Patient perspectives after receiving simulated preconception polygenic risk scores (PRS) for family planning.

PURPOSE: The study investigates patient perspectives on the use of Preimplantation Genetic Testing for Polygenic disease (PGT-P) to select embryos with lower risks for common polygenic diseases. Participant responses and attitudes were evaluated after receiving simulated embryo PRS generated from their personal genetic profile. METHODS: Couples seeking OB/GYN or Reproductive Endocrinology and Infertility (REI) care with an interest in genetic risks for common diseases in their prospective children participated. A tool provided PRS scores for 11 conditions, using parental DNA to simulate genet

Co-exposure to multiple air pollutants, genetic susceptibility, and the risk of myocardial infarction onset: a cohort analysis of the UK Biobank participants.

AIMS: The relationship between the long-term joint exposure to ambient air pollution and incidence of myocardial infarction (MI) and modification by genetic susceptibility remain inconclusive. METHODS AND RESULTS: We analysed 329 189 UK Biobank participants without MI at baseline. Exposure concentrations to particulate matter (PM2.5 and PM10), nitrogen dioxide (NO2), and nitrogen oxides (NOx) were obtained. Air pollution score assessing the joint exposure was calculated, and its association with MI was evaluated via Cox model under the P value aggregation framework. Genetic susceptibility to

Socioeconomic inequalities, genetic susceptibility, and risks of depression and anxiety: A large-observational study.

OBJECTIVES: This study aimed to investigate the interplay between genetic susceptibility and socioeconomic disparities on psychiatric disorders. METHODS: In this study, we utilized data from the UK Biobank to analyze the Generalized Anxiety Disorder (GAD)-7 scale (N = 74,425) and the Patient Health Questionnaire (PHQ)-9 (N = 74,101), along with the Index of Multiple Deprivation (IMD). The polygenic risk score (PRS) was calculated to assess the genetic risk associated with GAD-7/PHQ-9 scores, and the individuals were classified into low, medium, and high genetic risk groups according to tertil

Exploring causality in the association between circulating 25-hydroxyvitamin D and colorectal cancer risk: a large Mendelian randomisation study.

BACKGROUND: Whilst observational studies establish that lower plasma 25-hydroxyvitamin D (25-OHD) levels are associated with higher risk of colorectal cancer (CRC), establishing causality has proven challenging. Since vitamin D is modifiable, these observations have substantial clinical and public health implications. Indeed, many health agencies already recommend supplemental vitamin D. Here, we explore causality in a large Mendelian randomisation (MR) study using an improved genetic instrument for circulating 25-OHD. METHODS: We developed a weighted genetic score for circulating 25-OHD usin

Polygenic coronary artery disease association with brain atrophy in the cognitively impaired.

While a number of low-frequency genetic variants of large effect size have been shown to underlie both cardiovascular disease and dementia, recent studies have highlighted the importance of common genetic variants of small effect size, which, in aggregate, are embodied by a polygenic risk score. We investigate the effect of polygenic risk for coronary artery disease on brain atrophy in Alzheimer's disease using whole-brain volume and put our findings in context with the polygenic risk for Alzheimer's disease and presumed small vessel disease as quantified by white-matter hyperintensities. Prostate cancer risk-associated genetic markers and their potential clinical utility.

Prostate cancer (PCa) is one of the most common cancers among men in Western developed countries and its incidence has increased considerably in many other parts of the world, including China. The etiology of PCa is largely unknown but is thought to be multifactorial, where inherited genetics plays an important role. In this article, we first briefly review results from studies of familial aggregation and genetic susceptibility to PCa. We then recap key findings of rare and high-penetrance PCa susceptibility genes from linkage studies in PCa families. We devote a significant portion of this ar

Healthy Lifestyle, Genetic Susceptibility and Risk of Kidney Stone Disease in the UK Biobank: A Prospective Cohort Study.

OBJECTIVE: To explore the association between modifiable lifestyle and kidney stone disease (KSD) risk, and to investigate whether genetic susceptibility modifies this association. PATIENTS AND METHODS: We included 300,875 participants without KSD from the UK Biobank cohort (recruited between March 13, 2006, and October 10, 2010) in this analysis. A healthy lifestyle score was constructed based on seven lifestyle items: regular physical activity, BMI < 25 kg/m2, lower sedentary time, adequate sleep duration, no smoking, adequate fluid intake, and a healthy diet. Based on this score, participa

The impact of breast cancer polygenic risk score disclosure on decisional conflict around risk-reducing mastectomy in women with pathogenic BRCA1/2 variants.

PURPOSE: Women with BRCA1/2 pathogenic variants (PVs) face high breast cancer risks and complex preventive decisions, including whether to undergo risk-reducing mastectomy, which can generate decisional conflict. Polygenic risk scores (PRSs) offer individualized risk estimates that may support informed preventive decisions. We examined whether PRS disclosure reduces decisional conflict about risk-reducing mastectomy in women with BRCA1/2 PVs and whether PRS magnitude interacts with individual characteristics. METHODS: Unaffected women with BRCA1/2 PVs (N = 354) were assessed before PRS disclo

Genetic Variants Associated with Life Expectancy in Patients with Chagas Disease.

Single nucleotide polymorphisms (SNPs), as common genetic variations, can influence biological processes. Identifying these variations is cr … owed for 8 years to assess survival. The association between SNPs and survival was evaluated, and a genetic risk score was generated. Univariate and multivariate Cox regression models assessed the association between SNPs (coded as ordinal variables) and survival time. SNPs with p < 0.05 were selected to construct a risk score, which was then assessed using Kaplan-M …

Stratification by a polygenic risk score of common variation aids in Alzheimer's disease rare variant discovery.

INTRODUCTION: We utilized an Alzheimer's disease (AD) polygenic risk score (PRS) to discover associations with novel rare variants (RVs). METHODS: PRSs for European ancestry (EA) participants of the Alzheimer's Disease Sequencing Project were calculated using summary statistics from a large genome-wide association study. Participants were classified into high (n = 5738) and low (n = 5324) PRS groups based on the median PRS and on the lower and upper 35% of the PRS distribution. RESULTS: Risk variants were disproportionately enriched in the low-PRS group, while protective ones were disproport

Hereditary kidney tumor syndromes: structured evaluation of a questionnaire-based approach.

BACKGROUND: Up to 8% of renal tumors have a monogenic cause, yet hereditary renal cell carcinoma (hRCC) syndromes such as von Hippel-Lindau (VHL), Tuberous Sclerosis Complex (TSC), Birt-Hogg-Dubé (BHD), and Hereditary Leiomyomatosis and Renal Cell Cancer remain underdiagnosed. Early diagnosis is critical for patient management, genetic counseling, and family screening. We developed and prospectively validated a structured risk assessment tool (hRCC score) for identifying patients at risk of hereditary renal tumors. METHODS: A prospective single-center study was conducted at the University Hos

Associations between diet quality, genetic risk of psoriasis, and psoriasis incidence: A prospective cohort study.

BACKGROUND AND AIMS: Psoriasis is a chronic inflammatory skin condition that adversely affects quality of life. Given limited evidence to inform dietary recommendations for psoriasis prevention, this study used data from the UK Biobank to examine associations between diet quality and incident psoriasis, and whether genetic susceptibility moderates this relationship. METHODS: Participants without psoriasis at baseline who completed ≥2 Oxford WebQ 24-h dietary recalls were included. Psoriasis incidence was identified through self-report, primary care, and hospital records. Ten diet quality indi

Genetic Risk Underlying Psychiatric and Cognitive Symptoms in Huntington's Disease.

BACKGROUND: Huntington's disease (HD) is an inherited neurodegenerative disorder caused by an expanded CAG repeat in the HTT gene. It is diagnosed following a standardized examination of motor control and often presents with cognitive decline and psychiatric symptoms. Recent studies have detected genetic loci modifying the age at onset of motor symptoms in HD, but genetic factors influencing cognitive and psychiatric presentations are unknown. METHODS: We tested the hypothesis that psychiatric and cognitive symptoms in HD are influenced by the same common genetic variation as in the general p

Genetic Variant Score and Arrhythmogenic Right Ventricular Cardiomyopathy Phenotype in Plakophilin-2 Mutation Carriers.

INTRODUCTION: Whether detailed genetic information contributes to risk stratification of patients with arrhythmogenic right ventricular cardiomyopathy (ARVC) remains uncertain. Pathogenic genetic variants in some genes seem to carry a higher risk for arrhythmia and earlier disease onset than others, but comparisons between variants in the same gene have not been done. Combined Annotation Dependent Depletion (CADD) score is a bioinformatics tool that measures the pathogenicity of each genetic variant. We hypothesized that a higher CADD score is associated with arrhythmic events and earlier age

Genetic Relationships Between Coronet Score and Type Traits Related to Hoof and Body Size in Holstein Cattle in Japan.

Hoof disorders in dairy cows reduce productivity and increase culling risk, causing economic losses and animal welfare concerns. The coronet score is a 5-point indicator of hoof health (1 = healthy; 5 = severe) that may be useful in breeding programs. However, genetic relationships between coronet score and type traits remain unclear. We estimated genetic correlations between coronet score and type traits related to hoof and body size in Holstein dairy cows. Data from 24,174 first-lactation Holstein cows were analyzed. Coronet scores were obtained from test-day records, whereas type traits wer
