Targeting oncogenic acetyltransferase NAT10 to overcome anti-PD-1 resistance in nasopharyngeal carcinoma.

Targeted resistance to immune checkpoint therapy remains a clinical challenge, and a recent study reported by Xie et al provides novel insights into how RNA acetylation, particularly N4-acetylcytidine (ac4C), plays a pivotal role in shaping the tumor immune microenvironment in nasopharyngeal carcinoma (NPC). Through elucidation of the NAT10/DDX5/high mobility group box 1 axis, the authors demonstrate that enhanced ac4C modification suppresses CD4+ and CD8+ T-cell functionality, thereby facilitating tumor immune evasion and the resistance to anti-programmed cell death protein-1 therapy. The m6A methylome of SARS-CoV-2 in host cells.

The newly identified Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has resulted in a global health emergency because of its rapid spread and high mortality. The molecular mechanism of interaction between host and viral genomic RNA is yet unclear. We demonstrate herein that SARS-CoV-2 genomic RNA, as well as the negative-sense RNA, is dynamically N6-methyladenosine (m6A)-modified in human and monkey cells. Combined RIP-seq and miCLIP analyses identified a total of 8 m6A sites at single-base resolution in the genome. Especially, epidemic strains with mutations at these identified

The novel regulatory influence of Argonaute 1 on messenger RNA 5-methylcytosine modification in Arabidopsis thaliana.

5-Methylcytidine (m5C) modifications play a critical role in RNA metabolism. TRM4B (tRNA-specific methytransferase 4B) is the only known component of the plant m5C machinery that mediates mRNA (messenger RNA) methylation. To date, no regulators governing site-specific m5C modifications have been identified, apart from several degenerated sequence motifs. Drawing on relevant findings in animals, we investigated the involvement of AGO1 (Argonaute 1) in m5C regulation using an ago1 mutant of Arabidopsis. m5C-RIP-seq (m5C RNA immunoprecipitation followed by deep sequencing) identified high-confide

PRRC2A deficiency disrupts m6A-dependent RNA processing and meiosis in human spermatogenesis.

The formation of N6-methyladenosine (m6A) is the most prevalent internal RNA modification in eukaryotic transcripts and plays an essential role in spermatogenesis. PRRC2A is an m6A reader that is indispensable for meiotic progression and is essential for maintaining spermatocyte identity during mouse spermatogenesis. However, its contribution to human spermatogenesis remains unknown. In this study, we investigated a man diagnosed with non-obstructive azoospermia (NOA) and two with severe oligozoospermia, all carrying biallelic PRRC2A variants. All patients exhibited impaired spermatogenesis. T

METTL14 Hinders DCM Progression via Mediating the m6A Methylation of NRG4 in HG-Induced H9C2 Cells.

Diabetic cardiomyopathy (DCM) is a common form of cardiomyopathy that affects the cardiac muscle. It can lead to heart failure and threaten the life of human. Neuregulin 4 (NRG4) is a novel adipose factor released from brown adipose tissues and is considered to play an important role in metabolism, and affects the diabetic cardiomyopathy. N6-Methyladenosine (m6A) modification has been implicated in many bioprocesses. So we aim to explore the role of methyltransferase 14 (METTL14) and NRG4 in the progression of DCM. The high-glucose (HG) could increase the expression of NRG4. In HG-induced H9C2

m6A demethylase-driven reprogramming of leukemia-associated macrophages predicts improved outcomes in acute myeloid leukemia.

BACKGROUND: N6-methyladenosine (m6A) is a dynamic mRNA modification influencing transcript fate and cellular identity, especially in cancer. While oncogenic roles of m6A regulators in AML cells are known, their impact on the leukemic immune microenvironment is unclear. METHODS: In this study, we constructed a single-cell atlas of macrophages in AML by integrating publicly available scRNA-seq datasets from 129 patient cohorts. Data were batch-corrected using Seurat and Harmony. Macrophage subpopulations were identified, and the expression and activity of 29 m6A regulators were analyzed. Pseudo

Molecular Subtype Identification and Prognostic Prediction of Pancreatic Cancer Based on m6A/m5C/m1A-Related Genes.

Pancreatic cancer, characterized by an unfavourable prognosis, necessitates early diagnosis and prompt therapeutic intervention. This study aimed to delineate the functional implications of m6A/m1A/m5C-related genes in pancreatic carcinogenesis and establish a prognostic model. Utilizing data from the TCGA-PAAD cohort (n=178) combined with GTEx normal pancreatic tissues (n=332), we identified 43 genes associated with m6A/m1A/m5C methylation pathways. Significant expression differences in these genes were observed between neoplastic and non-neoplastic tissues, correlating with multiple biologic

Defective m6A RNA modification in macrophages exacerbates inflammation and promotes colitis-associated carcinogenesis by impairing efferocytosis.

Chronic inflammation resulting from unresolved tissue injury is a potent driver of tumorigenesis. Macrophages are essential for efferocytosis, the clearance of apoptotic cells, which prevents secondary necrosis and promotes inflammation resolution. However, the epigenetic mechanisms regulating this process under inflammatory stress remain incompletely understood. Here, we identify the m6A methyltransferase METTL3 as a critical regulator of macrophage efferocytosis. Using various models of tissue injury, METTL3 expression was significantly downregulated in macrophages under inflammatory conditi

Integrated analysis of m6A regulator mediated RNA methylation and key immune related genes in ischemic stroke.

The role of RNA N6-methyladenosine (m6A) modifications in modulating the immune microenvironment during ischemic stroke (IS) pathogenesis remains poorly characterized. This investigation systematically explores m6A-mediated immune regulation in IS and identifies critical immune-related biomarkers. Transcriptomic profiles from 108 IS samples were analyzed to discern m6A regulatory patterns. Single-sample gene set enrichment analysis (ssGSEA) and gene set variation analysis (GSVA) quantified immune cell infiltration and pathway activity across IS subtypes and controls. Cell-cycle-driven noncanonical ubiquitination of METTL14 orchestrates RNA methylation.

The precise temporal control of global mRNA methylation during the cell cycle is critical, yet the underlying mechanism remains unknown. Here we elucidate a noncanonical ubiquitination-mediated regulatory mechanism whereby RLIM deposits predominantly K27-linked ubiquitin (K27-Ub) on the m6A methyltransferase METTL14 in a cell-cycle-programmed manner. This modification modulates enzymatic activity by controlling METTL3-METTL14 complex assembly and RNA substrate engagement, independent of protein stability. We discover that transcriptome-wide mRNA m6A level, K27-Ub intensity on METTL14 and RLIM

m6A methylation regulatory network in pulmonary fibrosis: Molecular mechanisms from RNA stability to cellular phenotype transition (Review).

Pulmonary fibrosis (PF) is a progressive and often irreversible interstitial lung disease characterized by aberrant wound healing, excessive extracellular matrix deposition and distortion of lung architecture. N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, dynamically regulates RNA metabolism through m6A writers, erasers and readers. Evidence indicates that m6A dysregulation contributes to PF by modulating fibroblast activation, epithelial injury and senescence, epithelial-mesenchymal plasticity, macrophage-associated inflammation, oxidative stress respon

PUS1 Drives Renal Cancer Progression by Preventing Formation of Endogenous Double-stranded RNAs.

Pseudouridine (Ψ) modification is a prevalent epitranscriptomic mark with critical roles in carcinogenesis; however, the function of its catalytic "writer" enzyme, pseudouridine synthase 1 (PUS1), in renal cell carcinoma (RCC) remains elusive. Our analysis revealed that PUS1 mRNA is upregulated in RCC and is associated with an unfavorable prognosis. Strikingly, this transcriptional upregulation results in a concomitant and exclusive increase in the protein abundance of PUS1 isoform 2. Mechanistically, although PUS1 markedly enhances global mRNA translation, this effect is not directly mediated

NAT10 aggravates psoriasis by promoting keratinocytes fatty acid synthesis via stable FASN transcription.

RNA modifications have emerged as critical regulators of cellular function and disease pathogenesis; however, their contribution to psoriasis remains unclear. Here, we report that N-acetyltransferase 10 (NAT10) expression is markedly elevated in the epidermis of patients with psoriasis and in imiquimod (IMQ)-induced lesions. Single-cell RNA sequencing further identifies NAT10 upregulation in hyperproliferative and basal keratinocyte subsets. Keratinocyte-specific deletion of NAT10 attenuates IMQ-induced psoriatic phenotypes in mice. Integrative profiling of RNA sequencing and N4-acetylcytidine

YTHDF1-mediated translational activation of SETD2 suppresses aerobic glycolysis and sensitizes renal cell carcinoma to sunitinib.

Renal cell carcinoma (RCC) is a highly aggressive malignancy often characterized by metabolic rewiring and limited long-term responses to targeted therapies. In this study, we unveil a previously uncharacterized tumor-suppressive paradigm driven by the m6A reader YTHDF1. Through multi-omics integration and clinical validation, we demonstrate that YTHDF1 is downregulated in RCC and serves as a clinically relevant biomarker associated with favorable clinicopathological features. Mechanistically, YTHDF1 functions in an m6A-dependent manner to directly promote the translational efficiency of the e

Transcriptional and post-transcriptional convergence of HES1 and IGF2BP2 sustains CSF2-dependent metabolic adaptability in gastric Cancer.

This study delineates a hierarchical signaling axis driving gastric cancer (GC) progression through integrated transcriptomic and functional analyses. Single-cell sequencing and TCGA data identified CSF2 as a key oncogene, with elevated expression correlating with poor prognosis. Mechanistically, the transcription factor HES1 directly activates IGF2BP2 transcription, as confirmed by chromatin immunoprecipitation and dual-luciferase assays. IGF2BP2 subsequently stabilizes CSF2 mRNA via N6-methyladenosine (m6A) modification, validated through RNA immunoprecipitation and mRNA decay kinetics. Func

RNA cytidine acetyltransferase NAT10 maintains T cell pathogenicity in inflammatory bowel disease.

The emerging field of epitranscriptomics is reshaping our understanding of post-transcriptional gene regulation in inflammatory diseases. N4-acetylcytidine (ac4C), the only known acetylation modification in RNA catalyzed by N-acetyltransferase 10 (NAT10), is known to enhance mRNA stability and translation, yet its role in inflammatory bowel disease (IBD) remains unclear. In this study, we discovered that Nat10 expression correlates with inflammatory and apoptotic pathways in human ulcerative colitis CD4+ T cells. Our further analysis revealed that the deficiency of NAT10 led to a disruption of

RNA epigenetic modifications as dynamic biomarkers in cancer: from mechanisms to clinical translation.

RNA modifications are crucial for post-transcriptional gene regulation. Research on RNA modifications has become a novel frontier of epitranscriptomics. Up to now, over 170 kinds of modifications have been identified on mRNA and diverse non-coding RNA. Three classes of proteins (writers, erasers, and readers) regulate the addition, removal, and identification of epigenetic marks, thus affecting RNA biological functions. Increasing evidence identifies the dysregulation of RNA modifications in different cancer types and the therapeutic potential of targeting RNA-modifying enzymes. The ability of

Uncovering a role for METTL13 in malignant transformation of human hematopoietic stem cells and in the progression of pediatric leukemia.

Post-transcriptional RNA modifications, such as N6-methyladenosine (m6A) methylation and adenosine to inosine (A-to-I) editing, are critical regulators of hematopoietic stem cell (HSC) self-renewal and differentiation, yet their precise contributions to malignant transformation are not fully elucidated. In this study, we uncovered the epitranscriptomic landscape caused by knockdown of genes from the methyltransferase (METTL)-family in hematopoietic stem and progenitor cells (HSPCs). We identified both converging and distinct roles of METTL3 and METTL14, known members of the m6A writer complex,

Emerging Dual Roles of ALKBH5 in Ischemia-reperfusion Injury: Mechanisms and Therapeutic Implications.

Ischemia-reperfusion injury (IRI) is a critical pathological process affecting multiple organs, including the heart, brain, kidney, and liver. Recent studies have highlighted the dual roles of AlkB homolog 5 (ALKBH5), an N6-methyladenosine (m⁶A) RNA demethylase, in regulating cellular responses to IRI through epigenetic mechanisms. This review synthesizes current evidence on ALKBH5 involvement in metabolic reprogramming, autophagy, apoptosis, pyroptosis, and inflammation across organ-specific IRI models. We establish a unifying framework in which ALKBH5 functions as a double-edged sword, exert
