Negative Pressure Wound Treatment of Infections Caused By Extensively Drug-Resistant Gram-Negative Bacteria After Liver Transplantation: Two Case Reports.

Although survival after liver transplantation (LT) has progressively improved over the last years, an increased prevalence of clinically relevant infections in LT patients is well documented. In particular, the spread of infections sustained by extensively drug-resistant bacteria (XDR) produced an increase in the incidence of wound infections. Implementation of treatments for these life-threatening events is mandatory. This study describes 2 LT patients in whom XDR wound infection was effectively treated using negative pressure wound treatment (NPWT) combined with targeted local and systemic a

Disinfection mechanisms of gram-negative and gram-positive bacteria through multi-target damage under UV-C light at 222 and 254 nm.

Ultraviolet-C (UV-C) irradiation is a promising approach for inactivating viruses and microorganisms. Germicidal mercury lamps emitting at 254 nm and krypton chloride (KrCl) excimer lamps emitting at 222 nm both possess sterilization properties, but the KrCl lamp is considered less harmful to humans. UV-C at 222 and 254 nm induces cyclobutene pyrimidine dimers (CPDs) in Escherichia coli (E. coli). Although E. coli irradiated at 254 nm can undergo photoreactivation, cells exposed to 222 nm cannot, as CPDs are not effectively repaired. This study aimed to investigate the molecular mechanisms und

Managing Pneumonia Due to Rare Non-Fermenting Gram-Negative Bacteria: Epidemiology, Risk Factors and Therapeutic Strategies.

Pneumonia remains a leading cause of morbidity and mortality worldwide, with bacterial pathogens contributing significantly to its burden. While Pseudomonas aeruginosa and Acinetobacter baumannii complex are well-recognized non-fermenting Gram-negative bacteria (NFGNB) causing severe pneumonia, particularly in healthcare settings, an expanding array of other, rarer NFGNB species is increasingly implicated. These species include, but are not limited to, Achromobacter spp., Ochrobactrum spp., Burkholderia spp., Aeromonas spp., Roseomonas spp., Elizabethkingia spp., Chryseobacterium spp. Alcalige…

Discovery of eucalyptus essential oil as a novel colistin adjuvant eradicate multidrug-resistant Gram-negative bacteria.

Multidrug-resistant (MDR) Gram-negative bacteria poses a significant public health risk due to its multidrug resistance mechanism. The discovery of novel antibiotic adjuvant to combat clinical superbugs is an urgent priority for addressing the antibiotic resistance crisis. Here, through checkerboard assays, time-kill analyses, disk diffusion tests, biofilm inhibition assays, and microscopic examinations, eucalyptus essential oil (EEO) was identified as a potent enhancer of colistin activity. Mechanistic analyses further demonstrated that EEO downregulated bacterial genes associated with iron h…

Newly developed antibiotics against multidrug-resistant and carbapenem-resistant Gram-negative bacteria: action and resistance mechanisms.

Antimicrobial resistance stands as one of the most urgent global health concerns in the twenty-first century, with projections suggesting that deaths related to drug-resistant infections could escalate to 10 million by 2050 if proactive measures are not implemented. In intensive care settings, managing infections caused by multidrug-resistant (MDR) Gram-negative bacteria is particularly challenging, posing a significant threat to public health and contributing substantially to both morbidity and mortality. There are numerous studies on the antibiotics responsible for resistance in Gram-negativ…

Restoring colistin sensitivity in colistin-resistant gram-negative bacteria: combinatorial use of α-terpineol and colistin.

UNLABELLED: Multidrug-resistant gram-negative bacteria (GNB) have posed a serious public health hazard worldwide. Colistin (COL), the ultimate therapeutic option for treating GNB infections, has had escalating resistance rates concomitant with increased utilization. In this study, we attempted to increase the sensitivity to COL using a plant extract, α-terpineol (α-TP). Checkerboard and time-kill assays indicated that the combination of COL and α-TP exhibited pronounced synergistic antimicrobial effects against colistin-resistant (COL-R) GNB in vitro. Biofilm assessments revealed that COL comb…

Novel aminothiazoximone-corbelled ethoxycarbonylpyrimidones with antibiofilm activity to conquer Gram-negative bacteria through potential multitargeting effects.

The emergence of drug-resistant microorganisms threatens human health, and it is usually exacerbated by the formation of biofilm, which forces the development of new antibacterial agents with antibiofilm activity. In this work, a novel category of aminothiazoximone-corbelled ethoxycarbonylpyrimidones (ACEs) was designed and synthesized, and some of the prepared ACEs showed potent bioactivity against the tested bacteria. In particular, imidazolyl ACE 6c showed better inhibitory activity towards Acinetobacter baumannii and Escherichia coli with MIC values both of 0.0066 mmol/L than norfloxacin.
