Fusarium oxysporum and Aspergillus sp. as Keratinase Producers Using Swine Hair From Agroindustrial Residues.

Technological processes mediated by microorganisms and enzymes are promising alternatives for treatment of recalcitrant residues. Keratinases hydrolyze keratin, the primary component of some wastes generated in many industrial activities. The present study was designed to evaluate strategies for obtaining keratinases produced by fungi using submerged fermentation and two residues as substrates, chicken feathers and swine hair. Two fungi isolated from feather residues showed potential for keratinase production, Fusarium oxysporum and Aspergillus sp. These were subjected to submerged fermentatio…

Synergistic association of clioquinol with antifungal drugs against biofilm forms of clinical Fusarium isolates.

BACKGROUND: The influence of biofilm on the complexity of fungal diseases has been reported in recent years, especially in non-invasive mycoses such as keratitis and onychomycosis. The difficulty in treating cases of fusariosis in the human medical clinic exemplifies this situation, because when Fusarium spp. are present in the form of biofilm, the permeation of antifungal agents is compromised. OBJECTIVES: This study proposes an association of clioquinol, an inhibitor of fungal cells with antifungal drugs prescribed to combat fusariosis in humans. METHODS: Susceptibility was assessed by mic…

A Disseminated Fusarium fujikuroi Species Complex Infection Prior to Allogeneic Hematopoietic Stem Cell Transplantation.

A 53-year-old man was diagnosed with acute myeloid leukemia, which was refractory to chemotherapies. Systemic papules appeared afterward. The skin biopsies revealed filamentous fungal infection including fusariosis. Despite antifungal therapy, the infection did not resolve, because neutropenia persisted with the leukemia. He underwent hematopoietic stem cell transplantation (HSCT) to overcome the leukemia and restore normal hematopoiesis but died from fusariosis just before engraftment. Fusarium fujikuroi species complex was detected in blood cultures with poor antifungal susceptibility. Becau…

Field pathogenomics of Fusarium head blight reveals pathogen transcriptome differences due to host resistance.

Fusarium head blight (FHB), caused by Fusarium graminearum and other Fusarium species, is a detrimental disease that affects small grains such as wheat around the world. Management of FHB is difficult, and surveillance as well as a better understanding of pathogen aggressiveness is needed for improved control. F. graminearum disease severity varies depending on the resistance of the host genotype. In this study, we used the field pathogenomics method to investigate gene expression and population structure of isolates collected from wheat lines of varying resistance levels (susceptible, interme…

Fusaricidin Produced by Paenibacillus polymyxa WLY78 Induces Systemic Resistance against Fusarium Wilt of Cucumber.

Cucumber is an important vegetable crop in China. Fusarium wilt is a soil-borne disease that can significantly reduce cucumber yields. Paenibacillus polymyxa WLY78 can strongly inhibit Fusarium oxysporum f. sp. Cucumerium, which causes Fusarium wilt disease. In this study, we screened the genome of WLY78 and found eight potential antibiotic biosynthesis gene clusters. Mutation analysis showed that among the eight clusters, the fusaricidin synthesis (fus) gene cluster is involved in inhibiting the Fusarium genus, Verticillium albo-atrum, Monilia persoon, Alternaria mali, Botrytis cinereal, and …

Killing of diverse eye pathogens (Acanthamoeba spp., Fusarium solani, and Chlamydia trachomatis) with alcohols.

BACKGROUND: Blindness is caused by eye pathogens that include a free-living protist (Acanthamoeba castellanii, A. byersi, and/or other Acanthamoeba spp.), a fungus (Fusarium solani), and a bacterium (Chlamydia trachomatis). Hand-eye contact is likely a contributor to the spread of these pathogens, and so hand washing with soap and water or alcohol-based hand sanitizers (when water is not available) might reduce their transmission. Recently we showed that ethanol and isopropanol in concentrations present in hand sanitizers kill walled cysts of Giardia and Entamoeba, causes of diarrhea and dysen…

Molecular characterization, fitness and mycotoxin production of Fusarium graminearum laboratory strains resistant to benzimidazoles.

Six benzimidazole (BMZ)-resistant Fusarium graminearum strains were obtained after UV mutagenesis and selection on carbendazim (MBC)-amended medium. In vitro bioassays resulted in the identification of two resistant phenotypes that were highly HR (Rf: 40-170, based on EC50) and moderately MR (Rf: 10-20) resistant to carbendazim. Cross resistance studies with other fungicides showed that all mutant strains tested were also resistant to other BMZs, such as benomyl and thiabendazole, but retained their parental sensitivity to fungicides belonging to other chemical groups. A point mutation at codo…

Pathogen induced changes in the protein profile of human tears from Fusarium keratitis patients.

Fusarium is the major causative agent of fungal infections leading to corneal ulcer (keratitis) in Southern India and other tropical countries. Keratitis caused by Fusarium is a difficult disease to treat unless antifungal therapy is initiated during the early stages of infection. In this study tear proteins were prepared from keratitis patients classified based on the duration of infection. Among the patients recruited, early infection (n = 35), intermediate (n = 20), late (n = 11), samples from five patients in each group were pooled for analysis. Control samples were a pool of samples from …

Targeting iron acquisition blocks infection with the fungal pathogens Aspergillus fumigatus and Fusarium oxysporum.

Filamentous fungi are an important cause of pulmonary and systemic morbidity and mortality, and also cause corneal blindness and visual impa … erophore deferroxamine enhances fungal growth in infected mice. By examining mutant Aspergillus and Fusarium strains, we found that fungal transcriptional responses to low iron levels and extracellular siderophores are essential for fungal growth during infection. Further, we showed that targeting fungal iron acquisition or siderophore biosynthesis by topi …

Cellulitis and nodular skin lesions due to Fusarium spp in liver transplant: case report.

Fusariosis is one of the emerging invasive fungal infections over the last decade. However, its recent rise has been in its ability to produce disseminated infection in severely immunosuppressed patients with neutropenia. In solid organ transplantation, fusariosis remains an uncommon picture mainly with nodules, subcutaneous abscesses, ulcers, or necrotic skin lesions resembling erthyma gangrenosum. Herein, we have reported a case of cellulitis, subcutaneous nodules, and abscesses due to Fusarium spp in a liver transplantation patient who was successfully treated with polyenes and surgical res…

Nep1 Protein from Fusarium oxysporum Enhances Biological Control of Opium Poppy by Pleospora papaveracea.

ABSTRACT The fungus Pleospora papaveracea and Nep1, a phytotoxic protein from Fusarium oxysporum, were evaluated for their biocontrol potential on opium poppy (Papaver somniferum). Four treatments consisting of a control, P. papaveracea conidia, Nep1 (5 mug/ml), and P. papaveracea conidia plus Nep1 (5 mug/ml) were used in detached-leaf and whole-plant studies. Conidia of P. papaveracea remained viable for 38 days when stored at 20 or 4 degrees C. Nep1 was stable in the presence of conidia for 38 days when stored at 4 degrees C or for 28 days at 20 degrees C. The presence of Nep1 did not affect…

The role of Bacillus marisflavi in enhancing wheat crown rot through Fusarium pseudograminearum activation.

While many root-associated bacteria promote plant growth or induce resistance, Bacillus marisflavi interaction enhanced wheat crown rot (WCR) caused by Fusarium pseudograminearum. Unlike typical beneficial Bacillus species, B. marisflavi did not directly affect wheat growth but significantly increased pathogen colonization of roots and stem bases, exacerbating WCR incidence. This occurred via activation of F. pseudograminearum genes linked to growth and pathogenicity. The B. marisflavi metabolite D-mannitol promoted proliferation of F. pseudograminearum and related Fusarium spp., elevated myco…

Cold-hardy interspecific hybrid grape-associated Fusarium species are pathogenic on grapevines and barley.

Cold-hardy interspecific hybrid grapevines (CIHG; Vitis spp.) have expanded grape production in Northern regions of the United States, but mature vineyards in the upper Midwest are increasingly showing grapevine trunk disease-like symptoms, including cordon dieback, skipped spurs, stunted shoots, and vascular discoloration. Previous surveys recovered Fusarium spp. at high frequency from symptomatic CIHG tissues, but their roles as causal agents for vine decline remained unclear. In this study, we characterized Fusarium isolates recovered from symptomatic CIHGs in Iowa vineyards and evaluated t…

Fusarium verticillioides seed inoculation modifies maize cell wall responses to subsequent Sesamia nonagrioides infestation.

Maize resistance to stem borers and fungal infection is strongly influenced by cell wall composition, but little is known about how prior fungal seed colonization affects subsequent insect-induced cell wall responses. Therefore, this study evaluated whether seed inoculation with Fusarium verticillioides modifies constitutive and induced cell wall defences against Sesamia nonagrioides infestation in maize. Seven maize inbred lines were grown under four treatments combining the presence or absence of F. verticillioides seed inoculation and S. nonagrioides infestation. Tunnel length, lignin conte…

Exploring the potential of naked barley to manage deoxynivalenol accumulation from Fusarium head blight.

Developing and deploying multi-use naked barley varieties that accumulate lower levels of deoxynivalenol (DON) resulting from Fusarium head blight (FHB) disease could help farmers mitigate economic risks and expand market possibilities. Previous work established that a substantial amount of DON in covered barley accumulates in the hull and can be removed by pearling. We studied a diverse panel of 244 naked barley lines to determine if there is a genetic variation for the distribution of DON in Fusarium-infected spikes. We evaluated the panel genotyped with a 50K Barley SNP array for disease se…

Arabinoxylan and pectin features differentiate maize inbred lines with contrasting Fusarium stalk rot resistance.

Maize is susceptible to stalk rot caused by Fusarium graminearum, which reduces the forage quality and yield. This fungus spreads within the host, degrading pith cell walls and colonizing all other plant tissues. The aim of this research was to investigate genotypic differences in cell wall composition of two inbred lines (B73 and EA2024) with different resistance to Fusarium (susceptible and resistant respectively). We also compared the cell wall composition and structure of pith tissues between Fusarium-inoculated and non-inoculated plants. Cell wall analysis of non-inoculated plants showed …
