Prolongation of clotting times produced by hematin was investigated both in vitro and in vivo. Hematin-derived anticoagulant (HDA) was found to be due to a degradative product or derivative of hematin, and was generated in vitro in standing (aging) aqueous solutions of the parent compound. Generation of HDA in vitro was inhibited by antioxidants. The anticoagulant effect of HDA was inhibited by freshly prepared hematin, fresh Sn-protoporphyrin, imidazole, or the iron chelator desferrioxamine. Various metals have been shown to inhibit porcine brain lipoamidase activity at 0.1 mM, but not ferrous and ferric ions. Several mutants of Escherichia coli K-12 defective in aerobic metabolism were isolated. One such mutant was found to be deficient in cytochromes, heme, and catalase. Seventeen strains of haemophilus equigenitalis isolated from the cervix, clitoris, and urethra of mares were biochemically characterized with the API 10E and APIZYM test kit systems, conventional biochemical tests, and the porphyrin test. Chronic inflammation in such diseases as rheumatoid arthritis has been associated with the accumulation of iron in mononuclear phagocytes.
