Nitric oxide alters nitrogen metabolism and PIN gene expressions by playing protective role in arsenic challenged Brassica juncea L.

Plants have ability to adapt themselves through altering their growth process. In the present study, we examined exogenous application of nitric oxide (NO) on nitrogen metabolism and auxin (PIN) gene expression, and its possible role in alleviation of arsenic (As) toxicity in Brassica juncea seedlings. Seven days old hydroponically grown B. juncea seedlings were exposed to AsIII (150 μM), Sodium nitroprusside (NO donor, 100 μM), AsIII + SNP and control (without metal)for 48 h. Experimental results revealed that AsIII stress: enhanced the level of nitrite, NiR activity, NO3- and NH4+content as

Sulfur-oxidizing buffalo dung bacteria enhance growth and yield of Foeniculum vulgare Mill.

This study aimed to harness the benefits of sulfur-oxidizing beneficial bacteria from buffalo dung to improve crop yields of Foeniculum vulgare. A total of 61 bacterial isolates were screened from buffalo dung, of which 40 isolates exhibited plant-growth-promoting attributes, such as phosphate solubilization, indole-3-acetic acid production, and hydrogen cyanide production. Of these 40, four bacterial isolates, viz., BUFF12, BUFF14, BUFF23, and BUFF38, were the most potent, having plant-growth-promoting and sulfur-oxidizing properties. These four isolates produced phytase by solubilizing calci

Effect of calmodulin antagonists on the growth and graviresponsiveness of primary roots of maize.

We examined the effect of calmodulin (CaM) antagonists applied at the root tip on root growth, gravity-induced root curvature, and the movement of calcium across the root tip and auxin (IAA) across the elongation zone of gravistimulated roots. All of the CaM antagonists used in these studies delayed gravity-induced curvature at a concentration (1 micromole) that did not affect root growth. Calmodulin antagonists (> or = 1 micromole) inhibited downward transport of label from 45Ca2+ across the caps of gravistimulated roots relative to the downward transport of 45Ca2+ in gravistimulated roots wh

Nitric Oxide Pre-Treatment Advances Bulblet Dormancy Release by Mediating Metabolic Changes in Lilium.

The lily is a globally popular cut flower, and managing dormancy in lily bulblets is essential for continuous, year-round production. While nitric oxide (NO) has been shown to influence seed dormancy and germination, its role in dormancy release in lilies was previously unconfirmed. In this study, we investigated the effects of NO on dormancy release in lily bulblets using SNP and c-PTIO. Results showed that SNP treatment promoted dormancy release, while c-PTIO inhibited it. Measurement of endogenous NO levels in the bulbs, along with enzyme activities of NOS-like and NR and gene expression le

Argiotoxin636 inhibits NMDA-activated ion channels expressed in Xenopus oocytes.

Argiotoxin636, a component of the spider venom of argiope species, was chemically synthesized together with a number of derivatives in order to analyse their blocking activity on mammalian glutamate receptors. Xenopus laevis oocytes injected with rat brain mRNA served as assay system. The results showed that argiotoxin636 had a higher affinity for N-methyl-D-aspartate (NMDA) than for kainate receptors, blocking the corresponding ion channels in a voltage-dependent manner. Modifications of the polyamine tail or the terminal arginine residue strongly reduced the blocking potency. The iodinated m

RUBY expression alters development and metabolism in Arabidopsis.

Genetically encoded reporters are indispensable tools for monitoring gene expression, transformation efficiency, and diverse cellular processes in plants. The betalain-based RUBY reporter has recently emerged as a convenient visual marker requiring no substrates or specialized equipment, and it has been widely used for a variety of applications. However, the developmental and metabolic consequences of its expression remain unexplored. Here, we show that independent transgenic lines of Arabidopsis thaliana expressing RUBY exhibit profound alterations in growth and reproductive development. RUBY

Bridging sugar metabolism and signaling with hormones in shoot branching regulation.

Shoot branching, a major determinant of plant architecture and productivity, has long been attributed to the coordinated action of auxin, cytokinins (CKs), and strigolactones (SLs). Recent work shows that sugars act not only as metabolic resources but also as early developmental signals that regulate axillary bud outgrowth, the initial step in shoot branching. Sugar availability can rapidly trigger bud outgrowth, often preceding major hormonal changes. Mechanistically, sugars antagonize auxin and SL signaling while acting synergistically with CKs. These effects involve sugar signaling pathways

TaIAA25 negatively regulates wheat alkaline tolerance by inhibiting plasma membrane H+-ATPase activity.

Soil salinization/alkalization represents a universal challenge that severely constrains crop productivity. Elucidating the molecular mechanisms underpinning saline/alkaline tolerance of wheat (Triticum aestivum) holds profound significance for global food security and sustainable agriculture. In this study, we functionally characterized a wheat alkaline sensitive locus designated Wheat Alkaline Sensitive 1 (WAS1), through whole exome-capture sequencing-based bulked segregant analysis and fine mapping of was1 mutant populations. This locus encodes TaIAA25, a canonical member of the Auxin/Indol

Cytokinin represses NH4+ efflux by suppressing GH3.5 transcription through an ARR10/12-dependent pathway.

Ammonium (NH4+) is an essential nitrogen source for plants, yet its excessive accumulation triggers toxicity, a process typically coupled to futile NH4+ cycling in roots cells. While the cytokinin-ARR10/12 signaling pathway is known to regulate vacuolar NH4+ sequestration, it is not known whether it also coordinates NH4+ efflux across the root-cell plasma membrane to maintain intracellular homeostasis. Here, we identify the auxin-amido synthetase GH3.5 as a direct transcriptional target of ARR10, using a combination of yeast one-hybrid (Y1H) assays, dual-luciferase reporter assays, and CUT&RUN

miR169a-MhNF-YA2 module modulates H2S dosage-dependent lateral root formation via MhPIN1 and MhCYCB1;2 in Malus hupehensis.

Hydrogen sulfide regulates lateral root formation depending on its dosage, yet the underlying molecular mechanism remains not fully elucidated. Here, applying 0.5 mM and 0.05 mM sodium hydrosulfide promoted lateral root formation and upregulated miR169a expression in Malus hupehensis and Arabidopsis, respectively, whereas 1.0 mM sodium hydrosulfide inhibited it. Silencing miR169a reduced lateral root number and attenuated the improvement of 0.5 mM and 0.05 mM sodium hydrosulfide on lateral root formation, while its overexpression alleviated the 1.0 mM sodium hydrosulfide inhibition on lateral

Auxin biosynthesis and signaling drive virulence and plant adaptation in Dickeya dadantii.

Plants and their associated bacteria engage in complex bidirectional interactions mediated by a diverse array of signaling molecules of both plant and microbial origin. Indole-3-acetic acid (IAA) is a central phytohormone regulating plant growth and development and is increasingly recognized as an intra- and inter-kingdom signaling molecule that modulates diverse bacterial processes relevant during plant-microbe interactions. While IAA biosynthesis is widespread among plant-associated bacteria, the mechanisms through which this auxin regulates bacterial physiology and virulence, as well as tho

Lactobacillus acidophilus LA3 Suppresses Neuroinflammation via the Gut-Brain Axis.

The gut-brain axis represents a pivotal pathway through which gut microbiota influence neuroinflammation and neurological health. This study identifies Lactobacillus acidophilus LA3 as a potent probiotic strain capable of mitigating neuroinflammation. LA3 demonstrated superior efficacy in suppressing LPS-induced proinflammatory cytokines in BV-2 microglia. In an LPS-induced murine model, LA3 alleviated systemic and neuroinflammation, depressive-like behaviors, and cognitive impairment, while restoring blood-brain and gut-vascular barrier integrity. LA3 reversed gut microbiota dysbiosis, enrich

Tissue-specific consequences of tag fusions on protein expression in transgenic mice.

Genetic fusion of protein tags is widely used to study protein functions in vivo. It is well known that tag fusion can cause unwanted changes in protein stability, but whether this is an inherent property of the tagged protein, or can be influenced by the cell and tissue environment, is unclear. Using a series of genome edited mouse models, we show that tag-dependent changes in protein expression can vary across different primary cell and tissue contexts. In one case (Ncaph2), a C-terminal auxin-inducible degron fusion strongly increased protein stability in some tissues but decreased it in ot

Clostridium scindens attenuates acute kidney injury by producing indole-3-acetic acid.

The association between gut microbiota and acute kidney injury (AKI) has garnered increased attention recently. Herein, we investigated the effect of the gut microbiota and its metabolites on regulating AKI-associated kidney injury and inflammation. We observed that Clostridium scindens (CS) can ameliorate ischemia/reperfusion injury/folic acid-induced renal dysfunction, oxidative stress, and inflammation, and enhance intestinal barrier function. Mechanistically, CS can facilitate indole-3-acetic acid (IAA) production via the tryptophan metabolic pathway: tryptophan-indole-3-pyruvic acid-IAA.

PmLBD3 links auxin and brassinosteroid signalling pathways on dwarfism in Prunus mume.

BACKGROUND: Grafting with dwarf rootstock is an efficient method to control plant height in fruit production. However, the molecular mechanism remains unclear. Our previous study showed that plants with Prunus mume (mume) rootstock exhibited a considerable reduction in plant height, internode length, and number of nodes compared with Prunus persica (peach) rootstock. The present study aimed to investigate the mechanism behind the regulation of plant height by mume rootstocks through transcriptomic and metabolomic analyses with two grafting combinations, 'Longyan/Mume' and 'Longyan/Peach'. Indole-3-acetaldoxime delays root iron-deficiency responses and modify auxin homeostasis in Medicago truncatula.

Iron (Fe) is an essential plant micronutrient, being a major limiting growth factor in calcareous soils. To increase Fe uptake, plants induce lateral roots growth, the expression of a Fe(III)-chelate reductase (FCR), a Fe(II)-transporter and a H+-ATPase and the secretion of flavins. Furthermore, auxin hormone family is involved in the Fe-deficiency responses but the action mechanism remains elusive. In this work, we evaluated the effect of the auxin-precursor indole-3-acetaldoxime (IAOx) on hydroponically grown Medicago truncatula plants under different Fe conditions. Upon 4-days of Fe starvat

Effects of copper accumulation on growth and development of Scopelophila cataractae grown in vitro.

Scopelophila cataractae was cultured in vitro for 16 weeks to assess the contrasting effects of Cu on growth and reproduction, as well as gametophore stage. To induce buds and gametophores of S. cataractae, ten treatments (tr 1 to tr 10) of culture media were prepared using a combination of mineral salts, sugar, vitamin B complex, CuSO4, and exogenous hormones. Highest numbers of gametophores and buds were formed in media containing 500 µM CuSO4 in co-application with auxin and cytokinin, as shown in the modest Cu treatments (tr 6 and tr 7, 26 per cushion and 255 per 25 mm2, respectively). Melatonin Function and Crosstalk with Other Phytohormones under Normal and Stressful Conditions.

Melatonin was discovered in plants in the late nineties, but its role, signaling, and crosstalk with other phytohormones remain unknown. Research on melatonin in plants has risen dramatically in recent years and the role of this putative plant hormone under biotic and abiotic stress conditions has been reported. In the present review, we discuss the main functions of melatonin in the growth and development of plants, its role under abiotic stresses, such as water stress (waterlogging and drought), extreme temperature (low and high), salinity, heavy metal, and light-induced stress. Similarly, w
