[Precocious maturation of the testis associated with excessive secretion of estradiol and testosterone by Leydig cells].

We present the quantitative description of spermatogenesis in a 4.5-year-old boy with precocious puberty, where Leydig cell hyperplasia was associated with excessive secretion of testosterone (T), but predominantly with estradiol (E). The results were compared with data obtained from an age-matched group and adult men without hormonal abnormalities. We showed, that excessive secretion of T and E with relative deficiency of FSH is sufficient to induce testicular tubule maturation and qualitatively complete spermatogenesis, although with a poorer quantitative aspect.

The direct effect of hepatic peroxisome proliferators on rat Leydig cell function in vitro.

A review of the literature indicates that some compounds which produce hepatic peroxisome proliferation in rats also appear to produce Leydig cell adenomas, and some also affect the serum concentrations of testosterone and estradiol. Previous studies with the peroxisome proliferator ammonium perfluorooctanoate showed a direct effect on Leydig cells to alter steroidogenesis. It was therefore proposed that peroxisome proliferators in general may directly affect Leydig cell function to produce Leydig cell tumors by some undetermined mechanism. The present study investigated whether the following …

Compound heterozygous mutations of the luteinizing hormone receptor gene in Leydig cell hypoplasia.

The human LH receptor (hLHR) is a member of the G protein-coupled receptors characterized by the presence of seven-transmembrane (TM) helices. Inactivating mutations of the hLHR lead to Leydig cell hypoplasia (LCH), a form of male pseudohermaphroditism resulting from the failure of fetal testicular Leydig cell differentiation. We have identified three mutations of the hLHR in a patient with LCH: deletion of exon 8 (delta Exon 8), A872G transition resulting in Asn291Ser substitution in the extracellular domain, and C1847A transversion resulting in Ser616Tyr substitution in the seventh TM helix.

Leydig cell function after experimental testicular torsion despite loss of spermatogenesis.

Little is known about specific testicular cell responses to periods of testicular torsion. In particular, the steroidogenic capacity of Leydig cells in the post-torsion testis is unknown. Male Sprague-Dawley rats (450-550 g) underwent no torsion (control) or a 720 degrees unilateral testicular torsion for either 0 (sham), or 1 or 2 Such torsions have previously been shown to cause progressive damage to the rat testis. One, 15, or 30 days after torsion repair all animals (n = 5-10/group) were prepared for testicular venipuncture and intravenous infusion of ovine luteinizing hormone (LH) via the…

Ultrastructural and morphometrical analyses of Leydig and Sertoli cells in the testes of rats with hereditary polydactylism.

Rats of the polydactylous (PD) strain carry an autosomal recessive gene pd that causes polydactylism in homozygotes (pd/pd). Male homozygotes are sterile owing to an abnormality of spermatogenesis. In the present study, Leydig and Sertoli cells of 12-week-old pd/pd male rats were examined for ultrastructural alterations in an attempt to clarify the cause of the abnormal spermatogenesis. The relative volumes of the organelles were also determined with morphometry. Phenotypically normal pd/+ males served as controls. No morphological or morphometrical abnormalities were noted in the Leydig cells…

Testicular function and Leydig cell ultrastructure in long-term bilaterally cryptorchid rams.

This study was conducted to investigate the effects of bilateral cryptorchidism induced in adult rams on testicular function and Leydig cell ultrastructure. The results indicated that long-term bilateral cryptorchidism resulted in decreased testicular size, degeneration of seminiferous tubules, elevated serum LH levels, maintenance of normal testosterone concentrations in peripheral and spermatic vein serum, impairment of the magnitude and duration of androgen response to exogenous luteinizing hormone (LH), a 13-fold reduction in total number of Leydig cells/paired testes, and a 3-fold hypertr…

Leydig-cell tumor of the ovary: visualization using 131I-19-iodocholesterol scintigraphy.

A Leydig (Hilus)-cell tumor of the ovary was diagnosed in a 54-year-old woman with severe hirsutism and virilization. This case is unique in that it was diagnosed in a postmenopausal female 24 years after ovarian irradiation for sterilization, and the tumor was successfully localized preoperatively using 131I-19-iodocholesterol scintigraphy. This nuclear scanning technique may be of use in documenting the biologic activity of ovarian masses and in localizing functioning ovarian tumors which are too small to palpate or to localize by other means.

Sertoli--Leydig cell tumor with endometroid differentiation. Case report and discussion of histogenesis.

A case of Sertoli-Leydig cell tumor with endometrioid differentiations is reported. The tumor was associated with hirsutism and androgenic excess which normalized following surgical removal. On light-microscopic study, the tumor resembled a well-differentiated endometrioid tumor with luteinized stroma. With further study, the interstitial stromal cell were recognized as Leydig cells, containing scattered Reinke crystals. Tubules and solid cords reminiscent of sex cord elements were also recognized. The dilated glandular elements showed scant apical and luminal positivity with the PAS stain fol…

The effect of growth hormone on the Leydig cell response to chorionic gonadotrophin in boys with hypopituitarism.

Eleven boys with growth hormone (hGH) deficiency received human chorionic gonadotrophin (hCG) stimulation tests for the assessment of Leydig cell function before, during, and after 1 year of treatment with somatotrophin. Two patients entered puberty during the course of the study protocol. Analysis of the data in nine prepubertal boys revealed an augmentation of testosterone (T) responses to hCG in the presence of hGH. In six of these individuals in whom dihydrotestosterone (DHT) was determined, a similar augmentation in responsiveness of this steroid was found in the presence of hGH. Three pr

Temporal changes in rat Leydig cell function after the induction of bilateral cryptorchidism.

Adult rats were made bilaterally cryptorchid and studied at intervals of 3, 7, 14 or 21 days to study temporal changes in Leydig cell function. Serum FSH and LH levels were measured and the cross-sectional area of the Leydig cells assessed by morphometry. The function of the Leydig cells was judged by the binding of 125I-labelled hCG to testicular tissue in vitro and the testosterone response of the testis to hCG stimulation in vitro. By 3 days after cryptorchidism, the binding of labelled hCG to testicular tissue was significantly decreased compared to that of controls, but the testes were ab…

Autonomous cortisol secretion by a metastatic Leydig cell carcinoma associated with Klinefelter's syndrome.

We present the case of a 39-year-old man with Klinefelter's syndrome and a metastatic Leydig cell carcinoma in whom autonomous cortisol production induced by the interstitial cell tumor was found. Apart from the Cushing's syndrome the endocrine activity of the tumor was demonstrated by the secretion of estradiol, estrone, alkaline phosphatase, and testosterone. This is, to our knowledge, the first description of a Cushing's syndrome not caused via ACTH production but directly induced by ectopic steroid production. While being resistent to chemotherapy and radiation, the tumor responded favorab

Leydig cell density and function and their relation to gonadotropins in infertile oligospermic men with varicocele.

Circulating gonadotropin and testosterone levels in infertile men with varicocele and sperm counts of less than 10(7)/ml were correlated with testicular Leydig cell density and in vitro testosterone synthesis. A significant correlation was found between Leydig cell density and both plasma testosterone levels and in vitro testosterone synthesis. Luteinizing hormone (LH) levels paralleled the in vitro testosterone synthesis. Although in the majority of patients, in vitro testosterone synthesis and Leydig cell density were low, plasma testosterone and LH levels were within the normal range. The o

Molecular mechanism of testicular oxidative stress-induced ferroptosis in Leydig cells caused by cryptorchidism in yak.

Cryptorchidism is a prevalent reproductive disorder triggering severe testicular damage and male infertility. However, the underlying molecular mechanism in plateau yaks remains largely unclear. This study explored the pathogenesis of androgen deficiency and Leydig cell ferroptosis induced by yak cryptorchidism, and illuminated the relevant regulatory pathways. Testicular specimens were collected from healthy and cryptorchid yaks, the Leydig cells were isolated, purified and cultured in vitro combined with the histopathological staining, immunofluorescence, Western blotting and ELISA were cond…

Immunohistochemical evaluation of Sertoli and Leydig cells of scotch cat testis (Felis catus) at diverse ages.

BACKGROUND: The testicles are located in a muscular, skin-covered, highly pigmented sac called the scrotum. The testicles produce both sperm and androgens, such as testosterone. AIM: The current study aimed to evaluate the immunohistochemical characteristics of the testis in three age groups (kitten, adult, and senior) of Scottish cat breeds (Felis catus) during the postnatal period, focusing on changes in Sertoli cells and Leydig cells. These two cell types play essential roles in male reproductive function, as Sertoli cells provide structural and metabolic support for spermatogenesis, while…
