Mapping the social network: tracking lice in a wild primate (Microcebus rufus) population to infer social contacts and vector potential.

BACKGROUND: Studies of host-parasite interactions have the potential to provide insights into the ecology of both organisms involved. We monitored the movement of sucking lice (Lemurpediculus verruculosus), parasites that require direct host-host contact to be transferred, in their host population of wild mouse lemurs (Microcebus rufus). These lemurs live in the rainforests of Madagascar, are small (40 g), arboreal, nocturnal, solitary foraging primates for which data on population-wide interactions are difficult to obtain. We developed a simple, cost effective method exploiting the intimate r…

Toxoplasma gondii MAF1b Binds the Host Cell MIB Complex To Mediate Mitochondrial Association.

Many diverse intracellular pathogens, such as Legionella pneumophila, Chlamydia psittaci, Encephalitozoon sp., and Toxoplasma gondii, manipulate and relocate host cell organelles, including mitochondria. Toxoplasma tachyzoites use a secreted protein, mitochondrial association factor 1b (MAF1b), to drive the association between the host mitochondria and the membrane of the parasitophorous vacuole, in which the parasites grow. The identity of the host partner in this interaction, however, has not previously been identified. By exogenously expressing tagged MAF1b in mouse embryonic fibroblasts, w

Infection, specificity and host manipulation of Australapatemon sp. (Trematoda, Strigeidae) in two sympatric species of leeches (Hirudinea).

Factors that drive parasite specificity and differences in infection dynamics among alternative host species are important for ecology and evolution of host-parasite interactions, but still often poorly known in natural systems. Here, we investigated spatiotemporal dynamics of infection, host susceptibility and parasite-induced changes in host phenotype in a rarely explored host-parasite system, the Australapatemon sp. trematode infecting two sympatric species of freshwater leeches, Erpobdella octoculata and Helobdella stagnalis. We show significant variation in infection abundance between the…

The prevalence and immune response to coinfection by avian haemosporidians in wild Eurasian blackbirds Turdus merula.

Coinfection of a host by more than 1 parasite is more common than single infection in wild environments and can have differing impacts, although coinfections have relatively rarely been quantified. Host immune responses to coinfection can contribute to infection costs but are often harder to predict than those associated with single infection, due to the influence of within-host parasite–parasite interactions on infection virulence. To first quantify coinfection in a common bird species, and then to test for immune-related impacts of coinfection, we investigated the prevalence and immune respo

The Schistosoma mansoni phylome: using evolutionary genomics to gain insight into a parasite's biology.

BACKGROUND: Schistosoma mansoni is one of the causative agents of schistosomiasis, a neglected tropical disease that affects about 237 million people worldwide. Despite recent efforts, we still lack a general understanding of the relevant host-parasite interactions, and the possible treatments are limited by the emergence of resistant strains and the absence of a vaccine. The S. mansoni genome was completely sequenced and still under continuous annotation. Nevertheless, more than 45% of the encoded proteins remain without experimental characterization or even functional prediction. To improve …

Schistosomiasis Vector Snails and Their Microbiota Display a Phylosymbiosis Pattern.

Planorbidae snails are the intermediate host for the trematode parasite of the Schistosoma genus, which is responsible for schistosomiasis, a disease that affects both humans and cattle. The microbiota for Schistosoma has already been described as having an effect on host/parasite interactions, specifically through immunological interactions. Here, we sought to characterize the microbiota composition of seven Planorbidae species and strains. Individual snail microbiota was determined using 16S ribosomal DNA amplicon sequencing. The bacterial composition was highly specific to the host strain w

Circular RNAs: An Under-Recognized Part in Different Scenarios of Host-Parasite Interrelationships.

Circular RNAs (circRNAs) are a special category of non-coding RNAs. The latter are known for their pivotal regulatory functions over many cellular processes. In biology, circRNAs exert regulatory functions on many physiological events, most likely through the regulation of gene expression. They are also implicated in a variety of health-related issues in medicine. Advances in molecular techniques and bioinformatics are expected to expand our knowledge of circRNAs, better characterizing their nature and functions. Remarkably, parasites elaborate their own repertoire of circRNAs to regulate diff

Costs of parasite generalism revealed by abundance patterns across mammalian hosts.

Most parasites are able to infect several host species, yet generalism is anticipated to be a costly strategy. Parasite abundance provides an important indicator of fitness because it is promoted by longevity and fecundity, which are correlated with a parasite's basic reproductive number. Using a global database of parasitic helminths of terrestrial mammals, we show that the abundance of parasitic helminths is phylogenetically structured across helminth species. High parasite abundance can be achieved through three separate components: infecting abundant host species, having a high infection s

Intraspecific competition between co-infecting parasite strains enhances host survival in African trypanosomes.

It is becoming increasingly clear that under natural conditions parasitic infections commonly consist of co-infections with multiple conspecific strains. Multiple-strain infections lead to intraspecific interactions and may have important ecological and evolutionary effects on both hosts and parasites. However, experimental evidence on intraspecific competition or facilitation in infections has been scarce because of the technical challenges of distinguishing and tracking individual co-infecting strains. To overcome this limitation, we engineered transgenic strains of the protozoan parasite Tr

Transgenic Plasmodium knowlesi: relieving a bottleneck in malaria research?

Plasmodium knowlesi is a primate malaria parasite that is phylogenetically close to the major human parasite Plasmodium vivax. P. knowlesi causes life-threatening disease in humans, infects a wide range of non-human primates and is one of few malaria parasites amenable to cyclical in vitro propagation. A robust in vivo and in vitro genetic manipulation system has been developed for this parasite, enabling in vitro-in vivo shuttling of transgenes, which (together with recent characterization of its genome and that of its macaque experimental host) offers unique opportunities to gain insight in

Synergistic effects of seasonal rainfall, parasites and demography on fluctuations in springbok body condition.

1. Seasonality of rainfall can exert a strong influence on animal condition and on host-parasite interactions. The body condition of ruminants fluctuates seasonally in response to changes in energy requirements, foraging patterns and resource availability, and seasonal variation in parasite infections may further alter ruminant body condition. 2. This study disentangles the effects of rainfall and gastrointestinal parasite infections on springbok (Antidorcas marsupialis) body condition and determines how these factors vary among demographic groups. 3. Using data from four years and three study…

Inter-specific parasite competition: mixed infections of Schistosoma mansoni and S. rodhaini in the definitive host.

Competition between parasite species has been predicted to be an important force shaping parasite and host ecology and evolution, although empirical data are often lacking. Using the Mus musculus-Schistosoma mansoni and Schistosoma rodhaini host-parasite systems we characterized mate choice and inter-specific competition between these two schistosome species. Simultaneous infections revealed species-specific mate preferences for both species as well as suggesting mating competition, with male S. rodhaini appearing dominant over male S. mansoni. S. rodhaini homologous pairs were also shown to h

Ant parasitoidism in checkered beetles: Phyllobaenus obscurus developing inside intact cocoons of two species of the Ectatomma ruidum species complex.

Known parasitoids of ants include species from several families of flies, wasps, strepsipterans, nematodes, and mites. Curiously, while myrmecophily is heavily biased towards Coleoptera, one of the most diverse and speciose insect orders, no beetles specialized as parasitoids of ants have been recorded, although the parasitoid habit has evolved at least 13 times within this order. Here we report on observations that strongly suggest that a checkered beetle species behaves as a parasitoid of ant brood. A total of 146 colonies or part of colonies of three species of the Ectatomma ruidum species

Manipulation of the shore crab Metopograpsus thukuhar by the parasitic barnacle Polyascus planus (Crustacea: Cirripedia: Rhizocephala): Effects of the parasitism on hemolymph parameters and rootlet-infiltrated tissues of the host.

The present study assessed the impact of parasitism of Polyascus planus on the hemolymph parameters and rootlet-infiltrated tissues of the shore crab Metopograpsus thukuhar. First, hemolymph parameters were compared between hosts harboring internal infection (Hit) and non-parasitized host crabs (Hnp). The percentage of Hit with culturable bacteria in the hemolymph was significantly higher than that of Hnp when samples were cultured on Luria-Bertani (LB) agar, but the difference between Hit and Hnp was statistically insignificant when samples were cultured on Vibrio-selective X-VP agar.
