α-Synuclein Promotes Atherosclerosis by Impairing Macrophage Autophagic Flux.

Atherosclerosis, recognized as a prototypical age-associated vascular disease, is driven by macrophage foam cell formation. Individuals with Parkinson's disease (PD) exhibit an elevated risk for stroke. While α-synuclein (α-Syn) is known for its neuronal role in PD, its presence in the circulation and potential vascular effects remain unexplored. Here, we identify α-Syn as a previously unrecognized circulating factor that promotes atherosclerosis. α-Syn was detected in human atherosclerotic plaques. Genetic deletion of α-Syn reduced atherosclerotic lesion size and macrophage lipid accumulation

Lipophagic panniculitis in re-excision specimens.

Lipophagic panniculitis consists of a macrophage infiltrate in the subcutaneous tissue. The macrophages transform into foam cells within the panniculus; they replace lipocytes and may form giant cells. Although those pathologic features have been described as diagnostic of Weber-Christian disease, we report the occurrence of lipophagic panniculitis in re-excision specimens. Among 252 re-excision specimens from previously biopsied skin tumors, 5 cases in which masses of lipophages were infiltrating and replacing the subcutaneous tissue were found. The infiltrate was localized to the deep dermis…

Numerous intertriginous xanthomas in infant: A diagnostic clue for sitosterolemia.

Sitosterolemia is a very rare autosomal recessive lipoprotein metabolic disorder caused by homozygous or compound heterozygous mutations in … olesterolemia and elevated plant sterol levels. Histopathologically, xanthoma showed aggregation of foam cells in the dermis with a zone of mucin deposits in the dermal papilla. Electron microscopy showed numerous membrane-bound lipid droplets and multivesicular lipid bodies in the foam cells, a round cell containing lipid droplets in the basal cell layer an …

Non-coding RNA-Associated Therapeutic Strategies in Atherosclerosis.

Atherosclerosis has been the main cause of disability and mortality in the world, resulting in a heavy medical burden for all countries. It is widely known to be a kind of chronic inflammatory disease in the blood walls, of which the key pathogenesis is the accumulation of immunologic cells in the lesion, foam cells formation, and eventually plaque rupture causing ischemia of various organs. Non-coding RNAs (ncRNAs) play a vital role in regulating the physiologic and pathophysiologic processes in cells. More and more studies have revealed that ncRNAs also participated in the development of ath…

Scavenger receptors and modified lipoproteins: fatal attractions?

Lipoproteins modified by oxidation, glycation, alkylation, and nitration are generated by oxidative stress during inflammation, diabetes, an … d expression of scavenger receptors, may lead to cellular deregulation, apoptosis, and formation of foam cells. Diseases accompanied by oxidation of lipoproteins, such as atherosclerosis, Alzheimer disease, glomerulosclerosis, ataxia with vitamin E deficiency, and possibly age-dependent lipofuscin deposition, may share a common pathogenetic feature. This rev …

Monocyte Recruitment, Specification, and Function in Atherosclerosis.

Atherosclerotic lesions progress through the continued recruitment of circulating blood monocytes that differentiate into macrophages within plaque. Lesion-associated macrophages are the primary immune cells present in plaque, where they take up cholesterol and store lipids in the form of small droplets resulting in a unique morphology termed foam cell. Recent scientific advances have used single-cell gene expression profiling, live-cell imaging, and fate mapping approaches to describe macrophage and monocyte contributions to pro- or anti-inflammatory mechanisms, in addition to functions of mo

Experimental study of fat grafting under negative pressure for wounds with exposed bone.

BACKGROUND: The combination of fat grafting and negative pressure (VAC) therapy represents a synergistic interaction of all essential components for wound healing. The aim of this experimental study was to determine whether it could promote healing of wounds with exposed bone. METHODS: Full-thickness wounds with denuded bone in Sprague-Dawley rats were treated with either polyurethane foam dressing, fat grafting alone, polyurethane foam dressing with VAC, or polyurethane foam dressing with VAC combined with a single, or two administrations of fat graft. Wound healing kinetics, tissue growth,

Macrophages in Atherosclerosis, First or Second Row Players?

Macrophages represent a cell type that has been widely described in the context of atherosclerosis since the earliest studies in the 17th century. Their role has long been considered to be preponderant in the onset and aggravation of atherosclerosis, in particular by participating in the establishment of a chronic inflammatory state by the release of pro-inflammatory cytokines and by uncontrolled engorgement of lipids resulting in the formation of foam cells and later of the necrotic core. However, recent evidence from mouse models using an elegant technique of tracing vascular smooth muscle c…

Ketoconazole: a review of a workhorse antifungal molecule with a focus on new foam and gel formulations.

Topical ketoconazole, an imidazole, has been a mainstay of treatment for fungal infections and seborrheic dermatitis. Ketoconazole interferes with the fungal synthesis of ergosterol, a constituent of cell membranes, and has other antifungal effects. Shampoo, cream and oral formulations of ketoconazole have been available for many years. Recently, new foam and gel formulations of ketoconazole have been brought to market for the treatment of seborrheic dermatitis, which are effective against superficial fungal and yeast infections. The foam and gel formulations are likely to lead to a renascence

Mechanical properties of highly porous PEEK bionanocomposites incorporated with carbon and hydroxyapatite nanoparticles for scaffold applications.

Bone regeneration is of great importance worldwide, because of various bone diseases, such as infections, tumors, and resultant fracture, birth defects, and bone loss due to trauma, explosion, or accident. Bone regeneration can be achieved by several materials and templates manufactured through various fabrication techniques. Uses of different materials and scaffold fabrication techniques have been explored over the past 20 years. In this research, polyetheretherketone (PEEK) was used to fabricate highly porous bionanocomposite foams for bone scaffolding. Melt casting and salt porogen (200-500

Biomechanical evaluation of calcium phosphate-based nanocomposite versus polymethylmethacrylate cement for percutaneous kyphoplasty.

BACKGROUND CONTEXT: Polymethylmethacrylate (PMMA) is the most commonly used filling material when performing percutaneous kyphoplasty (PKP) for the treatment of osteoporotic vertebral compression fractures. However, there are some inherent and unavoidable drawbacks with the clinical use of PMMA. PMMA bone cement tends to leak during injection, which can lead to injury of the spinal nerves and spinal cord. Moreover, the mechanical strength of PMMA-augmented vertebral bodies is extraordinary and this high level of mechanical strength might predispose to adjacent vertebral fractures. A novel biod

A bioengineering investigation of cervical collar design and fit: Implications on skin health.

BACKGROUND: Cervical collars restrict cervical spine movement to minimise the risk of spinal cord injury. Collars apply mechanical loading to the skin putting it at risk of skin damage. Indeed, cervical collar-related pressure ulcers are unacceptably prevalent, especially at the occiput, mandibles, and chin. Collar design and fit are often key considerations for prevention. METHODS: This comprehensive study evaluated four commercial prehospital and acute care cervical collars. Pressure, microclimate, transepidermal water loss and skin hydration were measured at the interface between the devic

Alginate-Based Solid Foam Incorporating Rügen Chalk: A Novel Platform for Modern Application of Peloids.

Background/Objectives: Natural calcium carbonate materials such as Rügen chalk have a long history of use in balneology and rehabilitation, particularly for musculoskeletal disorders, yet their application remains largely confined to traditional, labour-intensive forms such as powders, suspensions, and packs, which limit usability and broader clinical translation. This study aimed to develop an alginate-based solid foam incorporating Rügen chalk and to evaluate how key formulation components influence its structural, mechanical, and thermal properties relevant for therapeutic use. Methods: Alg

From smooth to 'breaking bad': vascular smooth muscle cell phenotypic switching at the heart of plaque vulnerability.

PURPOSE OF REVIEW: Atherosclerosis persists despite LDL-lowering therapies, highlighting lipid-independent drivers of disease. This review examines vascular smooth muscle cell (VSMC) plasticity as a central determinant of plaque progression and stability and evaluates emerging therapeutic strategies targeting VSMC phenotypic transitions. RECENT FINDINGS: VSMCs exhibit marked phenotypic plasticity, contributing to up to 50% of plaque foam cells and generating diverse states including macrophage-like, fibromyocyte, osteogenic, and intermediate populations. Key regulators such as Krüppel-like fa…

An advanced method for quantitative measurements of cholesterol crystallization.

BACKGROUND: Cholesterol crystallization within an atherosclerotic plaque significantly contributes to the acceleration of plaque rupture - a problematic event due to the current lack of specific treatments to prevent such formations. Modelling this pathogenic process is also difficult due to the lack of suitable experimental models that enable quantitative analysis of crystal formation and bioactivity screening of potential therapeutic compounds. AIM: To develop an in vitro human cell model of cholesterol crystallization combined with an imaging system that incorporates both quantitative anal

Compressive Properties and Degradable Behavior of Biodegradable Porous Zinc Fabricated with the Protein Foaming Method.

A new protein foaming-consolidation method for preparing porous zinc was developed using three proteins (egg white protein (EWP), bovine bone collagen protein (BBCP), and fish bone collagen protein (FBCP)) as both consolidating and foaming agents. The preparation route utilized powder mixing and sintering processing, which could be divided into three steps: slurry preparation, low-temperature foaming, and high-temperature sintering. The morphological characteristics of the pore structures revealed that the porous zinc had an interconnected open-cell structure. Compared to the porous zinc prepa
