Radix Scrophulariae regulates proliferation, apoptosis, and autophagy of rat thyroid cells via the MST1/Hippo signaling pathway.

This study investigated the therapeutic effects of Radix Scrophulariae (RS) extract on hyperthyroidism and the associated mechanisms. A hyperthyroid cell model was established using thyrotropin receptor antibody and levothyroxine sodium tablets. FRTL-5 rat thyroid cells were treated with varying concentrations of RS-containing serum. Protein expression levels of Bcl-2, Caspase-3, PCNA, Cyclin D1, MST1, LC3-II/I, and ATG5 were assessed by Western blotting to determine the optimal concentration. Subsequent experiments included RS treatment with or without MST1 overexpression or the Hippo pathway

Qianlong Shutong Formula alleviates benign prostatic hyperplasia associated with the regulation of inflammation, fibrosis and the Hippo-YAP/TAZ pathway.

BACKGROUND: Clinically employed for benign prostatic hyperplasia management, the Qianlong Shutong Formula (QLSTF) represents an empirical therapeutic preparation, exhibiting properties of heat clearance, diuretic promotion, blood circulation activation, and nodule resolution. However, its molecular mechanisms in regulating inflammation and fibrosis remain unclear. OBJECTIVE: This study aimed to untangle whether QLSTF intervenes in the inflammatory and fibrotic processes of BPH by mediating the Th17/Treg immune microenvironment and the Hippo-YAP/TAZ pathway. MATERIALS AND METHODS: A rat BPH m

Trimethylamine-N-oxide disrupts spermatogenesis by inducing mitochondrial oxidative stress injury through Hippo signaling.

BACKGROUND: The gut-testis axis is increasingly recognized as a regulator of male reproductive health; however, the key microbial contributors, metabolites, and underlying mechanisms remain unclear. METHODS: We performed fecal metagenomic sequencing in 107 participants to identify microbial taxa associated with abnormal semen parameters. Serum trimethylamine-N-oxide (TMAO) levels were measured and correlated with semen quality. In mouse models, including fecal microbiota transplantation, dietary choline supplementation, mono-colonization, and direct TMAO administration, we assessed sperm morp

Hippo signaling and epithelial cell plasticity in mammalian liver development, homeostasis, injury and disease.

A traditional view of cellular differentiation is unidirectional: progenitor cells adopt specific fates in response to environmental cues resulting in deployment of cell-specific gene expression programs and acquisition of unique differentiated cellular properties such as production of structural and functional proteins that define individual cell types. In both development and in tissue repair stem and progenitor cells are thought to both self-renew to maintain the pool of precursors and to expand to give rise to transient amplifying and differentiated cell types. Recently, however, it has be

The MST4-14-3-3ζ complex promotes pancreatic cancer by activating YAP.

BACKGROUND: The mammalian STE20-like kinase 4 (MST4) has been implicated as a potential oncogene in various cancers, including pancreatic cancer (PC). However, the precise mechanisms through which MST4 promotes PC malignancy remain incompletely understood. METHODS: A Biotin identification (BioID)-based proximity labeling approach was employed to identify key MST4-interacting molecules in human PANC-1 pancreatic cancer (PC) cells, followed by cell migration assay to validate the synergistic promotional effect of the MST4-interacting complex on PC cell migration. Direct binding between MST4 and

From fitness to fate: Hippo-YAP-mediated cell competition influence on stem cells activity.

Cell competition is a fundamental quality-control mechanism where fitter 'winner' cells eliminate less-fit 'loser' counterparts, thereby fine-tuning cell populations during development and maintaining adult tissue integrity. This highly conserved, natural cellular process is absolutely crucial for organogenesis, but once dysfunctional, can instead be exploited by 'super-fit' cancer cells to promote tumourigenesis. This review aims to provide an overview of how heterogeneity is the root cause of cell competition, the factors which influence its emergence, the various modes of cell competition,

High fluid shear stress induces Hippo/YAP pathway in articular cartilage superficial layer cells: A potential mechanistic link to osteoarthritis.

Abnormal mechanical loading, which can lead to articular cartilage damage, is a significant contributor to the onset of osteoarthritis (OA). Articular cartilage superficial layer cells are among the first cells to respond to changes in the mechanical environment and are highly sensitive to mechanical stimuli. This study aimed to investigate the effects of high fluid shear stress on the articular cartilage superficial layer cells and the underlying mechanisms. We found that high fluid shear stress of 20 dyne/cm2 induces inflammation and promotes catabolic processes in these cells. Short-term hi

A novel 2q37 microdeletion containing human neural progenitors genes including STK25 results in severe developmental delay, epilepsy, and microcephaly.

2q37 microdeletion syndrome is a rare syndrome characterized by neurodevelopmental delay, bone, cardiovascular, and neurological alterations. This syndrome is typically associated with loss of genetic material of approximately 100 genes in the 2q37 band. However, the genes associated with neurodevelopmental phenotype in this syndrome are still unknown. We identified a deleted region of 496 kb by whole genome array CGH in a patient who fulfilled criteria for 2q37 microdeletion syndrome with developmental delay, microcephaly, hypoplasia of the corpus callosum, hand wringing, toe walking, and sei

Investigating the Transition of Pre-Symptomatic to Symptomatic Huntington's Disease Status Based on Omics Data.

Huntington's disease is a rare neurodegenerative disease caused by a cytosine-adenine-guanine (CAG) trinucleotide expansion in the Huntingtin (HTT) gene. Although Huntington's disease (HD) is well studied, the pathophysiological mechanisms, genes and metabolites involved in HD remain poorly understood. Systems bioinformatics can reveal synergistic relationships among different omics levels and enables the integration of biological data. It allows for the overall understanding of biological mechanisms, pathways, genes and metabolites involved in HD. The purpose of this study was to identify the

Hcfc1 and Ogt Mediate Zebrafish CNS Regeneration Through Hippo/Yap Signalling.

Regeneration of the central nervous system (CNS) is a complex and tightly regulated process, yet the precise molecular players and transcriptional regulators involved remain incompletely understood. Here, we identify Host Cell Factor-1 (Hcfc1), a transcriptional co-regulator, and O-GlcNAc transferase (Ogt), which cleaves and O-GlcNAcylates HCF-1, as crucial regulators of zebrafish brain and retinal regeneration. We uncover their interplay with the Hippo/Yap signalling pathway, a well-known regulator of tissue growth and repair. Knockdown of hcfc1a/b or Ogt activity inhibition disrupts regenera

Electroacupuncture regulates microglia polarization via lncRNA-mediated hippo pathway after ischemic stroke.

Microglia polarization and microglia-mediated inflammation play a crucial role in the development of ischaemic brain injury. Electroacupuncture (EA) has the function of anti-inflammatory, which has been thoroughly validated and utilized to treat ischemic brain damage. The fundamental mechanism by which EA alleviates ischemic brain damage by decreasing microglia polarization and microglia-mediated inflammation, however, remains unknown. In the current study, the activation of microglia and inflammatory cytokines was analyzed to confirm the anti-inflammatory function of EA in middle cerebral art

GNAS knockdown suppresses osteogenic differentiation of mesenchymal stem cells via activation of Hippo signaling pathway.

Bone marrow-derived mesenchymal stem cells (BMSCs) are a suitable option for cell-based tissue engineering therapies due to their ability to renew and differentiate into multiple different tissue types, such as bone. Over the last decade, the effect of GNAS on the regulation of osteoblast differentiation has attracted great attention. Herein, this study aimed to explore the role of GNAS in osteogenic differentiation of MSCs. A total of 85 GNASf/f male mice were selected for animal experiments and 10 GNASf/f male mice for BMSC isolation to conduct cell experiments. The mice and BMSCs were treat

Mst1-mediated phosphorylation of FoxO1 and C/EBP-β stimulates cell-protective mechanisms in cardiomyocytes.

The molecular mechanisms by which FoxO transcription factors mediate diametrically opposite cellular responses, namely death and survival, remain unknown. Here we show that Mst1 phosphorylates FoxO1 Ser209/Ser215/Ser218/Thr228/Ser232/Ser243, thereby inhibiting FoxO1-mediated transcription of proapoptotic genes. On the other hand, Mst1 increases FoxO1-C/EBP-β interaction and activates C/EBP-β by phosphorylating it at Thr299, thereby promoting transcription of prosurvival genes. Myocardial ischemia/reperfusion injury is larger in cardiac-specific FoxO1 knockout mice than in control mice. However

A Novel STK4 Mutation Impairs T Cell Immunity Through Dysregulation of Cytokine-Induced Adhesion and Chemotaxis Genes.

PURPOSE: Human serine/threonine kinase 4 (STK4) deficiency is a rare, autosomal recessive genetic disorder leading to combined immunodeficiency; however, the extent to which immune signaling and host defense are impaired is unclear. We assessed the functional consequences of a novel, homozygous nonsense STK4 mutation (NM_006282.2:c.871C > T, p.Arg291*) identified in a pediatric patient by comparing his innate and adaptive cell-mediated and humoral immune responses with those of three heterozygous relatives and unrelated controls. METHODS: The genetic etiology was verified by whole genome and

Restoring autophagy-apoptosis balance in diabetic cardiomyopathy via a biomimetic, ROS-responsive nanocarrier associated with Mst1 pathway modulation.

Diabetic cardiomyopathy (DCM) is driven by oxidative stress and an imbalance between autophagy and apoptosis. To improve myocardial delivery of dimethyl fumarate (DMF), a known NRF2-activating redox modulator, and evaluate its association with Mst1 pathway regulation, we developed a cardiac-targeting peptide (APT)-modified biomimetic, reactive oxygen species (ROS)-responsive nanoplatform (NP-APT). NP-APT comprises a ROS-sensitive core and a lipid-cell membrane hybrid coating modified with APT to achieve targeted delivery. The designed nanoplatform demonstrated favorable physicochemical propert

Therapeutic Effect of Ethanol Extract of Propolis Against Osteoarthritis in a Mouse Model via Modulation of NF-κB and Hippo Signaling Pathways.

Osteoarthritis (OA), a chronic degenerative joint disease, leads to disability and increases the health burden. This study identified the chemical components in the ethanol extract of propolis (EEP) and investigated its effects and mechanism on monosodium iodoacetate-induced OA in mice. Three phenolic acid esters, alongside 16 flavonoids and their derivatives, were tentatively characterized in EEP using HPLC-MS, and four main components were quantified. EEP treatment alleviated pathological changes in articular cartilage and reduced serum CTX-II levels in OA mice. Network pharmacology and mole
