The effect of different milk pretreatment methods on microbiome community development during Herrgårds cheese production and ripening.

One of the biggest challenges for dairy producers is the substantial variability in final product properties caused by changes in the produc … ing nine high-quality metagenome-assembled genomes. Our analysis revealed that a specific strain of Lactococcus lactis dominates the process, while other minor bacterial species persist at very low abundances (<1%), contributing non-negligibly to product properties. Notably, we detected DNA from Clostridium tyrobutyricum, a known bacterium whose heat-resistan …

The unconventional Xer recombination machinery of Streptococci/Lactococci.

Homologous recombination between circular sister chromosomes during DNA replication in bacteria can generate chromosome dimers that must be … r direct protein-protein interaction, which we demonstrated to be located at the division septum of Lactococcus lactis. Acquisition of the XerS recombination module can be considered as a landmark of the separation of Streptococci/Lactococci from other firmicutes and support the view that Xer recombination is a conserved cellular function in bacteria, but tha …

Metataxonomic insights into lactic acid bacteria diversity in artisanal coalho cheese from the Caatinga biome.

Artisanal raw-milk cheeses are complex microbial ecosystems that reflect local environments through spontaneous fermentation driven by lacti … 00 bp). The microbial community was dominated by LAB, particularly Enterococcus (~25%), followed by Lactococcus, Streptococcus, and Leuconostoc. The predominance of these genera reflects the spontaneous fermentation process typical of artisanal raw-milk cheeses and highlights their role in acidification, flavor development, and microbial stability. The microb …

Bacteriocin presence enhances phage-related risks in dairy fermentations.

Bacteriophages infecting dairy starter cultures are a serious threat in the dairy industry leading to failed or low-quality milk fermentations. The aim of this work was to assess if the presence of bacteriocins, either intentionally added or produced in situ during milk fermentation, increase the risk of fermentation failure in the event of phage contamination. Bacteriocin-phage interactions (BaPIs) were determined in vitro for nisin, a bacteriocin commonly used as a food biopreservative. Nisin increased the plaque size for the lactococcal phages P2 and bIL170. However, bIL170 burst size and l

Unveiling the volatilome of PGI Castellano cheese: Impact of ripening time and wild lactic-acid bacteria cultures on the volatile composition.

The influence of ripening and lactic acid bacteria consortia on the volatile profile of five Castellano cheese batches was evaluated using HS-SPME coupled with GC × GC-ToFMS. A control batch was produced with a commercial starter comprising two lactococcal strains, while four experimental batches combined this starter with autochthonous strains of Lactiplantibacillus plantarum, Lacticaseibacillus paracasei, and Leuconostoc mesenteroides. In one batch, the commercial acidifying culture was replaced with an autochthonous Lactococcus lactis. A non-targeted metabolomic approach detected 280 volati…

Transcriptome Analysis Reveals the Mechanism by which Probiotic Alleviate the Response of Juvenile American Shad (Alosa sapidissima) to High Temperatures.

Probiotics show great potential in mitigating the impacts of temperature stress on fish. American shad (Alosa sapidissima), a species highly sensitive to high temperatures, served as an ideal model for studying fish adaptation to climate change. This study integrated biochemical and hepatic transcriptomic analyses to demonstrate that the probiotic Lactococcus lactis L103 alleviated thermal stress via a regulatory network. The findings revealed that probiotic-mediated upregulation of irf1b and nfil3-2 suppressed pro-inflammatory responses, as evidenced by significantly reduced AST and ALT activ…

After a century of nisin research - where are we now?

It is almost a century since nisin was discovered in fermented milk cultures, coincidentally in the same year that penicillin was first described. Over the last 100 years this small, highly modified pentacyclic peptide has not only found success in the food industry as a preservative but has also served as the paradigm for our understanding of the genetic organization, expression, and regulation of genes involved in lantibiotic biosynthesis-one of the few cases of extensive post-translation modification in prokaryotes. Recent developments in understanding the complex biosynthesis of nisin have

The biological aspects of oral pathogenicity.

The bacteria in the oral region tend to localize in the form of plaque deposits on tooth surfaces and in gingival crevices. The "metabolic nature" of this complex flora focuses on the reversible interactions between the plaque flora and their environmental modifications. The environmental circumstances will influence the range of expression in situ for the plaque (oral) organisms. Then, it is suggested that the potential for caries induction or periodontal disease may develop when conditions approaching extremes of environment result in altered metabolism and antigenicity of particular oral or

A short update on new approaches to celiac disease.

Celiac disease is an autoimmune enteropathy of the small intestine, related to gluten intolerance occurring in genetically predisposed patients. Currently, the only available treatment is a lifelong gluten-free diet. However, the total avoidance of gluten is difficult and poses a challenge to patients, nutritionists and treating physicians. For this reason, scientists have developed in recent years new therapeutic approaches complementary to dietary treatment, such as modification of gluten to make gliadin non-toxic, reduction of the inflammatory response with elafin and Lactococcus Lactis, de…

In Vivo Assembly of Artificial Metalloenzymes and Application in Whole-Cell Biocatalysis*.

We report the supramolecular assembly of artificial metalloenzymes (ArMs), based on the Lactococcal multidrug resistance regulator (LmrR) and an exogeneous copper(II)-phenanthroline complex, in the cytoplasm of E. coli cells. A combination of catalysis, cell-fractionation, and inhibitor experiments, supplemented with in-cell solid-state NMR spectroscopy, confirmed the in-cell assembly. The ArM-containing whole cells were active in the catalysis of the enantioselective Friedel-Crafts alkylation of indoles and the Diels-Alder reaction of azachalcone with cyclopentadiene. Directed evolution resul

The membrane topology of immunity proteins for the two-peptide bacteriocins carnobacteriocin XY, lactococcin G, and lactococcin MN shows structural diversity.

The two-peptide bacteriocins produced by Gram-positive bacteria require two different peptides, present in equimolar amounts, to elicit optimal antimicrobial activity. Producer organisms are protected from their bacteriocin by a dedicated immunity protein. The immunity proteins for two-peptide bacteriocins contain putative transmembrane domains (TMDs) and might therefore be associated with the membrane. The immunity protein CbnZ for the two-peptide bacteriocin carnobacteriocin XY (CbnXY) was identified by heterologously expressing the cbnZ gene in sensitive host strains. Using protein topology
