Self-Growing Conductive Hydrogels Establish Volumetric Biointerfaces for Cardiac Conduction Restoration.

Restoring three-dimensional electrical conduction in infarcted myocardium remains a critical challenge, as conventional conductive hydrogel patches largely remain surface-confined and prevent electrical coupling of residual cardiomyocytes within fibrotic scars. Here, we present a self-growing conductive volumetric interface (SCOVE) that transforms surface-confined biointerfaces into tissue-integrated, three-dimensional conductive networks. SCOVE is delivered as an injectable hydrogel precursor containing the tissue-permeable conductive monomer 3,4-ethylenedioxythiophene-acetic acid sodium salt

Preparation of Quercetin-Loaded Lipid Nanoparticle-Embedded Hydrogels and Stability Studies.

Quercetin, a plant-derived flavonoid with potent antioxidant and anti-inflammatory properties, is limited for topical use by its poor aqueous solubility, low bioavailability, and chemical instability. This research developed and validated a formulation-driven strategy to stabilize quercetin by encapsulating it in lipid nanoparticles (QLNs) and embedding these in a Carbopol hydrogel, providing comprehensive physicochemical and functional stability data. A fully validated HPLC-PDA assay was used to quantify quercetin in the nanoparticle-hydrogel matrix. In vitro bioactivity testing showed notabl

In situ particle-to-fibre transformation of hydrogels for 3D printing.

Structural anisotropy is ubiquitous in nature and governs orientation-dependent properties and functions1,2. Recreating biomimetic anisotropy in hydrogels-water-rich soft materials widely used in biomedicine-has proven challenging3. Here we report a phenomenon termed shear-extensional in situ particle-to-fibre transformation (SHIFT) and demonstrate the SHIFT printing of highly aligned hydrogel microfibres (5-30 μm in diameter). In SHIFT, particle-embedded hydrogel is extruded in a controlled manner, in which the discrete particles and surrounding matrix experience opposite phase transitions, c…

Engineered responsive-hydrogels drive cGAS-STING pathway activation for cancer immunotherapy.

The cGAS-STING pathway holds considerable promise for cancer immunotherapy, yet its clinical translation is generally constrained by a narrow therapeutic window and the risk of systemic inflammation, making precision activation a central challenge. Engineered hydrogels provide a compelling strategy by serving as injectable, tissue-adherent depots that enable prolonged retention and programmable release within the tumor microenvironment. This review first outlines the immunological rationale and therapeutic window for cGAS-STING activation, then surveys hydrogel engineering strategies, includin…

Streptavidin functionalized hyaluronic acid hydrogels for controlled and customizable drug delivery.

Hyaluronic acid (HA)-based hydrogels have emerged as promising materials for tissue repair given their ability to rapidly gel in situ to precisely fill tissue defects, their mechanical tunability and customizable degradation kinetics, and their capacity to deliver therapeutic agents to the site of injury. HA hydrogels can also be used to encapsulate therapeutics for local delivery; however, release through diffusion may be too rapid to achieve a therapeutic window for many applications. To overcome this limitation, we functionalized norbornene-modified HA (NorHA) hydrogels with streptavidin us…

Copper-Coordinated Hyaluronic Acid Hydrogels with Antibacterial and Anti-Inflammatory Activities.

Chronic infected wounds are often characterized by persistent bacterial colonization, biofilm formation, excessive oxidative stress, and prolonged inflammation, which severely impair tissue regeneration. To address these challenges, a multifunctional wound dressing capable of antibacterial activity and microenvironment modulation was developed. In this study, amide-modified hyaluronic acid (HA-ADH) was used as the matrix, and a dynamic coordination network was constructed via Cu2+-hydrazide interactions to form an in situ HA-Cu hydrogel. Curcumin-loaded DSPE-PEG2000 micelles were further incor

Kinetic-Locked Fiber Networks Enable Anti-Fouling, Mechanically Robust Zwitterionic Hydrogels.

Zwitterionic hydrogels possess exceptional antifouling characteristics and biocompatibility, but their weak mechanical strength limits implantable applications. Existing enhancement strategies either introduce non-zwitterionic phases that compromise antifouling performance or employ zwitterionic fillers that still fall short of achieving high mechanical robustness. Here, we present a kinetic-locking strategy to fabricate strong, antifouling zwitterionic composite fibers for hydrogel reinforcement. Co-dissolving hydrophilic zwitterionic and hydrophobic polymers followed by electrospinning kinet…
