Prevalence of breech-strike in mulesed, clipped and unmulesed Merino hoggets in south-eastern Australia.

OBJECTIVE: To compare the prevalence of breech flystrike ('breech-strike') on 12-20-month-old Merino hoggets that were mulesed, had breech and tail clips applied ('clipped') or were left unmulesed but treated strategically with long-acting insecticide. DESIGN: A cohort study on three farms in southern Victoria. METHODS: Three treatment groups were established at lamb marking on each farm. The unmulesed group was treated with dicyclanil when 12 months old in September or October ('early-season treatment'). The prevalence of breech-strike and key risk factors, including soiling of the breech w

Regulation of growth-blocking peptide expression during embryogenesis of the cabbage armyworm.

Growth-blocking peptide (GBP) is an insect cytokine with diverse biological functions. Northern blot analysis revealed high heterogeneity in the size distribution of GBP mRNAs as well as in the tissues where they are detected. The spatio-temporal transcription pattern is dynamic, especially during embryogenesis. Gel shift assays demonstrated that the cabbage armyworm embryo nuclear extract specifically binds to a 178-bp element, at position +234 to +411 from the transcription start site of the 1.3 kb GBP transcript, in which two Drosophila Deformed (Dfd) binding sites are repeated in tandem. The function of engrailed and the specification of Drosophila wing pattern.

The adult Drosophila wing (as the other appendages) is subdivided into anterior and posterior compartments that exhibit characteristic patterns. The engrailed (en) gene has been proposed to be paramount in the specification of the posterior compartment identity. Here, we explore the adult en function by targeting its expression in different regions of the wing disc. In the anterior compartment, ectopic en expression gives rise to the substitution of anterior structures by posterior ones, thus demonstrating its role in specification of posterior patterns. The en-expressing cells in the anterior

Purification, sequence and antibacterial activity of two novel sapecin homologues from Sarcophaga embryonic cells: similarity of sapecin B to charybdotoxin.

Two sapecin homologues were purified from the culture medium of NIH-Sape-4, an embryonic cell line of Sarcophaga peregrina (flesh fly). These homologues contained six cysteine residues with exactly the same disulphide pairings as those in sapecin. The amino acid sequence of one of them, sapecin C, was also very similar to that of sapecin. The other homologue, sapecin B, was less similar to sapecin but showed significant similarity to charybdotoxin, an inhibitor of K+ channels isolated from a scorpion venom. Like sapecin, these homologues repressed the growth of various Gram-positive bacteria.

Regulation of a decapentaplegic midgut enhancer by homeotic proteins.

The clustered homeotic genes encode transcription factors that regulate pattern formation in all animals, conferring cell fates by coordinating the activities of downstream 'target' genes. In the Drosophila midgut, the Ultrabithorax (Ubx) protein activates and the abdominalA (abd-A) protein represses transcription of the decapentaplegic (dpp) gene, which encodes a secreted signalling protein of the TGF beta class. We have identified an 813 bp dpp enhancer which is capable of driving expression of a lacZ gene in a correct pattern in the embryonic midgut. The enhancer is activated ectopically in

The andropin gene and its product, a male-specific antibacterial peptide in Drosophila melanogaster.

In our study of the cecropin locus in Drosophila we have found a gene for a new peptide, andropin, with antibacterial properties. Transcripts from this gene, Anp, could be detected in newly eclosed males and reached steady-state levels after 1 day. Transcription was strongly induced in response to mating and is strictly confined to the ejaculatory duct of adult males. The deduced peptide sequence reveals a hydrophobic amino terminus with striking similarity to the signal peptide of the cecropins. The sequence of the predicted mature andropin shows no direct homology with the cecropins, but the

Royal jelly attenuates cadmium-induced nephrotoxicity in male mice.

Cadmium exposure induces nephrotoxicity by mediating oxidative stress, inflammation, and apoptosis. The purpose of this study was to examine the protective effect of royal jelly on Cd-induced nephrotoxicity. Adult male mice were distributed randomly into 4 clusters: untreated, royal jelly-treated (85 mg/kg, oral), CdCl2-treated (6.5 mg/kg, intraperitoneal), and pretreated with royal jelly (85 mg/kg) 2 h before CdCl2 injection (6.5 mg/kg, intraperitoneal) for seven consecutive days. Cd concentration in the renal tissue and absolute kidney weight of the Cd-treated mice were significantly higher

Female reproductive toxicity after exposure to malathion or diazinon: a systematic review of rodent and human studies.

Malathion and diazinon are pesticides widely used in agriculture as well as for domestic and veterinary purposes to control pests, such as scale insects, aphids, and fleas. However, these compounds may have harmful effects on the female genital system in humans and animals. This study conducted a systematic review of the scientific literature on the female reproductive effects in humans and rodents exposed to malathion or diazinon. The search was conducted from March to September 2024 and was updated in July 2025. It was carried out in the Embase, LILACS, PubMed, SciELO, Scopus, and Web of Sci

Hormonal induction of Dopa decarboxylase in the epidermis of Drosophila is mediated by the Broad-Complex.

The 2B5 early puff locus corresponds to the Broad-Complex BR-C) and encodes a family of transcription factors whose members are induced by the molting hormone ecdysone. Mutations in the br subcomplementation group substantially reduce the levels of Dopa decarboxylase (DDC) in the epidermis of mature third instar larvae but not in mature second instar organisms. Enzyme levels are normal in the central nervous system of the two mutants examined. The specificity of these effects suggests that a product of the BR-C locus mediates the rapid appearance of DDC in mature third instar larvae experienci

Ovarian transcriptome analyses indicate that weak juvenile hormone signaling underlies the molecular basis of oogenesis deficiencies in mosquitoes.

BACKGROUND: Juvenile hormone (JH) is synthesized by the corpora allata (CA) and controls development and reproduction in insects. We recently used CRISPR/Cas9 to establish a line lacking the enzyme that catalyzes the final step of JH biosynthesis in mosquitoes, a P450 epoxidase. The CA of the epox-/- mutants do not synthesize epoxidized JH III but methyl farneosate (MF), a weak agonist of the JH receptor. Female epox-/- mosquitoes have reduced JH signaling and show a substantial loss of reproductive fitness. To understand the molecular basis of this loss of fitness, we constructed ovarian mRNA

Adult nutrient shortage impairs female reproduction via the attenuated juvenile hormone signaling during vitellogenesis in Helicoverpa armigera (Lepidoptera: Noctuidae).

In insect reproduction, vitellogenesis is a prerequisite to oocyte maturation and critically depends on nutrient acquisition, supporting reproductive success. In Helicoverpa armigera (Hübner 1809), a hallmark of female reproduction is that vitellogenin (Vg) synthesis is initiated following adult eclosion, and juvenile hormone (JH) plays a principal role in this process. The adults commonly visit flowers to feed on nectar, which implies the possible linkage between adult nutrient and vitellogenesis. The preliminary experiment demonstrated that the supplemental nutrition with 10% honey during vi

Dual roles of methoprene-tolerant gene TaMet in male molting and female reproduction of the tomato leafminer, Tuta absoluta (meyrick).

The tomato leafminer (Tuta absoluta) is a highly destructive global quarantine pest. The methoprene-tolerant (Met) protein, a member of the bHLH/PAS family of transcription factors, forms complexes with other family members to transduce the juvenile hormone signal, which regulates insect growth and development. However, the functions of the TaMet gene have rarely been studied in T. absoluta. Herein, we investigated the significance of TaMet in T. absoluta. Spatiotemporal expression analysis revealed that TaMet exhibited comparable expression patterns in males and females, with high expression

Neurobiology of egg-laying behavior in Drosophila: neural control of the female reproductive system.

Egg-laying is one of the key aspects of female reproductive behavior in insects. Egg-laying has been studied since the dawn of Drosophila melanogaster as a model organism. The female's internal state, hormones, and external factors, such as nutrition, light, and social environment, affect egg-laying output. However, only recently, neurobiological features of egg-laying behavior have been studied in detail. fruitless and doublesex, two key players in the sex determination pathway, have become focal points in identifying neurons of reproductive significance in both central and peripheral nervous

Co-option of immune effectors by the hormonal signalling system triggering metamorphosis in Drosophila melanogaster.

Insect metamorphosis is triggered by the production, secretion and degradation of 20-hydroxyecdysone (ecdysone). In addition to its role in developmental regulation, increasing evidence suggests that ecdysone is involved in innate immunity processes, such as phagocytosis and the induction of antimicrobial peptide (AMP) production. AMP regulation includes systemic responses as well as local responses at surface epithelia that contact with the external environment. At pupariation, Drosophila melanogaster increases dramatically the expression of three AMP genes, drosomycin (drs), drosomycin-like

The Venus Flytrap Dionaea muscipula Counts Prey-Induced Action Potentials to Induce Sodium Uptake.

Carnivorous plants, such as the Venus flytrap (Dionaea muscipula), depend on an animal diet when grown in nutrient-poor soils. When an insect visits the trap and tilts the mechanosensors on the inner surface, action potentials (APs) are fired. After a moving object elicits two APs, the trap snaps shut, encaging the victim. Panicking preys repeatedly touch the trigger hairs over the subsequent hours, leading to a hermetically closed trap, which via the gland-based endocrine system is flooded by a prey-decomposing acidic enzyme cocktail. Here, we asked the question as to how many times trigger h

Whole transcriptome responses among females of the filariasis and arbovirus vector mosquito Culex pipiens implicate TGF-β signaling and chromatin modification as key drivers of diapause induction.

Culex pipiens mosquitoes are important disease vectors inhabiting temperate zones, worldwide. The seasonal reduction in temperature and photoperiod accompanying late summer and early fall prompts female mosquitoes to enter diapause, a stage of developmental arrest and physiological conditioning that enhances survival during the winter months. To investigate the molecular mechanisms underlying diapause induction, we used custom whole transcriptome microarrays to identify differences in gene expression following exposure to nondiapause (long days, 25 °C) and diapause-inducing (short days, 18 °C)

Whiteflies glycosylate salicylic acid and secrete the conjugate via their honeydew.

During insect feeding, a complex interaction takes place at the feeding site, with plants deciphering molecular information associated with the feeding herbivore, resulting in the upregulation of the appropriate defenses, and the herbivore avoiding or preventing these defenses from taking effect. Whiteflies can feed on plants without causing significant damage to mesophyll cells, making their detection extra challenging for the plant. However, whiteflies secrete honeydew that ends up on the plant surface at the feeding site and on distal plant parts below the feeding site. We reasoned that thi

Genome-wide identification of neuropeptides from Plutella xylostella (Lepidoptera: Plutellidae) and elucidating the essential roles of adipokinetic hormone in lipid mobilisation and food intake.

The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a major pest of cruciferous crops and has developed high levels of resistance to many widely used insecticides. Neuropeptides are involved in regulating essential insect biological processes, which could be considered potential target genes for pest control. In this study, a total of 45 neuropeptide precursor genes were identified in the genome of P. xylostella, including two adipokinetic hormone genes (PxAKH1 and PxAKH2). The qPCR analysis showed that PxAKH genes were highly expressed in third- and fourth-instar lar

Repeated exposure of bumblebee workers to field-realistic doses of di-2-ethylhexyl and di-n-butyl phthalates lowers worker survival and induces obesity-related phenotypes in workers and brood.

Wild bees, like many other insects, are declining. Multiple drivers have been identified, but the potential effects of endocrine disruptors such as phthalates, remain poorly studied. Yet, di-2-ethylhexyl phthalate (DEHP) and di-n-butyl phthalate (DnBP), two of the most common phthalates that pervade the environment, are known to affect key hormonal and metabolic pathways in invertebrates even at field realistic doses. These ubiquitous pollutants are semi-volatile compounds used to increase the flexibility of plastic materials, or as solvents for certain detergents, pesticides, and other produc
