Rapid Peptide Mapping of Monoclonal Antibodies with Direct Infusion Mass Spectrometry.

Peptide mapping is a critical tool for characterizing biotherapeutic proteins and is essential for the development of monoclonal antibody drugs. Here we describe a new direct infusion technology that streamlines peptide mapping data collection and analysis, accelerating the method by up to 100-fold. This method, which we term RaPiD-mAb-MS, combines high-throughput plate-based sample preparation with direct infusion mass spectrometry analysis. RaPiD-mAb-MS allows analysis of 96 samples within ∼ 1.5 to 2 hours, routinely achieves >95% sequence coverage, and has been successfully applied to 28 un…

Bioactive compounds in functional foods using mass spectrometry: A systematic review.

This systematic review provides a comprehensive, data-centric overview of bioactive compound discovery in functional foods using mass spectrometry (MS) between 2020 and 2025. Evaluating 250 foodomics studies, we identified over 2000 unique molecules across 14 food macro-categories and 11 molecular classes. Phenolic compounds, specifically ferulic acid, gallic acid, and rutin, emerged as the most pervasive markers, particularly in fruits and botanicals. Methodological analysis reveals that while LC-ESI-QTOF-MS/MS dominates untargeted discovery, significant analytical vulnerabilities remain, inc…

Forced degradation study of abacavir using the electrochemistry-mass spectrometry setup.

This article investigates the electrochemical oxidation of abacavir using a simple online setup, coupling an electrochemical flow cell with a mass spectrometer, as an alternative approach to forced oxidative degradation studies. Electrochemical oxidation of abacavir was performed on a glassy carbon electrode in only 3 min, using four different solvents: three containing 0.5 mol/L ammonium acetate, pH 7.0, with 0, 50, and 90% (v/v) methanol, and one containing 0.1 mol/L NaClO4 in pure methanol. A total of seven degradation products were observed, with detector signals in different ratios depend

MALDI versus DESI mass spectrometry imaging of lipids in atherosclerotic plaque.

Mass spectrometry imaging (MSI) is a powerful tool for detecting lipids in tissue sections, with matrix-assisted laser desorption/ionization (MALDI) and desorption electrospray ionization (DESI) as its key ionization techniques. In this study, we examine how MALDI compares with state-of-the-art DESI ionization in identifying lipids in heterogeneous samples, specifically atherosclerotic plaques. Carotid plaques (n = 4) from patients undergoing endarterectomy were snap-frozen, stored at -80°C, and then sectioned for MSI analysis and H&E staining. Measurements were conducted using a SYNAPT XS mas…

Mass spectrometry identifies tau C-terminal phosphorylation cluster during neuronal hyperexcitation.

Tau is a microtubule-associated protein implicated in Alzheimer's disease (AD) and other neurodegenerative disorders termed tauopathies. Pat … erexcitation promotes extracellular secretion and trans-synaptic propagation of tau. Using unbiased mass spectrometry, we identified a novel phosphorylation signature in the C-terminal domain of tau detectable only during neuronal hyperactivity in primary cultured rat hippocampal neurons. These sites influenced tau localization to dendrites as well as the size of e …

Mass Spectrometry-Based Proteomics of Epithelial Ovarian Cancers: A Clinical Perspective.

Increasing proteomic studies focused on epithelial ovarian cancer (EOC) have attempted to identify early disease biomarkers, establish molecular stratification, and discover novel druggable targets. Here we review these recent studies from a clinical perspective. Multiple blood proteins have been used clinically as diagnostic markers. The ROMA test integrates CA125 and HE4, while the OVA1 and OVA2 tests analyze multiple proteins identified by proteomics. Targeted proteomics has been widely used to identify and validate potential diagnostic biomarkers in EOCs, but none has yet been approved for

Extended Similarity Methods for Efficient Data Mining in Imaging Mass Spectrometry.

Imaging mass spectrometry is a label-free imaging modality that allows for the spatial mapping of many compounds directly in tissues. In an imaging mass spectrometry experiment, a raster of the tissue surface produces a mass spectrum at each sampled x, y position, resulting in thousands of individual mass spectra, each comprising a pixel in the resulting ion images. However, efficient analysis of imaging mass spectrometry datasets can be challenging due to the hyperspectral characteristics of the data. Each spectrum contains several thousand unique compounds at discrete m/z values that result …

High-Temperature Dissociation of Neopentanol: Shock Tube/Photoionization Mass Spectrometry Studies.

The pyrolysis mechanism of 2,2-dimethylpropan-1-ol (neopentanol) has been investigated at high temperatures (1128-1401K) and high pressures (5 and 15 bar). The experiments were performed in a miniature shock tube coupled to a time-of-flight mass spectrometer. Cations were generated by tunable vacuum ultraviolet photoionization resulting in multidimensional data sets containing mass and photoionization spectra and the time histories of species. At the elevated temperatures and pressures of this work, neopentanol was determined to dissociate primarily by the scission of a C-C bond yielding tert-

Uncertainty estimation for margin detection in cancer surgery using mass spectrometry.

PURPOSE: Rapid evaporative ionization mass spectrometry (REIMS) is an emerging technology for clinical margin detection. Deployment of REIMS depends on construction of reliable deep learning models that can categorize tissue according to its metabolomic signature. Challenges associated with developing these models include the presence of noise during data acquisition and the variance in tissue signatures between patients. In this study, we propose integration of uncertainty estimation in deep models to factor predictive confidence into margin detection in cancer surgery. METHODS: iKnife is us…

Understanding glycoprotein structural heterogeneity and interactions: Insights from native mass spectrometry.

Protein glycosylation is critical since it connects complex metabolic pathways to diverse proteoforms, fine-tunes protein structures and exerts biological functions. Aberrant glycosylation on the other hand is associated with many diseases, including cancers, inflammation and metabolic disorders. By resolving monosaccharide residues on intact glycoprotein complexes, native mass spectrometry can shed light on glycan heterogeneity, glycoprotein structure and molecular recognition. Here, we focus on the two most prevalent forms of glycosylation, namely N- and O- linked, and discuss recent progres…

Charge detection mass spectrometry on human-amplified fibrils from different synucleinopathies.

Amyloid fibrils are self-assembled mesoscopic protein aggregates, which can accumulate to form deposits or plaques in the brain. In vitro amplification of fibrils can be achieved with real-time quaking-induced conversion (RT-QuIC). However, this emerging technique would benefit from a complementary method to assess structural properties of the amplification products. This work demonstrates the feasibility of nanospray-charge-detection-mass-spectrometry (CDMS) performed on α-synuclein (αSyn) fibrils amplified from human brains with Parkinson's disease (PD) or Dementia with Lewy bodies (DLB) and

A TRUSTED targeted mass spectrometry assay for pan-herpesvirus protein detection.

The presence and abundance of viral proteins within host cells are part of the essential signatures of the cellular stages of viral infections. However, methods that can comprehensively detect and quantify these proteins are still limited, particularly for viruses with large protein coding capacity. Here, we design and experimentally validate a mass spectrometry-based Targeted herpesviRUS proTEin Detection (TRUSTED) assay for monitoring human viruses representing the three Herpesviridae subfamilies-herpes simplex virus type 1, human cytomegalovirus (HCMV), and Kaposi sarcoma-associated herpesv…
