Model-specific effects of bumetanide on epileptiform activity in the in-vitro intact hippocampus of the newborn mouse.

The immature brain has a higher susceptibility to develop seizures, which often respond poorly to classical pharmacological treatment. It has been recently suggested that bumetanide, which blocks Na(+)-dependent K(+)-Cl(-)-cotransporter isoform 1 (NKCC1) and thus attenuates depolarizing GABAergic responses, could soothe epileptiform activity in immature nervous systems. To evaluate whether bumetanide consistently attenuates epileptiform activity, we investigated the effect of 10 microM bumetanide in five different in-vitro epilepsy models using field potential recordings in the CA3 region of i

Antimineralocorticoid canrenoate enhances secretory activity of the hypothalamus-pituitary-adrenocortical (HPA) axis in humans.

In rats, both hippocampal glucocorticoid and mineralocorticoid receptors (MR) have been shown to participate in the regulation of basal hypothalamus-pituitary-adrenocortical (HPA) secretory activity. Inhibition of hippocampal MRs enhanced the activity of the HPA axis in these animals. We tested the influence of potassium cancrenoate, a selective MR antagonist, on basal cortisol secretion in 10 healthy young men during sleep. Cortisol, ACTH, vasopressin and growth hormone (GH) were determined at 22.00, 23.00, 01.00, 04.00 and 07.00 h. Sleep was monitored by somnopolygraphy. Potassium canrenoate

Ameliorating effect of veratric acid against mercury-induced neurologic deficit and reproductive toxicity in female rats.

The aim of this investigation was to explore the potential effects of veratric acid on central nervous and reproductive toxicity caused by mercuric chloride in female rats. Six mercury-exposed female rats in each group were separately treated with 12.5, 25, and 50 mg/kg of veratric acid. The brain and ovaries were then evaluated for biochemical and histopathological analysis. Based on the results obtained through this study's assessment methods, mercuric chloride (HgCl2) administration may harm the central nervous and reproductive systems in females. Evidence suggested that test subjects exper

Heart rate variability (HRV) in kidney failure: measurement and consequences of reduced HRV.

A common cause of death in end-stage renal disease (ESRD) patients on dialysis is sudden cardiac death (SCD). Compared to the general population, the percentage of cardiovascular deaths that are attributed to SCD is higher in patients treated by dialysis. While coronary artery disease (CAD) is the predominant cause of SCD in dialysis patients, reduced heart rate variability (HRV) may play a role in the higher risk of SCD among other risk factors. HRV refers to beat-to-beat alterations in heart rate as measured by periodic variation in the R-R interval. HRV provides a non-invasive method for in

Evidence of a selective increase in cardiac sympathetic activity in patients with sustained ventricular arrhythmias.

BACKGROUND: Although enhanced efferent cardiac sympathetic nervous activity has been proposed as an important factor in the genesis of ventricular arrhythmias and sudden cardiac death, direct clinical evidence has been lacking. METHODS: We measured the rates of total and cardiac norepinephrine spillover into the plasma, which reflect respectively overall and cardiac sympathetic nervous activity, in 12 patients who had recovered from a spontaneous, sustained episode of ventricular tachycardia or ventricular fibrillation outside the hospital 4 to 48 days earlier. The results were compared with

Association of MMP-2 and MMP-9 with clinical outcome of neurocysticercosis.

Matrix metalloproteinases (MMPs) are the major endopeptidases involved in proteolysis of blood brain barrier (BBB) during central nervous system (CNS) infections. The present study detected serum levels and activities of MMP-2 and MMP-9 in patients with neurocysticercosis (NCC) and their association with symptomatic disease. In total, 68 individuals with NCC (36 symptomatic patients with active seizures and 32 asymptomatic individuals) and 37 healthy controls were enrolled for the study. Serum MMP-2 and MMP-9 levels and their activities were measured by ELISA and gel zymography respectively. M

[Meningitis associated with tsutsugamushi disease--detection of intrathecal interferon-gamma synthesis and lymphocyte subsets in blood and CSF].

We report a case of meningitis associated with tsutsugamushi disease. The lymphocyte subpopulation study of peripheral blood revealed a reversed CD4/CD8 ratio (0.14), an increased number of activated T cells and an elevated cytotoxic T cell activity. In cerebrospinal fluid (CSF), the CD4/CD8 ratio was much higher (0.84) and activated T cells were less prevalent than in blood. These pictures of lymphocyte subsets were characteristic and different from that of viral meningitis. We also verified an elevation of interferon-gamma levels in CSF as well as in serum and intrathecal interferon-gamma sy

CD73 and PD-L1 expression in WHO grade 1 and grade 2 meningiomas: an exploratory analysis of purinergic and immune-related profiles.

BACKGROUND: Meningiomas are the most common primary central nervous system tumors and exhibit heterogeneous biological behavior across WHO grades. Emerging evidence suggests that purinergic signaling and immune-related pathways may contribute to meningioma biology. OBJECTIVE: This study evaluated the expression and activity of CD39/ENTPD1 and CD73/NT5E, the expression of PD-L1/CD274, and the systemic inflammatory profile in patients undergoing surgical resection of meningiomas. RESULTS: Forty patients were included, with specific subgroups available for molecular, immunohistochemical, enzyma

Elevated resting and exercise-induced cortisol levels after mineralocorticoid receptor blockade with canrenoate in healthy humans.

Activation of central nervous mineralocorticoid receptors (MRs) has been shown to inhibit the activity of the hypothalamo-pituitary-adrenocortical (HPA) axis in animals. Here, we examined whether MRs in humans likewise regulate HPA activity in response to a physiological stressor. In a balanced, randomized, double-blind, cross-over trial, 12 healthy men were treated with either two injections of 200 mg canrenoate or placebo 24 and 8 h before an intense physical exercise taking place between 1600 and 1700 h. Exercising was preceded by a 60-min rest period and followed by another 90-min rest. Bl

Renal denervation attenuates cardiac fibrosis and improves left ventricular function in rats with myocardial infarction.

The sympathetic nervous system (SNS) plays a critical role in regulating inflammatory responses after myocardial infarction (MI). We aimed to investigate whether modulation of the SNS through renal denervation (RDN) influences myocardial remodeling following MI. MI in rats was induced 2 days after bilateral RDN or Sham surgery. Cardiac MRI showed that left ventricular ejection fraction was significantly higher in RDN-treated MI rats compared to Sham-treated MI rats. RNA sequencing of left ventricular tissue revealed that RDN suppresses fibrosis and inflammation related pathways at the whole tr

Developmental roles of the spontaneous depolarization wave in synaptic network formation in the embryonic brainstem.

One of the earliest activities expressed within the developing central nervous system is a widely propagating wave-like activity, which we referred to as the depolarization wave. Despite considerable consensus concerning the global features of the activity, its physiological role is yet to be clarified. The depolarization wave is expressed during a specific period of functional synaptogenesis, and this developmental profile has led to the hypothesis that the wave plays some roles in synaptic network organization. In the present study, we tested this hypothesis by inhibiting the depolarization

Metabolomic profiling of CSF in multiple sclerosis and neuromyelitis optica spectrum disorder by nuclear magnetic resonance.

Multiple sclerosis (MS) and neuromyelitis optica spectrum disorder (NMOSD) are inflammatory diseases of the central nervous system. Although several studies have characterized the metabolome in the cerebrospinal fluid (CSF) from MS and NMOSD patients, comparative analyses between them and between the relapse and the remission of each disease have not been performed. Both univariate and multivariate analyses were used to compare 1H-NMR spectra of CSF from MS, NMOSD, and healthy controls (HCs). The statistical analysis showed alterations of eight metabolites that were dependent on the disease. L
