Dexamethasone loaded nanoparticles exert protective effects against Cisplatin-induced hearing loss by systemic administration.

Ototoxicity is one of the most important adverse effects of cisplatin chemotherapy. As a common treatment of acute sensorineural hearing loss, systemic administration of steroids was demonstrated ineffective against cisplatin-induced hearing loss (CIHL) in published studies. The current study aimed to evaluate the potential protective effect of dexamethasone (DEX) encapsulated in polyethyleneglycol-coated polylactic acid (PEG-PLA) nanoparticles (DEX-NPs) against cisplatin-induced hearing loss following systemic administration. DEX was fabricated into PEG-PLA nanoparticles using emulsion and ev

Hearing Loss in Young and Middle-Aged Adults as a Modifiable Risk Factor for Late-Life Dementia: A Systematic Review and Meta-Analysis.

BACKGROUND: Individuals with untreated hearing loss often experience cognitive decline as a result of increased cognitive load and reduced sensory stimulation. Despite the well-established link between untreated hearing loss and cognitive decline in older adults, its impact on cognition in young and middle-aged adults has not been systematically examined. Given the Lancet Commission's identification of midlife hearing loss as the leading modifiable risk factor for dementia, early identification of cognitive decline is essential. This review explored the cognitive impact of untreated hearing lo

PKHD1L1, a gene involved in the stereocilia coat, causes autosomal recessive nonsyndromic hearing loss.

Identification of genes associated with nonsyndromic hearing loss is a crucial endeavor given the substantial number of individuals who remain without a diagnosis after even the most advanced genetic testing. PKHD1L1 was established as necessary for the formation of the cochlear hair-cell stereociliary coat and causes hearing loss in mice and zebrafish when mutated. We sought to determine if biallelic variants in PKHD1L1 also cause hearing loss in humans. Exome sequencing was performed on DNA of four families segregating autosomal recessive nonsyndromic sensorineural hearing loss. Compound het

Audiological and Subjective Benefits in a Child with Microtia and Atresia After Sequential Bilateral Implantation with Active Bone Conduction Devices: A Case Study.

BACKGROUND: With bilateral hearing loss, the main problems for the patient are speech understanding in noise and, especially in asymmetrical hearing loss, an inability to correctly localize sound sources. There are multiple methods of treatment and rehabilitation for people with conductive hearing loss, and one of them is to use an active bone conduction implant. This case study is designed to evaluate the auditory benefits and sound localization accuracy with active bilateral bone conduction implants-in comparison to unilateral ones-in a patient with congenital bilateral conductive hearing lo

Auditory brainstem response artefact caused by caloric testing.

In the external ear canal, residual water from caloric testing or any other irrigation may act to simulate a conductive hearing loss and interfere with subsequent auditory brainstem response recording leading to increased latencies and reduced amplitudes. This artefact in the ABR may be mistaken for a retrocochlear lesion.

Working Memory, Sleep, and Hearing Problems in Patients with Tinnitus and Hearing Loss Fitted with Hearing Aids.

BACKGROUND: Tinnitus is a common condition and there is a need to evaluate effects of tinnitus management in relation to moderating factors such as degree of hearing loss. As it is possible that tinnitus influences concentration, and thus is likely to disturb cognitive processing, the role of cognitive functioning also needs to be investigated. PURPOSE: To compare a group of patients with sensorineural hearing loss and tinnitus to a control group with only sensorineural hearing loss (and no tinnitus). To investigate working memory, sleep, and hearing problems measured before and after hearing

Age-related hearing loss accelerates the decline in fast speech comprehension and the decompensation of cortical network connections.

Patients with age-related hearing loss face hearing difficulties in daily life. The causes of age-related hearing loss are complex and include changes in peripheral hearing, central processing, and cognitive-related abilities. Furthermore, the factors by which aging relates to hearing loss via changes in auditory processing ability are still unclear. In this cross-sectional study, we evaluated 27 older adults (over 60 years old) with age-related hearing loss, 21 older adults (over 60 years old) with normal hearing, and 30 younger subjects (18-30 years old) with normal hearing. We used the outc

Functional hearing difficulty in Alzheimer's disease: Selective links to verbal fluency and behavioral and psychological symptoms of dementia severity.

BackgroundHearing loss is a well-established risk factor for dementia. However, the clinical relevance of everyday listening difficulties after the diagnosis of Alzheimer's disease (AD) remains insufficiently understood, particularly in relation to cognition and behavioral and psychological symptoms of dementia (BPSD). Functional hearing difficulty refers to perceived difficulties in everyday listening situations that are not fully captured by standard audiometric thresholds.ObjectiveTo examine the associations between caregiver-rated functional hearing difficulty, cognitive measures commonly

Early Physiological and Cellular Indicators of Cisplatin-Induced Ototoxicity.

Cisplatin chemotherapy often causes permanent hearing loss, which leads to a multifaceted decrease in quality of life. Identification of early cisplatin-induced cochlear damage would greatly improve clinical diagnosis and provide potential drug targets to prevent cisplatin's ototoxicity. With improved functional and immunocytochemical assays, a recent seminal discovery revealed that synaptic loss between inner hair cells and spiral ganglion neurons is a major form of early cochlear damage induced by noise exposure or aging. This breakthrough discovery prompted the current study to determine ea

The neonate has a temporary conductive hearing loss due to fluid in the middle ear.

Postnatal functional changes in the activity of the ear and auditory pathway in neonatal guinea pigs [from day of birth (postnatal day, PND = 0), PNDs 1-4, 7 and then weekly up to 7 weeks] were studied as a model of maturation of hearing in human neonates. On the day of birth there were signs of a conductive hearing loss: negative middle ear pressure, auditory nerve brainstem evoked response (ABR) threshold elevation, ABR wave 1 latency prolongation and low amplitude otoacoustic emissions. The conductive hearing loss is probably a result of the (amniotic) fluid found in the neonatal middle-ear

In vivo optical mapping of the tympanic membrane impulse response.

The wide frequency range of the human hearing could be narrowed by various pathologies in the middle ear and in the tympanic membrane that lead to conductive hearing loss. Diagnosing such hearing problems is challenging, however, often relying on subjective hearing tests supported by functional tympanometry. Here we present a method for in vivo 2D mapping of the impulse response of the tympanic membrane, and demonstrate its potential on a healthy human volunteer. The imaging technique is based on interferometric spectrally encoded endoscopy, with a handheld probe designed to scan the human tym

Different Audiologic Outcomes in Twins with Congenital Cytomegalovirus Infection.

Congenital cytomegalovirus (cCMV) infection is a major cause of hearing loss in children. A few cases of cCMV twin pregnancies are reported in the literature. Twins can react differently to maternal infection, but hearing loss is rarely evaluated. Two couples of twins with cCMV infection and different audiologic outcomes are reported. The first couple of twins was composed by two male twins, both affected by cCMV infection. The first born had normal hearing function, and the second born had sensorineural hearing loss (SNHL). In the second couple, a male and a female twin, only the male twin wa

[Successful decompression of the acoustic nerve after postarachnitic hearing damage (author's transl)].

Arachnitic adhesions of the pontine angle can damage the VIIIth cranial nerve to complete hearing loss. If there exists only a circumscript damage of the myeline sheet, this functional loss is reversible after decompression of the nerve. Preoperatively, the electrocochleography and the cisternoscopy discover this type of nerve damage and so, the probability of the postoperative hearing recovery is quite predictable.

[Evoked otoacoustic emissions in diagnosis of cochlear hearing disorders].

The authors consider the possibility of routine use of evoked otoacoustic emissions (EOE) in diagnosis of sensory hearing loss by comparison of EOE curves with relative curves obtained by pure tone audiometry. 180 ears with sensory hearing loss of different aetiologies were examined. EOE were obtained by application of Bray and Kemp's Evoked Otoacustic Measurement Package (ILO88). The results obtained show a close correlation between presence of EOE and real hearing losses of different frequencies obtained by pure tone audiometric evaluation. We could detect no EOE with hearing losses over 50

[Presbycusis - Age Related Hearing Loss].

Presbycusis or age related hearing loss can be defined as a progressive, bilateral and symmetrical sensorineural hearing loss due to age related degeneration of inner ear structures. It can be considered a multifactorial complex disorder with environmental and genetic factors. The molecular, electrophysiological and histological damage at different levels of the inner ear cause a progressive hearing loss, which usually affects the high frequencies of hearing. The resulting poor speech recognition has a negative impact on cognitive, emotional and social function in older adults. Recent investig

Auditory brainstem responses in infants recovering from bacterial meningitis. Audiologic evaluation.

Auditory brainstem response (ABR) was used to assess possible hearing loss in 60 patients recovering from bacterial meningitis. The ABR results were consistent with either unilateral or bilateral hearing loss in 35% of the cases tested. Of these, 15% were conductive-type hearing loss. Twelve percent had sensorineural hearing losses and normal brainstem function. The remaining 8% had elevated ABR thresholds coincident with findings suggestive of neuropathology of the auditory brainstem pathways. A case of reversible sensorineural hearing loss was documented. Various clinical and demographic fac

Hearing/vestibular problems, racial differences, and associations with physical function impairment among breast cancer survivors.

BACKGROUND: Breast cancer survivors face many health challenges, including tinnitus, hearing loss, and vertigo which will grow with an aging population and improved treatment outcomes. However, the prevalence of hearing/vestibular problems, racial differences, and relationships to physical function are poorly characterized in breast cancer survivors. METHODS: Between July and September 2023, we surveyed the Chicago Multiethnic Epidemiologic Breast Cancer Cohort. Tinnitus, hearing loss, and vertigo were self-reported and verified through electronic health records. Physical function impairment
