Interplay between proteostasis pathways and innate immune responses in Caenorhabditis elegans.

Microbial pathogens frequently manipulate host protein homeostasis to undermine immunity by targeting protein synthesis, folding, trafficking, and degradation. Conversely, effective immune responses themselves impose substantial proteostatic demands, as the rapid production of antimicrobial effectors increases the burden on cellular quality-control systems. This bidirectional pressure has likely driven the evolution of surveillance mechanisms that sense disruptions in protein homeostasis as indicators of infection. Using Caenorhabditis elegans as a genetically tractable model, recent studies h

Feeding Systems and the Gut Microbiome: Gut-Brain Interactions With Relevance to Psychiatric Conditions.

BACKGROUND: Physical and mental health is dependent on the environment, and feeding is a prime example of this environmental exchange. While the hypothalamus controls both feeding behavior and the stress response, the integration of the neural control centers and the peripheral gut allows for disruption in the gastrointestinal systems and dysfunctional communication to the brain. OBJECTIVE: The purpose of this review is to familiarize clinicians with the physiology controlling feeding behavior and its implications for psychiatric conditions, such as anorexia nervosa and depression. Growing un

Endometriosis and chronic pain syndrome: The role of stress mechanisms.

Patients with endometriosis report increased levels of perceived stress, which correlates with disease severity and pain intensity, creating feedback loops that perpetuate the cycle of endometriosis inflammation, chronic pain syndrome, and pathological stress response. Stress and chronic pain form an integrated biopsychosocial system in which neuroendocrine dysregulation, central sensitization, neuroinflammation, large-scale network reorganization, and maladaptive threat learning mutually reinforce each other. Sex differences, developmental experiences, and resilience factors modulate this int

Macrophage NRF1 promotes mitochondrial protein turnover via the ubiquitin proteasome system to limit mitochondrial stress and inflammation.

Macrophage elaboration of inflammatory responses is dynamically regulated, shifting from acute induction to delayed suppression during the c … l proteins accumulate to trigger severe mitochondrial stress. Such mitochondrial stress engages the integrated stress response-ATF4 axis, which limits mitochondrial translation to attenuate mitochondrial stress but amplifies inflammatory responses to augment susceptibility to septic shock. Therefore, NRF1 mediates a dynamic regulation of mitochondrial proteostasis in inflam …

Cytoprotection by pre-emptive conditional phosphorylation of translation initiation factor 2.

Transient phosphorylation of the alpha-subunit of translation initiation factor 2 (eIF2alpha) represses translation and activates select gene expression under diverse stressful conditions. Defects in the eIF2alpha phosphorylation-dependent integrated stress response impair resistance to accumulation of malfolded proteins in the endoplasmic reticulum (ER stress), to oxidative stress and to nutrient deprivations. To study the hypothesized protective role of eIF2alpha phosphorylation in isolation of parallel stress signaling pathways, we fused the kinase domain of pancreatic endoplasmic reticulum…

Acute hypernatremia dampens stress-induced enhancement of long-term potentiation in the dentate gyrus of rat hippocampus.

Stress often occurs within the context of homeostatic threat, requiring integration of physiological and psychological demands to trigger appropriate behavioral, autonomic and endocrine responses. However, the neural mechanism underlying stress integration remains elusive. Using an acute hypernatremic challenge (2.0M NaCl subcutaneous), we assessed whether physical state may affect subsequent responsiveness to psychogenic stressors. We found that experienced forced swimming (FS, 15min in 25°C), a model of psychogenic stress, enhanced long-term potentiation (LTP) induction in the dentate gyrus

DOF Transcription Factors: Regulatory Hubs Coordinating Plant Growth, Development and Stress Tolerance.

Under global climate warming, plants evolve unique growth and developmental strategies to cope with frequent abiotic stresses. The plant-specific DNA-BINDING WITH ONE FINGER (DOF) transcription factors (TFs) originated in green algae and are phylogenetically divided into four subfamilies (A, B, C, D), governing plant growth development and stress adaptation. Subfamily B mainly regulates vegetative growth, subfamily C is predominantly involved in seed development and vegetative growth, and subfamily D1 (CYCLING DOF FACTOR, CDF) participates extensively in vegetative growth, flowering, and stres

Mov10 mitigates hematopoietic stem cell exhaustion under stress by modulating the Camp-mediated inflammatory pathway.

The integrity of hematopoietic stem cell (HSC) function is crucial for robust hematopoietic regeneration following stress. Inflammatory responses are pivotal drivers of HSC stress response, yet the precise modulation of inflammatory pathways remains incompletely defined. In this study, we identify the RNA helicase Mov10 as a negative regulator of stress-induced inflammatory pathways in HSC. Our study indicates that Mov10, which is critically required for HSC maintenance, is highly expressed in HSC, and its loss adversely affects HSC fitness and survival during hematopoietic stress induced by b

Identification of myokines associated with the pathological stress response in the mdx mouse model of Duchenne muscular dystrophy.

PURPOSE: Skeletal muscle constitutes 30-40% of total body mass and is now considered an endocrine organ, given its secretion of a variety proteins, metabolites, and cytokines. We have previously shown that the absence of dystrophin in skeletal muscle contributes to lethal systemic stress pathology in the mdx mouse model of Duchenne muscular dystrophy through a mechanism that remains to be identified. Here we searched for secreted protein signaling factors, or myokines, released from dystrophin-deficient skeletal muscle that influence the organism-wide integrated stress response. We performed s…

Plant chloroplast stress response: insights from mass spectrometry metabolites analysis.

Plant chloroplasts produce excess reactive oxygen species (ROS) during photosynthesis, particularly under biotic and abiotic stress conditions. These adverse environmental stresses lead to significant alterations in various cellular components, especially within the chloroplast, which serves as a key stress-sensor organelle. The stress response of chloroplasts can trigger plastid-to-nucleus retrograde signaling and enhance the biosynthesis of biologically active compounds and phytohormones, which are mechanisms that aid plants in acclimating to environmental stress. While ROS act as signaling

Rainbow trout integrated response after recovery from short-term acute hypoxia.

Overcoming a stress situation, such as hypoxia episodes, which involve an allostatic load, will depend on the ability of fish to modulate physiological and biochemical systems to maintain homeostasis. The aim of the study was to determine the integrated stress response after acute hypoxia of the rainbow trout considering the different elements and areas of the stress response: systemic and mucosal, local and global, and from the systemic hypothalamic-pituitary-interrenal axis to skin mucosa. For this purpose, trout were subjected to acute hypoxia (dissolved O2 down to 2 mg/L) for 1 h and then …

Neuroimmune integration in chronic stress: mechanisms of TRPV1-CB2 crosstalk, inflammation, and neural resilience.

Under the relentless demands of contemporary life, stress orchestrates a sophisticated neurochemical cascade, engaging the hypothalamic-pituitary-adrenal axis and modulating key neurotransmitters-norepinephrine, serotonin, and dopamine-as though forcing a finely tuned thermostat beyond its limits. While acute stress can transiently sharpen adaptive responses, chronic exposure exacts an escalating allostatic burden, gradually compromising synaptic integrity and heightening vulnerability to psychiatric and neurological disorders. Guided by the Neuroimmune Integration Model, this review integrate

A paradigm shift in oxygen sensing with a twist in the tale!

AMP-activated protein kinase (AMPK) is pivotal to metabolic homoeostasis in eukaryotes, serving as a critical energy sensor. Increased AMPK activity during oxygen deprivation (hypoxia) protects against potentially catastrophic deficits in ATP supply. Although the nervous system circuitry for elaboration of the complex cardiorespiratory response to hypoxia has been understood in some detail for many decades, there is continued and considerable interest in the molecular machinery underpinning the mechanism(s) of oxygen sensing. In this issue of the Biochemical Journal, Evans et al. [(2016) Bioch

Rats Selected for Different Nervous Excitability: Long-Term Emotional-Painful Stress Affects the Dynamics of DNA Damage in Cells of Several Brain Areas.

The maintenance of genome stability is critical for health, but during individual ontogenesis, different stressors affect DNA integrity, which can lead to functional and/or structural changes in the cells of target organs. In the nervous system, cell genome destabilization is associated with different neurological and psychiatric diseases, but experiments in vivo, where a link between stress and DNA instability has been demonstrated, are relatively rare. Here, we use rat strains selected for the contrast excitability of the tibialis nerve (n. tibialis) and nonselected Wistar rats to investigat

Exposure-based treatment for co-occurring posttraumatic stress disorder and obsessive compulsive disorder in veterans: The feasibility of massed models.

Exposure-based therapies are widely accepted as the gold-standard intervention for both obsessive compulsive disorder (OCD) and posttraumatic stress disorder (PTSD). Despite their co-occurrence, little research has explored exposure-based treatment in individuals who experience both OCD and PTSD. At an academic medical center mental health program, four different treatment approaches combining exposure and response prevention (ERP) and prolonged exposure (PE) therapies were piloted for veterans with co-occurring OCD and PTSD. We present each approach with a brief case study. Two sequential mod

A review of flavonoids at the crossroads of plant defense: integrating biotic and abiotic stress tolerance through AI- and CRISPR/Cas-guided metabolic reprogramming.

Flavonoids are multifunctional phenylpropanoid-derived metabolites that occupy a central position in plant adaptation to environmental stress. Beyond their established roles in antioxidant protection, they contribute to defense against pathogens and herbivores, signaling processes, and physiological acclimation to adverse environmental conditions. Although flavonoid responses to individual biotic or abiotic stresses have been extensively investigated, considerably less attention has been given to how flavonoid-associated regulatory networks function when multiple stresses occur simultaneously.
