A biologically based pharmacodynamic model for lipid peroxidation stimulated by trichloroethylene in vitro.

It is often necessary for chemical risk assessment to determine a quantitative relationship between the internal dose of a chemical and its biological effect. The tool best suited for this purpose is a biologically based pharmacodynamic (BBPD) model. Such a BBPD model was developed previously (10) to simulate chemically induced lipid peroxidation, and it was experimentally calibrated in precision-cut mouse liver slices in vitro. The BBPD model simulated formation of lipid hydroperoxides and thiobarbituric acid reactive substances (TBARS) over time and was originally calibrated with different c…

Lipid peroxidation and antioxidant system in the blood of cancerous patients with metastasis.

Free-radical-mediated damages may play an important role during metastasis. To investigate their relevance in the metastatic process MDA levels, glutathione peroxidase (GPX) and superoxide dismutase (SOD) activities, and selenium, zinc and copper contents were determined in plasma and erythrocytes from 20 cancerous patients with metastasis and 30 age-matched controls. Significantly higher concentrations of MDA in plasma as well as in erythrocytes were found comparing to the control group. In both plasma and erythrocytes, GPX activity and selenium and zinc levels were significantly lower in pat

The clinical manifestation of the kwashiorkor syndrome is related to increased lipid peroxidation.

Along with the onset of severe kwashiorkor symptoms, a 20-month-old child showed biochemical signs of markedly increased lipid peroxidation, with a decrease of plasma antioxidants and decreased proportions of polyunsaturated fatty acids in plasma and red cell phospholipids. Additionally, plasma concentrations of the lipid peroxidation products malondialdehyde and hexanal, as well as the urinary excretion of leukotriene E4, were found to be increased. All biochemical alterations normalized along with subsequent clinical improvement. These findings suggest that the extent of lipid peroxidation i…

Menadione-induced cell degeneration is related to lipid peroxidation in human cancer cells.

The role of lipid peroxidation, intracellular glutathione and Ca2+ concentration in menadione-mediated toxicity was investigated in human hepatoma cell lines, Hep G2 and Hep 3B, and in human leukemia cell lines, CCRF-CEM and MOLT-3. Incubation of these cells with 80 microM menadione at 37 degrees C resulted in depletion of intracellular glutathione, increased intracellular Ca2+, and increased lipid peroxidation, events leading to cell degeneration. The sensitivity of these cells to menadione, in order, was: Hep G2 cells > Hep 3B cells > CCRF-CEM cells and MOLT-3 cells. The extent of menadione-…

Lipoxidation products as biomarkers of oxidative damage to proteins during lipid peroxidation reactions.

Oxidative stress is implicated in the pathogenesis of numerous disease processes including diabetes mellitus, atherosclerosis, ischaemia reperfusion injury and rheumatoid arthritis. Chemical modification of amino acids in protein during lipid peroxidation results in the formation of lipoxidation products which may serve as indicators of oxidative stress in vivo. The focus of the studies described here was initially to identify chemical modifications of protein derived exclusively from lipids in order to assess the role of lipid peroxidative damage in the pathogenesis of disease. Malondialdehye…

[Lipid peroxidation and blood serum peroxidase activity in diseases of the hepatobiliary system].

Patients with hepatobiliary pathology showed an augmentation of peroxide oxidation of lipids (LP), its degree dependent upon the severity of the clinical course of the medical condition in question, being the highest in those patients with viral hepatitis, carcinoma of the head of pancreas, and portal cirrhosis of the liver. A parallel investigation into the blood serum peroxidase activity, the main indicator of the antioxidant activity, in the above patients, showed a noticeable decrease in PA activity. LP and blood serum PA measurements allow some judgement about the gravity of the course of

Vitamin E slows the rate of free radical-mediated lipid peroxidation in cells.

Much of what is known about the antioxidant mechanism of vitamin E has been learned from studies of lipid dispersions, solutions, or subcell … also altered the kinetics of ascorbate free radical formation. We conclude that vitamin E inhibits lipid peroxidation in cells by slowing the rate of lipid peroxidation; but with iron/ascorbate as the initiating system, vitamin E does not delay the onset of peroxidation. Of special interest is that these free radical peroxidation events parallel cell membrane damag …

Preventive effect of melatonin against cyanide-induced seizures and lipid peroxidation in mice.

Subcutaneous injection of potassium cyanide (6, 8, and 9 mg/kg) caused a severe tonic seizure in a dose-dependent manner. However, the incidence of seizures induced by potassium cyanide was significantly inhibited by preadministration of melatonin (20 mg/kg, s.c.) Lipid peroxidation in homogenates from whole brain of mice was significantly increased (39%, 106% and 132%, respectively) by the exposure to potassium cyanide (0.01, 0.05, and 0.1 mM). The potassium cyanide (0.1 mM)-induced lipid peroxidation was prevented by melatonin (0.1, 0.5, 1.0, or 2 mM) in a concentration-dependent manner. The…

Hemoglobin-induced lipid peroxidation in the retina: a possible mechanism for macular degeneration.

To investigate a possible link between subretinal hemorrhage and macular degeneration, oxyhemoglobin (HbO2) or methemoglobin (metHb) was incubated with retinal homogenate and unsaturated phospholipid peroxidation was monitored by (a) assay of thiobarbituric acid-reactive substances (TBARS), (b) luminescence originating from an energy transfer of lipid-degraded products to rose bengal, and (c) the decrease in composition of highly unsaturated fatty acids of phospholipids. TBARS formation and rose bengal luminescence in the case of metHb-induced lipid peroxidation were about 1.5 times greater th…

Role of lipid peroxidation in cadmium-induced impairment of the gastric mucosal barrier.

Peroxidative tissue damage has been reported to contribute to several pathological disorders. Despite high exposure to both exogenous and endogenous oxidant stress, the strong cell defence mechanism of the gastric mucosa protects mucosal epithelial cells against these noxious stimuli. However, some environmental factors involved in lipid peroxidation (such as cadmium), which disrupt gastric mucosal protection, may impair the mucosal barrier and facilitate the occurrence of gastric ulcers. In an experimental study to investigate this hypothesis, the level of cadmium-induced lipid peroxidation p…

Isoprostanes. Novel markers of endogenous lipid peroxidation and potential mediators of oxidant injury.

It was recently discovered that a series of structurally unique prostaglandin F2-like compounds (F2-isoprostanes) capable of exerting potent biological activity are produced in vivo in humans by a noncyclooxygenase mechanism involving free radical catalyzed peroxidation of arachidonic acid. Considerable evidence has been obtained suggesting that quantification of F2-isoprostanes represents an important advance in our ability to assess oxidant status in vivo in humans. This has allowed us to implicate oxidant stress in the pathogenesis of human disease-for example, the hepatorenal syndrome. In

Benzylamine antioxidants: relationship between structure, peroxyl radical scavenging, lipid peroxidation inhibition, and cytoprotection.

Three homologous series of 3,5-dialkoxy-4-hydroxybenzylamines were prepared and tested (1) as peroxyl radical scavengers in homogeneous aqueous solution, (2) as inhibitors of iron-dependent peroxidation of rabbit brain vesicular membrane lipids, and (3) as cytoprotective agents using primary cultures of rat hippocampal neurons exposed to hydrogen peroxide. The structural requirements for efficient radical trapping in homogeneous solution differed from those for effective lipid peroxidation inhibition: In homogeneous solution a kinetic preference existed for smaller, less sterically encumbered …

Increased cerebrospinal fluid protein and lipid peroxidation products in patients with cerebral malaria.

Membrane lipid peroxidation by reactive oxygen species leading to increased capillary permeability is considered an important event in the pathogenesis of severe malaria. A significant decrease in plasma albumin and increases in cerebrospinal fluid (CSF) protein and malondialdehyde (MDA) were observed in 73 patients with cerebral malaria, compared to values in 23 control patients. The greatest effect was noticed in the most severely ill patients. The ratio of CSF protein to plasma albumin was increased in the patients compared to the controls, and in fatal cases of cerebral malaria compared to…

The increase of membrane capacitance as a consequence of radiation-induced lipid peroxidation.

The electrical capacitance of lipid membranes may increase by up to 50% on exposure to ionizing radiation. This is the consequence of lipid peroxidation induced by primary or secondary radicals of water radiolysis in the presence of oxygen. The polar products of this process give rise to an increase of the dielectric constant of the membrane. This in turn leads to the increase of membrane capacitance observed. An important consequence of this phenomenon is the reduction of the inner energy barrier between the two membrane/water interfaces influencing the movement of charged particles across th…

Lipid peroxidation and oxidation of lignoceric acid in kidneys from thioridazine treated rats.

We have investigated lipid peroxidation and oxidation of lignoceric acid in response to oral thioridazine administration, to better understand the effects of phenothiazines, which are one of the more commonly used therapeutic agents. Measurements at different time intervals showed that levels of lipid peroxides in rat kidney were markedly decreased after thioridazine feeding, however, the oxidation of lignoceric acid was found to be elevated immediately after the start of thioridazine treatment. These biochemical changes were noted to be associated with mitochondrial proliferation and lipid ac…

Detection of lipid peroxidation in lung and in bronchoalveolar lavage cells and fluid.

Inhalation of toxic materials such as asbestos, silica, 100% oxygen, ozone, or nitrogen dioxide may lead to an increased production of reactive oxygen metabolites which may initiate lipid peroxidation. Measurement of lipid peroxidation in cells and fluid obtained by bronchoalveolar lavage (BAL), as well as in lung tissue, may aid in monitoring the development and extent of pulmonary damage after inhalation of a toxic substance. In this study, we employed a sensitive assay for detection of malondialdehyde (MDA), a breakdown product of lipid peroxidation. By separation of the adduct with thiobar…

Oxygen radicals, lipid peroxidation, and neutrophil infiltration after small-intestinal ischemia and reperfusion.

In this study the role of free radicals, lipid peroxidation, and neutrophil infiltration as mediators of ischemia and reperfusion-induced intestinal mucosal damage were investigated. We used a rat experimental model in which a ligated loop of the distal ileum was subjected to ischemia and reperfusion and the ensuing mucosal damage was assessed by means of lysosomal enzyme release and intestinal permeability measurements. We also determined the mucosal content of malondialdehyde, a lipid peroxidation product, and the mucosal activity of myeloperoxidase, a neutrophil granulocyte marker. Ischemia…

Correlation among lipid peroxidation, brain energy metabolism and brain oedema in cerebral ischaemia.

In order to study the involvement of lipid peroxidation in the generation of ischaemic cell damage, sequential measurements were made on the chemiluminescence, energy metabolism, water content and the volume of Na+ and K+ in the cerebral cortex using a rat model for severe cerebral ischaemia. It was found that the amount of chemiluminescence increased due to ischaemia and increased further following recirculation. In chemiluminescence spectral analysis, peaks were found at 480, 520-530, 570, 620-640, 680-700 nm. It is thought that these wavelengths reflect the release of energy due to the redu…
