cyp51A-based mechanism of azole resistance in Aspergillus fumigatus: Illustration by a new 3D Structural Model of Aspergillus fumigatus CYP51A protein.

Mutations of CYP51A protein (Cytochrome P450 14-α Sterol demethylase) play a central role in the azole resistance of Aspergillus fumigatus The available structural models of CYP51A protein ofA. fumigatus are built based on that of Homo sapiens and that of Mycobacterium tuberculosis, of which the amino acid homology is only 38% and 29% compared with CYP51A protein ofA. fumigatus, respectively. In the present study, we constructed a new 3D structural model ofA. fumigatus CYP51A protein based on a recently resolved crystal structure of the homologous protein in the fungus S. cerevisiae, which sha

Parallel beta-helix proteins required for accurate capsule polysaccharide synthesis and virulence in the yeast Cryptococcus neoformans.

The principal capsular polysaccharide of the opportunistic fungal pathogen Cryptococcus neoformans consists of an alpha-1,3-linked mannose backbone decorated with a repeating pattern of glucuronyl and xylosyl side groups. This structure is critical for virulence, yet little is known about how the polymer, called glucuronoxylomannan (GXM), is faithfully synthesized and assembled. We have generated deletions in two genes encoding predicted parallel beta-helix repeat proteins, which we have designated PBX1 and PBX2. Deletion of either gene results in a dry-colony morphology, clumpy cells, and dec

Cryptococcus neoformans invasion of the orbital compartment and periocular spaces following intracerebral infection in mice.

Cryptococcus neoformans is an encapsulated and neurotropic fungus known for causing life-threatening meningoencephalitis in immunocompromised individuals. We used a murine intracerebral infection model to study C. neoformans brain infection and microglia-fungal interactions. Mice infected with the C. neoformans H99 strain exhibited hydrocephalus, cryptococcoma formation, and significantly higher brain fungal burden than the acapsular cap59 strain at 7-days post infection (dpi). Immunofluorescence imaging showed robust microgliosis, especially near cryptococcomas. Cytokine analysis revealed a h

Fungal infection of the ear. Etiology and therapy with bifonazole cream or solution.

Fungal infections of the auditory canal and middle ear are usually left unnoticed and regarded as a harmless saprophytic growth. However, these infections may become clinically significant in immunocompromised patients or patients undergoing long-term antibiotic treatment. This is demonstrated by characteristic case reports and confirmed by light and electron microscopic findings. Moreover, studies on animals with experimentally induced fungal infections have furnished evidence for the risk of the infection's spreading to the inner ear and causing serious damage to the organ of Corti; indirect

Strasseriolides A-D, A Family of Antiplasmodial Macrolides Isolated from the Fungus Strasseria geniculata CF-247251.

A novel family of four potent antimalarial macrolides, strasseriolides A-D (1-4), has been isolated from cultures of Strasseria geniculata CF-247251, a fungal strain obtained from plant tissues. The structures of these compounds, including their absolute configurations, were elucidated by HRMS, NMR spectroscopy, and X-ray single-crystal diffraction. The four compounds gave respective IC50 values of 9.810, 0.013, 0.123, and 0.128 μM against Plasmodium falciparum 3D7 parasites with no significant cytotoxicity against the HepG2 cell line.

Discovery of a new P450 enzyme DkpdB for cyclo(Trp-Phe) dimerization in the marine strain Aspergillus sydowii MNP-2.

Diketopiperazine (DKP) dimers are one of the important microbial natural products owing to their structural diversity and a broad spectrum of biological properties. However, few studies on fungal P450-catalyzed DKP dimerization have been reported. Herein, a novel cytochrome P450 monooxygenase DkpdB was originally discovered in the marine-derived fungus Aspergillus sydowii MNP-2 and shown to catalyze the formation of the DKP dimer WIN 64821 from cyclo(Trp-Phe) through a proposed C3-C3' oxidative coupling reaction. Heterologous expression in Aspergillus nidulans RJMP1.5 together with in vivo and

Antifungal drug resistance in Candida: a special emphasis on amphotericin B.

Invasive fungal infections in humans caused by several Candida species, increased considerably in immunocompromised or critically ill patients, resulting in substantial morbidity and mortality. Candida albicans is the most prevalent species, although the frequency of these organisms varies greatly according to geographic region. Infections with C. albicans and non-albicans Candida species have become more common, especially in the past 20 years, as a result of aging, immunosuppressive medication use, endocrine disorders, malnourishment, extended use of medical equipment, and an increase in imm

Fungi between threat and promise: global perspectives on health and innovation.

Fungi play a dual role as indispensable ecological engineers and as major agents of disease in humans, animals, and plants. Recent estimates highlight their substantial impact, with millions of invasive infections annually and severe agricultural losses threatening food security. At the same time, fungi underpin ecosystem services such as decomposition, soil aggregation, and carbon sequestration, while also serving as prolific sources of enzymes, metabolites, and sustainable biomaterials. Advances in single-cell and spatial omics, cryo-electron microscopy, AlphaFold-based structural prediction

Structural analysis of mycobacterial homoserine transacetylases central to methionine biosynthesis reveals druggable active site.

Mycobacterium tuberculosis is the cause of the world's most deadly infectious disease. Efforts are underway to target the methionine biosynthesis pathway, as it is not part of the host metabolism. The homoserine transacetylase MetX converts L-homoserine to O-acetyl-L-homoserine at the committed step of this pathway. In order to facilitate structure-based drug design, we determined the high-resolution crystal structures of three MetX proteins, including M. tuberculosis (MtMetX), Mycolicibacterium abscessus (MaMetX), and Mycolicibacterium hassiacum (MhMetX). A comparison of homoserine transacety

Histological considerations in implant patients.

The selection of patients for cochlear implantation requires consideration of a variety of anatomical and histological facts. Mechanical impediments to cochlear implantation, including the consequences of chronic otitis media, bony obliteration of the membranous labyrinth, or labyrinthitis ossificans, following bacterial labyrinthitis or vascular compromise of the inner ear, obliterative otosclerosis, and middle-ear dysplasias may present difficulties in access to the inner ear and placement of electrodes. Consideration of the histopathological features of known causes of profound deafness wil

Targeting multidrug-resistant Candida: CuxTe nanoflowers synergistically facilitate fungal clearance via cuproptosis death and host immune activation.

Candida species are common infectious pathogens that can cause invasive infections with potentially life-threatening consequences. Current treatments for invasive Candida infections are primarily azoles and echinocandins. However, emerging antifungal drug resistance compromises therapeutic efficacy. Herein, a stable CuxTe nanoflower was designed to combat fungal infections via cuproptosis death and immune activation. Firstly, Cu and Te precursors were added to an auxiliary reducing agent and solvent, and CuxTe nanoflowers with a multibranched dendritic structure were synthesized via a temperat

Cows' reproductive performances and parity order influences the cervicovaginal fungal community.

Fungi have already been described as etiological agents of reproductive diseases such as endometritis and infertility in cows. However, few studies have been developed to elucidate the entire cervicovaginal fungal communities in cows. Therefore, our study aimed to characterize the fungal community present in the cervix of cows with different reproductive performances. Cervicovaginal mucus was collected from 36 Angus breed cows (1.5-12 years old) on a commercial beef cattle ranch. Twenty-one cows had a history of infertility in the year prior to the collection, showing early return to estrus. T

Preliminary study of the lethal fungus Candida albicans phosphofructokinase-1 as a potential therapeutic target.

Our previous research revealed that phosphofructokinase-1 (Pfk1) is a key regulator in fungal virulence [1]. However, the unclear structural characterization and catalytic mechanisms hinder the research on whether it can become a potential antifungal drug target. Here, we report the first purification of Pfk1 from the lethal fungal C. albicans using affinity and ion exchange chromatography, yielding two 100 kDa protein bands. Mass spectrometry identified these bands as the α- and β-subunits. Interestingly, the α subunit has serine at position 164, which differs from the asparagine shown in the

Synthesis of 4-(Diethylamino)salicylaldehyde Derivatives and Research on Their Antifungal Mechanism against Rhizoctonia solani.

To enhance the antifungal activity of 4-(diethylamino)salicylaldehyde (DSA), 20 salicylaldehyde Schiff base compounds (L1-L20), including nine novel derivatives, were synthesized and characterized by MS, 1H NMR, and 13C NMR. In vitro assays identified compound L5 as the most active agent, with an EC50 of 10.797 μg/mL (2-fold higher than DSA), and pot experiments confirmed its efficacy against cotton damping-off disease. Mechanistic investigations via electron microscopy, transcriptomics, physiological assays and molecular docking suggest that L5 may target fatty acid synthase (FAS) to interfer

Discovery of Thiram-Inspired Thiocarbamate Derivatives as Novel Laccase Inhibitors against Sclerotium rolfsii.

Peanut stem rot, caused by the soil-borne pathogen Sclerotium rolfsii, poses a significant threat to global peanut production and results in substantial yield losses. A series of thiocarbamate derivatives were synthesized and evaluated against ten phytopathogenic fungi. Over 50% showed broad-spectrum fungicidal activity at 40 mg/L. Notably, compound b6 exhibited superior activity against S. rolfsii (EC50 = 0.75 mg/L), surpassing Thiram (EC50 = 0.91 mg/L). In vivo assays confirmed robust protective and curative efficacies on peanut seedlings. b6 also demonstrated potent laccase inhibition (IC50

Discovery of Novel Cyanoacrylates as Potential Myosin I Inhibitors with Remarkable Activity against Fusarium Species.

Fusarium head blight (FHB) is a devastating wheat disease causing yield losses and mycotoxin contamination. Phenamacril, a myosin I-targeting fungicide, has a limited antifungal spectrum. Using structure-based drug design focusing on M375 in Fusarium graminearum myosin I (FgMyoI), we synthesized 25 phenamacril derivatives. The II-2 emerged as the most potent inhibitor (EC50 = 0.0077 mg/L against F. graminearum), with broad-spectrum activity against various phytopathogens. Biochemical and computational analyses confirmed that II-2 tightly binds FgMyoI and inhibits its ATPase activity. In vivo t

Structural determinants for subnanomolar inhibition of the secreted aspartic protease Sapp1p from Candida parapsilosis.

Pathogenic Candida albicans yeasts frequently cause infections in hospitals. Antifungal drugs lose effectiveness due to other Candida species and resistance. New medications are thus required. Secreted aspartic protease of C. parapsilosis (Sapp1p) is a promising target. We have thus solved the crystal structures of Sapp1p complexed to four peptidomimetic inhibitors. Three potent inhibitors (Ki: 0.1, 0.4, 6.6 nM) resembled pepstatin A (Ki: 0.3 nM), a general aspartic protease inhibitor, in terms of their interactions with Sapp1p. However, the weaker inhibitor (Ki: 14.6 nM) formed fewer nonpolar
