Psychedelic substances have great promise for the treatment of many conditions, and they are the subject of intensive research. As with other medical treatments, both research and clinical use of psychedelics depend on our ability to ensure informed consent by patients and research participants. However, some have argued that informed consent for psychedelic use may be impossible, because psychedelic experiences can be transformative in the sense articulated by L. A. Paul (2014). For Paul, transformative experiences involve either the acquisition of knowledge that cannot be obtained in any oth

A number of organizations have developed or are developing psychedelic integration groups, held in person or online. In parallel, there have been calls to make enhanced integration available in the community after clinical trials. Here, we explore a potential reason why individuals may seek out these groups: namely, the feelings of disconnection and loneliness that may arise after the psychedelic experience (noting that there may be other reasons that individuals may seek out these groups). Integration groups can offer significant benefits to participants (e.g. a sense of community, shared cop

INTRODUCTION: After a long period of obscurantism, a possible role of psychedelics in clinical practice has progressively become a tangible perspective during the last two decades. However, the resounding enthusiasm linked to such 'psychedelic renaissance' runs the risk to unduly minimize the possible hazards associated with these compounds, while expanding their alleged benefits to improbable panacea-like proportions. In order to avoid mystifying or demonizing the properties of 5-HT2a agonists on emotional grounds, this subject requires a strictly unprejudiced and cautious approach to the evi

Several naturally occurring molecules exhibit unique potential in treating certain elements of psychiatric illnesses and are being actively pursued in therapeutic development. Among these are molecules termed entheogens, a preferred name for plant-derived compounds that alter human consciousness for religious or spiritual purposes, which are especially important to various Indigenous groups, and their use within these cultures precedes that of the contemporary medicalized use. Here, we acknowledge that some entheogens were included in the DARK Classics issues of ACS Chemical Neuroscience and t

In recent years, there has been a growing movement to apply psychedelic drugs for treating psychiatric disorders, including depression. Although the therapeutic effects of psychedelic drugs were verified in the 1960s, their abuse by the public led to their designation as narcotics in 1970, and clinical research on them was suspended for a time. However, since 1994, clinical trials have been conducted in large numbers and elicited great excitement. In this review, we consider the history of psychedelic drugs with potent therapeutic effects and outline the prospects for this field.

A recent report by Chen et al. describes the discovery of RmNMT, a highly active and promiscuous tryptamine N-methyltransferase from the cane toad, Rhinella marina. N,N-dimethyltryptamine derivatives produced by this enzyme were then evaluated for their potential to serve as next-generation treatments for mental health disorders.

As individuals and communities around the world confront mounting physical, psychological, and social threats, three complimentary mind-body-spirit pathways toward health, wellbeing, and human flourishing remain underappreciated within conventional practice among the biomedical, public health, and policy communities. This paper reviews literature on psychedelic science, contemplative practices, and Indigenous and other traditional knowledge systems to make the case that combining them in integrative models of care delivered through community-based approaches backed by strong and accountable he

The extremes of human experiences, such as those occasioned by classic psychedelics and psychosis, provide a rich contrast for understanding how components of these experiences impact well-being. In recent years, research has suggested that classic psychedelics display the potential to promote positive enduring psychologic and behavioral changes in clinical and nonclinical populations. Paradoxically, classic psychedelics have been described as psychotomimetics. This review offers a putative solution to this paradox by providing a theory of how classic psychedelics often facilitate persistent i

This work reports the synthesis, characterization, and sensing behavior of a hybrid nanodevice for the detection of the potent abuse drug 25I-NBOMe. The system is based on mesoporous silica nanoparticles, loaded with a fluorescent dye, functionalized with a serotonin derivative and capped with the 5-HT2A receptor antibody. In the presence of 25I-NBOMe the capping antibody is displaced, leading to pore opening and rhodamine B release. This delivery was ascribed to 5-HT2A receptor antibody detachment from the surface due to its stronger coordination with 25I-NBOMe present in the solution. The pr

Psychedelics are compounds that alter consciousness by acting on serotonin receptors in the brain. The term 'psychedelic', from the Greek for mind manifesting, refers to the drugs' subjective effects and was first proposed by Humphry Osmond in 1956. Other terms have been used to emphasize different aspects of the psychological experiences produced by various related compounds, including hallucinogens (perceptual), entheogens (spiritual), and empathogens or entactogens (social/emotional). The diversity in terminology reflects the existence of hundreds of potential psychedelic compounds with a s…

We assessed the incidence of mechanical injury in drug users. Incidence was 21.9% and exceeds the global average injury rate by 2.4 times Mortality from mechanical injury among drug users ranges from 4.46% to 5%. This value is 4.46-5%, which is 64-71 times higher than the average mortality rates from injury and 89-100 times higher than trauma mortality rate in Russia. Psychoactive drugs from the groups of neurostimulators and neurodepressors have a significant impact on the course of traumatic brain injury; hallucinogens only increase the risk of traumatic injury. In cases of mild traumatic br…

This study examines panic attacks and substance use in a sample of incoming college freshman (n = 399 ) using questionnaires. Panickers (n = 47 ) were significantly more likely than nonpanickers (n = 290) to report having ever used sedatives, stimulants, opiods, and other drugs, but not tobacco, alcohol, cocaine, or hallucinogens. Gender and race did not substantially moderate the associations between substance use and panic attacks. Sedative, stimulant, opiod, and other drug use was not associated with panic attack frequency or the occurrence of unexpected attacks. The relationships of anxiet…

A hierarchical cluster analysis was performed on 273 self-admitted high-school drug users' descriptions of how frequently they had employed marijuana, stimulants, depressants, hallucinogens, solvents, and opiates during the preceding year. Four types of polydrug users were identified and considered as representing an ordinal scaling of the students along a continuum of overall involvement with drugs, and each user was assigned an index score based on his typal membership. A stepwise multiple regression was then performed to discover which of the students' background characteristics and attitud…

As psychedelic interventions move from controlled clinical trials into broader implementation, critical dimensions of lived experience among patients, trial participants, families, and caregivers remain systematically undermeasured in research, service design, and oversight. The resulting gap between protocol-defined outcomes and real-world experiences over time creates blind spots in current approaches, particularly regarding safety, care quality, post-treatment stabilization, aftercare, and long-term vulnerability. I argue that lived experience is a form of systems-relevant evidence essentia

Psychedelics are a group of substances within the heterogeneous class of hallucinogenic drugs. Via binding to the serotonin (5-HT) 2A receptor, psychedelics exert profound alterations in various mental domains, including sensation, cognition, emotions, and self-perception. Psychedelics comprise phenethylamines (e.g., mescaline), tryptamines (e.g., psilocybin), and ergolines (e.g., LSD). These drugs have been used recreationally for decades but have also regained attention as potential treatments for various psychiatric as well as neurological illnesses. While psychedelics are generally conside
