Insects experience a wide array of chemical pressures from plant allelochemicals and pesticides and have developed several effective counterstrategies to cope with such toxins. Among these, cytochrome P450 monooxygenases are crucial in plant-insect interactions. Flavin-dependent monooxygenases (FMOs) seem not to play a central role in xenobiotic detoxification in insects, in contrast to mammals. However, the previously identified senecionine N-oxygenase of the arctiid moth Tyria jacobaeae (Lepidoptera) indicates that FMOs have been recruited during the adaptation of this insect to plants that

Alternative splicing (AS) is a major source of protein diversity in eukaryotes, but less is known about its evolution compared to gene duplication (GD). How AS and GD interact is also largely understudied. By constructing the evolutionary trajectory of the serpin gene PpSerpin-1 (Pteromalus puparum serpin 1) in parasitoids and other insects, we found that both AS and GD jointly contribute to serpin protein diversity. These two processes are negatively correlated and show divergent features in both protein and regulatory sequences. Parasitoid wasps exhibit higher numbers of serpin protein/domai

The white-backed planthopper (Sogatella furcifera) serves as the vector for the southern rice black-streaked dwarf virus (SRBSDV), with varying transmission efficiencies across populations. This research identifies Wolbachia, a common insect symbiont, as a key facilitator in breaching the salivary gland barrier for SRBSDV, revealing a mechanism by which the virus exploits an insect symbiont for transmission. Through field and laboratory investigations, it was observed that high-transmission (HT) planthopper populations contained high levels of Wolbachia, while low-transmission (LT) populations

A new anti-insect neurotoxin, AaH IT4, has been isolated from the venom of the North African scorpion Androctonus australis Hector. This polypeptide has a toxic effect on insects and mammals and is capable of competing with anti-insect scorpion toxins for binding to the sodium channel of insects; it also modulates the binding of alpha-type and beta-type anti-mammal scorpion toxins to the mammal sodium channel. This is the first report of a scorpion toxin able to exhibit these three kinds of activity. The molecule is composed of 65 amino acid residues and lacks methionine and, more unexpectedly

During Leishmania transmission sand flies inoculate parasites and saliva into the skin of vertebrates. Saliva has anti-haemostatic and anti-inflammatory activities that evolved to facilitate bloodfeeding, but also modulate the host's immune responses. Sand fly salivary proteins have been extensively studied, but the nature and biological roles of protein-linked glycans remain overlooked. Here, we characterised the profile of N-glycans from the salivary glycoproteins of Lutzomyia longipalpis, vector of visceral leishmaniasis in the Americas. In silico predictions suggest half of Lu. longipalpis

Edible insects, exemplified by honeybee pupa, represent a sustainable protein source but are associated with undercharacterized allergenic risks. This study innovatively addresses this gap by targeting honeybee pupa-derived Profilin, a cross-reactive pan-allergen, through conjugation with caffeic acid phenethyl ester (CAPE), a bioactive polyphenol derived from propolis. Structural and biochemical analyses revealed that CAPE-induced covalent polymerization and conformational remodeling (characterized by a shift from α-helix dominance to β-sheet dominance, accompanied by tertiary structural pert

Many insects feed on blood or tissue from mammalian hosts. One potential strategy for the control of these insects is to vaccinate the host with antigens derived from the insect. The larvae of the fly Lucilia cuprina feed on ovine tissue and tissue fluids causing a cutaneous myiasis associated with considerable host morbidity and mortality. A candidate vaccine antigen, peritrophin 95, was purified from the peritrophic membrane, which lines the gut of these larvae. Serum from sheep vaccinated with peritrophin 95 inhibited growth of first-instar L. cuprina larvae that fed on this serum. Growth i

The molecular dialogue between rice and the brown planthopper (BPH; Nilaparvata lugens) is mediated by insect salivary effectors, yet the molecular mechanisms underlying plant perception and immune activation remain largely unclear. Here, we characterized the BPH salivary protein NlSP2, which is secreted into rice tissues during feeding. Silencing of NlSP2 in BPH impairs salivary sheath formation, feeding ability, and survival, whereas its ectopic expression in rice enhances resistance to BPH via MAPK activation, H2O2 accumulation, and induction of defense-related genes. We identify the rice c

Bacillus thuringiensis (Bt) is widely used for the control of agricultural insect pests; however, trace residues of Bt may induce sublethal effects in insects. In this study, Bombyx mori was used as a model insect to investigate the mechanisms underlying midgut injury and tissue apoptosis induced by continuous exposure to a low concentration of Bt. The results showed that prolonged exposure to a low concentration of Bt progressively worsened structural damage to midgut epithelial cells, accompanied by chromatin condensation. Moreover, the apoptotic rate of midgut cells was significantly higher

Saliva plays important roles in insect feeding, but its roles in insect reproduction were rarely reported. Here we reported that the knockdown of a salivary gland-specific gene NlG14 disrupted the reproduction through inhibiting the ovulation of the brown planthopper (BPH), Nilaparvata lugens (Stål), one of the most devastating rice pests in Asia. NlG14 knockdown caused the displacement of the lateral oviduct secreted components (LOSC), leading to the ovulation disorder and the accumulation of mature eggs in the ovary. The RNAi-treated females laid much less eggs than their control counterpart

BACKGROUND: The neurotoxin peptide ω-ACTX-Hv1a, fused to the carrier molecule GNA, presents potential for insect control as a biopesticide, being orally toxic to insect pests from different orders. However, thorough evaluation is required to assure its safety towards non-target invertebrates. Effects of this novel biopesticide on the parasitoid Eulophus pennicornis via its host Lacanobia oleracea are presented.
RESULTS: Hv1a/GNA did not cause mortality when injected or fed to fifth-stage L. oleracea, but caused up to 39% reduction in mean larval weight (P < 0.05) and increased developmental t

Specific respiration rate ( [Formula: see text]) is a key parameter to understand cell metabolism and physiological state, providing useful information for process supervision and control. In this work, we cultivated different insect cells in a very controlled environment, being able to measure [Formula: see text]. Spodoptera frugiperda (Sf9) cells have been used through virus infection as host for foreign protein expression and bioinsecticide production. Transfected Drosophila melanogaster (S2) cells can be used to produce different proteins. The objective of this work is to investigate respi

Prenylated proteins play key roles in several human diseases including cancer, atherosclerosis and Alzheimer's disease. KRAS4b, which is frequently mutated in pancreatic, colon and lung cancers, is processed by farnesylation, proteolytic cleavage and carboxymethylation at the C-terminus. Plasma membrane localization of KRAS4b requires this processing as does KRAS4b-dependent RAF kinase activation. Previous attempts to produce modified KRAS have relied on protein engineering approaches or in vitro farnesylation of bacterially expressed KRAS protein. The proteins produced by these methods do not

All kinetoplastids contain membrane-bound microbodies known as glycosomes, in which several metabolic pathways including part of glycolysis are compartmentalized. Peroxin 2 is essential for the import of many proteins into the microbodies of yeasts and mammals. The PEX2 gene of Trypanosoma brucei was identified and its expression was silenced by means of tetracycline-inducible RNA interference. Bloodstream-form trypanosomes, which rely exclusively on glycolysis for ATP generation, died rapidly upon PEX2 depletion. Insect-form (procyclic) trypanosomes do not rely solely on glycolysis for ATP sy

Human malaria, a major public health burden in tropical and subtropical countries, is transmitted exclusively by the bite of a female Anopheles mosquito. Malaria control strategies aimed at inducing sexual sterility in natural vector populations are an attractive alternative to the use of insecticides. However, despite their importance as disease vectors, limited information is available on the molecular mechanisms regulating fertility in Anopheles mosquitoes. In the major malaria vector, An. gambiae, the full complement of sperm and seminal fluid required for a female's lifelong egg productio

INTRODUCTION: Mexican leaf-cutter ants (Atta mexicana), also known as chicatana ants, have been consumed in Mexico since pre-Columbian times as a seasonal food commodity and delicacy. Most studies on this edible insect have focused on its nutritional value as a protein source. Because of its proximate composition, this insect can also be considered a source of other important macro- and micronutrients. However, due to its geographic location and collection practices, food safety risks may be present.
METHODS: In this study, chicatana ants were collected in Huatusco, Veracruz, Mexico, and anal

Insect-transmitted trypanosomatid parasite infections cause life-threatening neglected tropical diseases (NTDs), including African sleeping sickness, Chagas disease and leishmaniasis. In these parasites, glycosomes are unique organelles that are essential for the parasite survival. Proper biogenesis of glycosomes is crucial to ensure correct compartmentation of the glycosomal metabolism. Genetic or chemical disruption of the glycosome biogenesis leads to a mislocalization of the glycosomal enzymes into the cytosol, which results in toxicity to the parasites. Here, we describe a detailed protoc

Rapidly expanding insect farming industries worldwide are raising concerns about introducing contaminants into food chains. One of the dominant species in this new bioindustry, the black soldier fly (Hermetia illucens), offers diverse benefits ranging from protein production to waste management, biofuel, and pharmaceutical applications. Recent studies have highlighted the ability of insects to accumulate metals; however, knowledge of their ability to eliminate contaminants is needed. Similarly, information regarding the accumulation of polycyclic aromatic hydrocarbons (PAHs) is lacking. This s

Arthropod-borne viruses cause major global health burdens, yet few vaccines or antivirals exist. Targeting host factors required for viral replication offers a promising approach. The DEAD-box RNA helicase eIF4A, a core component of the translation initiation complex eIF4F, unwinds structured 5' UTRs and is therefore critical also for the translation of many viral RNAs. The compound classes rocaglates and pateamines are potent eIF4A inhibitors in mammalian cells. Here we show that the natural rocaglate silvestrol strongly inhibits Rift Valley fever virus replication (RVFV) in human cells witho
