Divergence in virus-host protein interactomes across species explains disparities in translational therapeutic success.

Infectious viral diseases remain a major and persistent threat to human health, and findings from animal models often translate poorly to clinical applications. Here, we constructed protein interactomes for seven species, including humans, revealing substantial differences in research biases. A random sampling strategy was employed to generate human interactomes, with sizes matched to those of the other six species to ensure cross-species comparability. We found that the clustering of virally targeted proteins within the interactomes (measured as largest connected component [LCC] proportion) a

Studying Microbial Communities In Vivo: A Model of Host-mediated Interaction Between Candida Albicans and Pseudomonas Aeruginosa in the Airways.

Studying host-pathogen interaction enables us to understand the underlying mechanisms of the pathogenicity during microbial infection. The prognosis of the host depends on the involvement of an adapted immune response against the pathogen. Immune response is complex and results from interaction of the pathogens and several immune or non-immune cellular types. In vitro studies cannot characterise these interactions and focus on cell-pathogen interactions. Moreover, in the airway, particularly in patients with suppurative chronic lung disease or in mechanically ventilated patients, polymicrobial

High-dimensional linear state space models for dynamic microbial interaction networks.

Medical researchers are increasingly interested in knowing how the complex community of micro-organisms living on human body impacts human health. Key to this is to understand how the microbes interact with each other. Time-course studies on human microbiome indicate that the composition of microbiome changes over short time periods, primarily as a consequence of synergistic and antagonistic interactions of the members of the microbiome with each other and with the environment. Knowledge of the abundance of bacteria-which are the predominant members of the human microbiome-in such time-course

Flipons, Noncoding Transcripts and Retroelements: A Universal Host Defense.

Encoding genetic information involves both structure and sequence. Non-protein-coding (NCR) RNAs modulate host immune responses by guiding and assembling stress-specific protein effectors, with flipons enabling dynamic transitions between states. Small NCRs transcribed by both RNA Polymerases 2 and 3 normally modulate these pathways. The miRNAs, tRNA fragments, mitrons, piRNAs, and endo-small-interfering RNA products target the guide effector complexes to their targets. These NCRs filter the subset of RNAs that are translated, a process modulated by other NCRs that fine-tune RNA stability, loc

Bridging two kingdoms: binding interactions between enteric viruses and intestinal bacteria.

Enteric viruses are major causes of gastrointestinal disease worldwide and are primarily transmitted through the fecal-oral route. Upon entering the gastrointestinal tract, these viruses encounter the dense and diverse population of intestinal microbiota, leading to frequent interactions with commensal bacteria. Over the past decade, numerous studies have demonstrated that bacteria can enhance enteric viral replication, stability, and overall pathogenesis. Although direct binding interactions between enteric viruses and bacteria have been observed, the molecular determinants governing these in

Intracellular pathogen manipulation of ER-phagy: direct receptor targeting and indirect pathway perturbation.

Selective autophagy of the endoplasmic reticulum (ER-phagy) maintains intracellular homeostasis by selectively removing damaged ER membranes and contained misfolded proteins. This review delineates the core molecular mechanisms of canonical autophagy, then systematically describes three types of ER-phagy (macro-ER-phagy, micro-ER-phagy, and vesicular ER-phagy), their signature receptors, subdomain specificities, and regulatory mechanisms, alongside a comparative analysis of the evolutionary divergence between yeast and mammalian ER-phagy receptor systems. At the direct regulation level, divers

Extracellular vesicles in human fungal pathogens: Biogenesis, functions, and translational applications.

Fungal extracellular vesicles (EVs) have emerged as critical mediators of fungal physiology, virulence, and host-pathogen interactions. Since their first description in Cryptococcus neoformans, EVs have been identified in several fungal species and shown to transport a broad repertoire of bioactive cargo. Increasing evidence indicates that fungal EVs participate in multiple biological processes, including cell wall remodeling, stress adaptation, biofilm formation, antifungal resistance, and modulation of host immune responses. Recent advances in cryo-electron microscopy, multi-omics approaches…

A Trojan horse pathogen breaking through partner-choice barriers in the insect gut.

Mutualistic symbioses are potentially vulnerable to exploitation, particularly in hosts that acquire symbionts from the environment, where harmful exploiters inhabit. The independent evolution and persistence of intricate partner-choice mechanisms in many symbioses testify the threat by specialized exploiters of mutualisms, although only few have been documented in nature. We report here a lethal "Trojan horse" pathogen, Burkholderia sp. SJ1, exploiting the stinkbug-Caballeronia gut symbiosis. This bacterium resembles symbionts by using wrapping motility to traverse the host's sorting organ, i

From biogenesis to host modulation: the expanding biology of oral streptococcal membrane vesicles.

Bacterial extracellular membrane vesicles (bEVs) provide fundamental biological functions through communication, ecological adaptation, and mediation of host interactions. Gram-positive oral streptococci were long considered to be incapable of bEVs production due to their thick peptidoglycan. In contrast, recent advances in imaging, biochemical isolation, and multi-omics profiling have revealed that bEVs production is common and may represent a general trait of streptococci. This review summarizes current research on the biogenesis, cargo composition, and functional roles of bEVs, with an emph

The interplay between ecological networks drives host-plasmid community dynamics.

Plasmids drive evolution by transferring traits across microbial hosts. Transmission depends on both host-plasmid (infection) and plasmid-plasmid (compatibility) interactions, yet how the structure of these networks shapes transmission remains poorly understood. We hypothesized that these two ecological networks interact in non-additive ways to influence community outcomes. To test this, we developed a stochastic agent-based model that embeds both network structures and simulates coupled host-plasmid dynamics. We systematically varied the structure of each network, both individually and in com

Beyond Pathogenesis: The Nematode Immune Network as the Arbiter of a Host-Virus Truce.

The phylum Nematoda is host to a vast and diverse virosphere, yet severe viral diseases are rarely observed. This paradox between pervasive infection and limited pathology suggests the existence of a highly effective host-virus "truce". In this review, we argue that this truce is not a result of viral attenuation but is actively arbitrated by a multi-tiered host immune network, whose primary characteristic is not destructive power but exquisite cost-benefit management. We deconstruct this network into two functional tiers. The first, the "effector layer", comprises a diverse arsenal of antivir

Obligate intracellular bacteria and host cell death pathways-the matter of life and death.

To limit the damage caused by pathogenic bacteria, host organisms possess different defense systems and mechanisms for preventing infection, combating the pathogens, and creating a memory that will avert recurrent infections. Pathogens, on the other hand, have developed countermeasures to enable their replication and spreading. For intracellular pathogenic bacteria, the battleground is localized at the cellular level. Different cell types, including phagocytic, epithelial, and endothelial cells, fibroblasts, and trophoblasts, not only are equipped with diverse defense tools, but also provide d

More than just membranes: membrane contact sites as crossroads for infections.

Membrane contact sites (MCSs), tethering zones between organelles, have emerged as critical hubs for regulating cellular metabolism, homeostasis, and immune responses. Recent discoveries reveal that a wide range of intracellular pathogens, including bacteria, viruses, parasites, and fungi, exploit MCSs to establish and maintain their replicative niches within host cells. By co-opting the host MCS machinery, these pathogens create specialized interfaces between their vacuoles, replication complexes, or cytosolic domains and host organelles, enabling nutrient acquisition, immune evasion, and spa

The 'ins and outs' of Brucella intracellular journey.

Members of the genus Brucella are the causative agents of brucellosis, a worldwide zoonosis affecting wild and domestic animals and humans. These facultative intracellular pathogens cause long-lasting chronic infections by evolving sophisticated strategies to counteract, evade, or subvert host bactericidal mechanisms in order to establish a secure replicative niche necessary for their survival. In this review, we present recent findings on selected Brucella effectors to illustrate how this pathogen modulates host cell signaling pathways to gain control of the vacuole, promote the formation of

Time-series gene expression patterns and their characteristics of Beauveria bassiana in the process of infecting pest insects.

Beauveria bassiana has been widely used as an important biological control fungus for agricultural and forest pests, and clarifying the interaction mechanism between B. bassiana and its host will help to better exert the efficacy of the mycoinsecticide. Here, we proposed a novel pattern analysis (PA) method for analyzing time-series data and applied it to a transcriptomic data set of B. bassiana infecting Galleria mellonella. We screened out 14 patterns including 868 genes, which had some characteristics that were not inferior to differentially expressed genes (DEGs). Compared with the previou

GroEL protein of the Leptospira spp. interacts with host proteins and induces cytokines secretion on macrophages.

BACKGROUND: Leptospirosis is a zoonotic disease caused by infection with spirochetes from Leptospira genus. It has been classified into at least 17 pathogenic species, with more than 250 serologic variants. This wide distribution may be a result of leptospiral ability to colonize the renal tubules of mammalian hosts, including humans, wildlife, and many domesticated animals. Previous studies showed that the expression of proteins belonging to the microbial heat shock protein (HSP) family is upregulated during infection and also during various stress stimuli. Several proteins of this family are

Interplay Between Toxoplasma gondii, Autophagy, and Autophagy Proteins.

Survival of Toxoplasma gondii within host cells depends on its ability of reside in a vacuole that avoids lysosomal degradation and enables parasite replication. The interplay between immune-mediated responses that lead to either autophagy-driven lysosomal degradation or disruption of the vacuole and the strategies used by the parasite to avoid these responses are major determinants of the outcome of infection. This article provides an overview of this interplay with an emphasis on autophagy.

The periodontal war: microbes and immunity.

Maintenance of periodontal health or transition to a periodontal lesion reflects the continuous and ongoing battle between the vast microbial ecology in the oral cavity and the array of resident and emigrating inflammatory/immune cells in the periodontium. This war clearly signifies many 'battlefronts' representing the interface of the mucosal-surface cells with the dynamic biofilms composed of commensal and potential pathogenic species, as well as more recent knowledge demonstrating active invasion of cells and tissues of the periodontium leading to skirmishes in connective tissue, the locali

Leptospirosis in human: Biomarkers in host immune responses.

Leptospirosis remains one of the most widespread zoonotic diseases caused by spirochetes of the genus Leptospira, which accounts for high morbidity and mortality globally. Leptospiral infections are often found in tropical and subtropical regions, with people exposed to contaminated environments or animal reservoirs are at high risk of getting the infection. Leptospirosis has a wide range of clinical manifestations with non-specific signs and symptoms and often misdiagnosed with other acute febrile illnesses at early stage of infection. Despite being one of the leading causes of zoonotic morbi
