between an amplicon from the pOV-A6-specific probe sequence, a 162-bp repeated sequence specific to O. viverrini and specific fluorophore-labeled probes. The real-time FRET PCR could detect as little as a single cercaria artificially introduced in a pool of 30 non-infected snails. The O. viverri

Higher triplet excited states of benzophenones and bimolecular triplet energy transfer measured by using nanosecond-picosecond two-color/two-laser flash photolysis.

The lifetimes of benzophenone in the higher triplet excited state (BP(T(n))) and several BP derivatives in the T(n) states were measured directly to be tau(T(n))=37+/-7 ps and 20-33 ps, respectively, by using the nanosecond-picosecond (ns-ps) two-color/two-laser flash photolysis method. Based on the direct measurements of tau(T(n)) of BP(T(n)), the triplet energy transfer (TET) from BP(T(n)) to quenchers (Q), such as carbon tetrachloride (CCl4), benzene (Bz), and p-dichlorbenzene (DCB), was investigated. The fast TET from BP(T(n)) to Q can be attributed to the lifetime-dependent quenching proc

Unlocking the power of π-π stacking in mesogenic lanthanide complexes: a machine learning enhanced approach to controlling energy transfer and supramolecular assembly.

The rational design of highly luminescent lanthanide(III) complexes has long been hampered by the inability to simultaneously capture the subtle interplay of electronic structure, supramolecular packing, and thermally driven dynamics within these systems. Here, we shatter this barrier by introducing a multiscale computational framework that seamlessly integrates density functional theory, semiempirical methods, molecular dynamics, and state-of-the-art machine learning interatomic potentials - to achieve ab initio accuracy at scales previously inaccessible for lanthanide complexes. This approac

A paper-supported sandwich immunosensor based on upconversion luminescence resonance energy transfer for the visual and quantitative determination of a cancer biomarker in human serum.

In this paper, a paper-supported analytical device based on a sandwich immunoreaction and luminescence resonance energy transfer (LRET) was reported for the visual and quantitative determination of a cancer biomarker, in which upconversion nanoparticles (UCNPs) were located on the surface of the paper as energy donors and gold nanoparticles (AuNPs) were used as energy acceptors. Upon the recognition of the cancer biomarker by two rationally selected antibodies, the upconversion luminescence was quenched by the AuNPs in a biomarker concentration-dependent manner. As a model target, CEA was dete

Amplified electrochemiluminescence detection of DNA-binding protein based on the synergy effect of electron and energy transfer between CdS nanocrystals and gold nanoparticles.

In this work, we firstly enunciated the presence of synergy effect between the electron and energy transfer in an electrochemiluminescence (ECL) system involving the CdS NCs and Au NPs, based on which an amplified ECL biosensor was constructed for sensitive detection of DNA-protein interactions. Specifically, Au NPs modified on electrode, following by assembled probe DNA. Then the modified GCE was hybridized with SiO(2)@CdS/DNA conjugates. With DNA duplex as a rigid spacer, Au NPs could not only accelerate the electron transfer but also produce the surface plasma resonance upon excited ECL of

Förster resonance energy transfer (FRET) as a tool for dissecting the molecular mechanisms for maturation of the Shigella type III secretion needle tip complex.

Förster resonance energy transfer (FRET) provides a powerful tool for monitoring intermolecular interactions and a sensitive technique for studying Å-level protein conformational changes. One system that has particularly benefited from the sensitivity and diversity of FRET measurements is the maturation of the Shigella type III secretion apparatus (T3SA) needle tip complex. The Shigella T3SA delivers effector proteins into intestinal cells to promote bacterial invasion and spread. The T3SA is comprised of a basal body that spans the bacterial envelope and a needle with an exposed tip complex t

Fluorescence Resonance Energy Transfer Assay at the Crossroad: Urgent Reexamination of Assay Design for Severe Acute Respiratory Syndrome Coronavirus 2 Main Protease Inhibitors.

The main protease (Mpro) from coronaviruses represents an attractive therapeutic target for antiviral development. The fluorescence resonance energy transfer (FRET) assay is widely used for high-throughput screening (HTS) of Mpro inhibitors, but there has been a significant increase in false positives stemming from flawed assay design in previous studies. Here, we provide an overview of the FRET assay, discuss the key points of this method design, and highlight the corresponding solutions. We hope that this issue should receive increased attention from researchers.

Implementation of a fully automated ADAMTS13 activity assay utilizing fluorescence resonance energy transfer with a practical approach to address high background fluorescence interference.

OBJECTIVE: This study evaluates an automated fluorescence resonant energy transfer (FRET)-based ADAMTS13 activity assay on the Ceveron S100 instrument for the diagnosis of thrombotic thrombocytopenic purpura. It addresses the challenge of high background fluorescence (HBF), a known concern from our manual FRET assay, and proposes strategies to minimize erroneous results. METHODS: We compared FRET-Ceveron results with FRET-Manual (n = 100) and Technozym (Technoclone) enzyme-linked immunosorbent assay (ELISA) (n = 52) using retrospective and prospective patient samples collected throughout 2024

Switchable electrochemiluminescence aptasensor coupled with resonance energy transfer for selective attomolar detection of Hg2+ via CdTe@CdS/dendrimer probe and Au nanoparticle quencher.

In the present study, an ultrasensitive electrochemiluminescence (ECL) aptasensing assay for selective detection of Hg2+ was designed. In this electrochemiluminescence resonance energy transfer (ECL-RET) approach, Fe3O4@SiO2/dendrimers/QDs exhibited amplified ECL emissions (switch "on" state) and with the hybridization between T-rich ssDNA(S1) immobilized on the Fe3O4@SiO2/dendrimers/QDs and AuNPs modified with complementary aptamer (AuNPs-S2), the ECL of QDs nanocomposites was efficiently quenched (switch "off" state). In the presence of Hg2+ ions, formation of strong and stable T-Hg2+-T comp

CdS/TiO2-fluorescein isothiocyanate nanoparticles as fluorescence resonance energy transfer probe for the determination of trace alkaline phosphatase based on affinity adsorption assay.

The CdS/TiO(2)-fluorescein isothiocyanate (FITC) luminescent nanoparticles (CdS/TiO(2)-FITC) with the particle size of 20 nm have been synthesized by sol-gel method. CdS/TiO(2)-FITC could emit the fluorescence of both FITC and CdS/TiO(2). The fluorescence resonance energy transfer (FRET) occurred between the donor CdS/TiO(2) and the acceptor FITC in the CdS/TiO(2)-FITC. Taking advantages of the excellent characteristics of FRET, a new CdS/TiO(2)-FITC FRET labeling reagent and a CdS/TiO(2)-FITC-wheat germ agglutinin (CdS/TiO(2)-FITC-WGA) fluorescent probe have been developed. The FRET occurring

Fluorescence resonance energy transfer-based sensor Camui provides new insight into mechanisms of calcium/calmodulin-dependent protein kinase II activation in intact cardiomyocytes.

RATIONALE: Calcium/calmodulin-dependent protein kinase II (CaMKII) is a key mediator of intracellular signaling in the heart. However, the tools currently available for assessing dynamic changes in CaMKII localization and activation in living myocytes are limited. OBJECTIVE: We use Camui, a novel FRET-based biosensor in which full-length CaMKII is flanked by CFP and YFP, to measure CaMKII activation state in living rabbit myocytes. METHODS AND RESULTS: We show that Camui and mutant variants that lack the sites of CaMKII autophosphorylation (T286A) and oxidative regulation (CM280/1VV) serve a

Phenol formaldehyde resin nanoparticles loaded with CdTe quantum dots: a fluorescence resonance energy transfer probe for optical visual detection of copper(II) ions.

A novel fluorescence resonance energy transfer (FRET) system has been designed for the Cu2+ ions detection with optical visual assays. In this FRET reaction, the biocompatible, green luminescent monodisperse phenol formaldehyde resin nanoparticles (PFR NPs) synthesized by a simple hydrothermal method were used as the acceptor and the luminescent CdTe quantum dots (QDs) were selected as the donor. By the layer-by-layer method, the polyelectrolyte (PEI/PSS/PEI) were absorbed alternately on the surface of the PFR NPs. As a result, the amino groups were stably modified onto the surface of the PFR

Rapid detection of Dirofilaria immitis in mosquito vectors and dogs using a real-time fluorescence resonance energy transfer PCR and melting curve analysis.

A real-time fluorescence resonance energy transfer (FRET) PCR supplemented with melting curve analysis for the rapid molecular detection of Dirofilaria immitis in mosquito vectors and dog blood samples was developed. This real-time FRET PCR was based on the fluorescence melting curve analysis of a hybrid between an amplicon generated from the D. immitis ribosomal RNA gene sequence and specific fluorophore-labeled probes. The sensitivity, specificity, accuracy, and positive and negative predictive values of this method were all 100%. Besides being highly sensitive and specific, this PCR is fast

A Novel Inverse Algorithm To Solve the Integrated Optimization of Dose, Dose Rate, and Linear Energy Transfer of Proton FLASH Therapy With Sparse Filters.

PURPOSE: The recently proposed Integrated Physical Optimization Intensity Modulated Proton Therapy (IPO-IMPT) framework allows simultaneous optimization of dose, dose rate, and linear energy transfer (LET) for ultra-high dose rate (FLASH) treatment planning. Finding solutions to IPO-IMPT is difficult because of computational intensiveness. Nevertheless, an inverse solution that simultaneously specifies the geometry of a sparse filter and weights of a proton intensity map is desirable for both clinical and preclinical applications. Such solutions can reduce effective biologic dose to organs at

Effect of sulphate-limited growth on mitochondrial electron transfer and energy conservation between reduced nicotinamide-adenine dinucleotide and the cytochromes in Torulopsis utilis.

1. Conditions have been established for the sulphate-limited growth of Torulopsis utilis in continuous culture. 2. Mitochondria prepared from sulphate-limited cells lack both piericidin A sensitivity and the first energy-conservation site (site 1). Sensitivity to antimycin A or cyanide and the second and third energy-conservation sites were apparently unaffected by sulphate-limited growth. 3. Aerobic incubation for 8h of sulphate-limited cells with a low concentration of sulphate (50mum or less) resulted in the recovery of mitochondrial piericidin A sensitivity and site 1. The use of higher co

The Effect of Steric Hindrance on Triplet-Triplet Energy Transfer Rates: A Predictive Model Based on Spin-Density-Bearing van der Waals Surfaces.

Triplet-triplet energy transfer (TTET) via the Dexter mechanism is central to many photochemical applications, yet a quantitative structure-kinetic relationship for bimolecular quenching rate constants (kq) is still lacking. Here, using two photosensitizers and a series of anthracene-based acceptors with tailored steric profiles, we systematically investigate how nonconjugated substituents regulate kq. The most significant results are when the triplet energy gap exceeds ∼0.2 eV, kq is governed solely by steric hindrance and the suppression originates from spin-density-free units that block don

Cutaneous integration of a new wireless transcutaneous energy transfer system for durable long-term ventricular assist device systems in an experimental pig model.

Driveline infections remain a major complication of mechanical circulatory support (MCS) systems. This study evaluated the surgical implantation of a novel wireless transcutaneous energy transfer implant designed to eliminate driveline-associated infections by achieving durable cutaneous integration. Nine pigs were used across 4 trials to optimize implant geometry and surgical methods. Early prototypes caused skin necrosis due to excessive stretching, which was mitigated by increasing pocket size but led to seroma formation. A final implant design with reduced cross-sectional area and refined
