Histamine Content in Selected Production Stages of Fish Products.

INTRODUCTION: Histamine intoxication, known as scombroid fish poisoning, is caused by the consumption of foods with high levels of histamine. This biogenic amine is formed as a result of histidine decarboxylation by bacterial decarboxylases present in food, including fish and fish products. The aim of this study was to investigate the content of histamine at different production stages of canned, marinated and smoked fish. MATERIAL AND METHODS: Raw fish, semi-finished fish products, and the final products of the same production batches were collected between 2019 and 2022 from different fish …

Proliferative Effects of Histamine on Primary Human Pterygium Fibroblasts.

Purpose. It has been confirmed that inflammatory cytokines are involved in the progression of pterygium. Histamine can enhance proliferation and migration of many cells. Therefore, we intend to investigate the proliferative and migratory effects of histamine on primary culture of human pterygium fibroblasts (HPFs). Methods. Pterygium and conjunctiva samples were obtained from surgery, and toluidine blue staining was used to identify mast cells. 3-[4, 5-Dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) was performed to evaluate the proliferative rate of HPFs and human conjunctival fib…

Regional characteristics of histamine uptake into neonatal rat astrocytes.

Histaminergic signalling constitutes an attractive target for treatment of neuropsychiatric disorders. One obstacle to developing new pharmacological options has been failure to identify putative specific histamine transporter responsible for histamine clearance. Although high-affinity histamine uptake was detected in neonatal cortical astrocytes, its existence in other brain regions remains largely unexplored. We investigated whether cerebellar and striatal astrocytes participate in histamine clearance and evaluated the role of organic cation transporters (OCT) in astroglial histamine transpo…

Histamine-induced Ca2+ release in bovine adrenal chromaffin cells.

The histamine-induced biphasic increase of the intracellular free [Ca2+] ([Ca2+]i) was studied in bovine adrenal chromaffin cells using fura-2 microfluorimetry and the whole-cell patch-clamp technique. Both the rapid, transient Ca2+ rise and the sustained plateau component of elevated [Ca2+]i were independent of extracellular Ca2+. Incubation with the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) blocker thapsigargin diminished histamine-induced changes in [Ca2+]i. When Ca2+ release was either stimulated by IP3 or blocked with the competitive inhibitor heparin, histamine was unable to elicit…

Histamine and tissue fibrinolytic activity in duodenal ulcer disease.

Patients with duodenal ulcer have lower gastroduodenal mucosal histamine and a reduced tissue fibrinolysis at the ulcer edge. In the duodenal mucosa fibrinolysis is regulated by the tissue-type and urokinase-type plasminogen activators; and inhibitors type 1 and type 2. Trends across ordered groups leading from mucosa of nonulcer control subjects, "normal" mucosa of ulcer patients, to ulcer edge were found in tissue-type plasminogen activator concentration, histamine concentration, and histamine methyltransferase activity. Concentrations of tissue-type activator, inhibitor type 1, and histamin…

Monoclonal antibodies against glutaraldehyde-conjugated histamine: application to immunocytochemistry.

We have developed mouse monoclonal antibodies (AHA-1-5, all IgG1 sub-isotype mAbs) against histamine (HA) conjugated to bovine serum albumin using glutaraldehyde-NaBH4. Among these, AHA-1 mAb was found to be the most useful for HA immunocytochemistry (ICC) in terms of specificity and sensitivity without non-specific immunobinding. AHA-1 was demonstrated to be specific to HA with an enzyme-linked immunosorbent assay (ELISA) binding test, simulating the ICC of tissue sections, and not reactive to any of the other amino acids and peptides with N-terminal histidine tested. By use of this antibody,…

Interactions between neuronal histamine and halothane anesthesia in rats.

Using an in vivo microdialysis method, we measured the release of histamine in the anterior hypothalamic area (AHy) of rats under several concentrations of halothane anesthesia (1, 0.5, and 0.2%). The release of histamine increased to 341 and 325% at halothane concentrations of 0.5 and 0.2%, compared with the basal level at anesthesia induced by 1% halothane. alpha-Fluoromethylhistidine (100 mg/kg i.v.), a specific and irreversible inhibitor of histidine decarboxylase, reduced the histamine release to <35% of the basal value at 1% halothane anesthesia in the AHy, and also decreased the anesthe…

Histamine effect on human cutaneous blood flow: regional variations.

Different reactivities of small blood vessels to the histamine released by exogenous and endogenous substances may play a role in the regional variations of the elicited cutaneous response. To study the regional dependence of cutaneous blood flow in response to histamine, the compound was administered intradermally (prick introduction), thereby bypassing the spatially dependent penetration process. The induced response was quantified with cutaneous blood flow measurements utilizing laser Doppler flowmetry. Extent of response and time parameters were compared. Three anatomical sites, the back, …

Hypothalamic neuronal histamine regulates feeding circadian rhythm in rats.

To clarify involvement of hypothalamic neuronal histamine in feeding circadian rhythm, we analyzed rat behavioral patterns using chemical probes which affect endogenous histaminergic activity. Sustained infusion of alpha-fluoromethylhistidine (FMH), a specific suicide inhibitor of a histamine-synthesizing enzyme, into the rat third cerebral ventricle disrupted light-dark cycles of feeding, drinking, and ambulatory behavior. Food and water intake and ambulatory activity during the 12-h light period increased, and those during the 12-h dark period decreased after the infusion. The ratio of the l…

Indirect excitatory actions of histamine on isolated chicken oesophagus.

The histamine-induced contraction on chicken oesophagus was antagonised non-competitively by atropine, hexamethonium, cocaine, methysergide, indomethacin, theophylline and verapamil. Physostigmine slightly potentiated the excitatory action of histamine. These results indicate that histamine excreted its excitatory action by involving a number of mechanisms as suggested in guinea pig oesophagus.

Histamine-induced mucociliary dysfunction and otitis media with effusion.

Histamine, which has been found in middle ear effusions, is a potent pharmacological mediator released at an early stage of allergic reactions or general inflammatory process, increasing permeability of small blood vessels. Histamine might be involved in the origin and chronicity of middle ear effusions. In this study we studied the effect of histamine on the mucociliary function. First we examined the effect of histamine and its H1 and H2 blockers on the ciliary activity in the middle ear. 10(-2) M of histamine deteriorated ciliary activity; however, at lower concentrations ciliostimulatory e…

Histamine H3-receptors inhibit neurogenic microvascular leakage in airways.

We studied the role of histamine H3-receptors on neurogenic microvascular leakage in guinea pig airways by measuring extravasation of Evans blue dye. In guinea pig airways the major component of the neurogenic leakage is nonadrenergic noncholinergic (NANC) mediated, due to release of tachykinins. Anesthetized guinea pigs were pretreated with propranolol and atropine to block the beta-adrenergic and cholinergic responses and with mepyramine and cimetidine to block histamine H1- and H2-receptors, respectively. Vagal stimulation significantly increased leakage of dye in trachea (Tr), main bronchi…

Histamine poisoning (scombroid fish poisoning): an allergy-like intoxication.

Histamine poisoning results from the consumption of foods, typically certain types of fish and cheeses, that contain unusually high levels of histamine. Spoiled fish of the families, Scombridae and Scomberesocidae (e.g. tuna, mackerel, bonito), are commonly implicated in incidents of histamine poisoning, which leads to the common usage of the term, "scombroid fish poisoning", to describe this illness. However, certain non-scombroid fish, most notably mahi-mahi, bluefish, and sardines, when spoiled are also commonly implicated in histamine poisoning. Also, on rare occasions, cheeses especially …

Histamine release from human leukocytes by platelet-activating factor.

Platelet-activating factor (PAF) was found to induce histamine release from human basophils in mixed leukocytes in the presence of cytochalasin B. The reaction was rapid and dependent on temperature. The release was lower at a high concentration of PAF and was inhibited by EDTA, suggesting a noncytolytic mechanism. CV-3988, a PAF antagonist, inhibited the reaction dose-dependently. PAF acted as an IgE-independent stimulus, but PAF-induced histamine release from leukocytes of allergic asthmatics, who had elevated serum IgE levels, was significantly higher than that from leukocytes of controls. …
