Effects of post-extinction l-DOPA administration on the spontaneous recovery and reinstatement of fear in a human fMRI study.

Relapse is a pertinent problem in the treatment of anxiety disorders. In the laboratory, relapse is modeled as return of conditioned fear re … ediately after extinction. In mice, this effect could be attributed to an enhancement of extinction memory consolidation. In our human study, we could not exclude that l-DOPA might have acted by interfering with the consolidation of the original fear memory. In the present study, we therefore used a combined differential cue and context conditioning paradigm where ini …

Anxiolytic and sedative drugs: Sleep modulation and memory implications.

Sleep plays a crucial role in cognitive processes, particularly in memory consolidation, with Non-Rapid Eye Movement (NREM) and Rapid Eye Movement (REM) sleep phases playing distinct roles. Different classes of anxiolytics and sedatives influence sleep architecture and, consequently, memory function, with varying effects on learning, synaptic plasticity, and neurophysiological activity. While some drugs promote sedation and anxiolysis via GABAergic modulation, others act on serotonergic, dopaminergic, or orexinergic pathways, producing divergent effects on memory function. Rodent studies indic…

Aging-related changes in retrograde amnesia for mice.

Aging mice showed a retrograde amnesia for passive avoidance behavior when a single electroconvulsive shock (ECS) was given at post-training … of aging mice to the amnesic effect of ECS could be related to aging-related changes in the rate of memory consolidation or to the duration of neural chemical changes that disrupt processes such as consolidation, storage, and/or retrieval of the memory trace.

Blockade of striatal 5-HT2 receptors produces retrograde amnesia in rats.

We have recently reported that intrastriatal administration of the serotonin (5-HT) releasing drug p-chloroamphetamine, and of 5-HT itself, produces a significant retention deficit of inhibitory avoidance. It is not known which of the 5-HT receptors are involved in the amnesic effect of serotonin. The present experiment was aimed at determining whether 5-HT2 receptors within the striatum are involved in memory consolidation. Ketanserine (0.5, 1.0, 2.0, or 4.0 ng) was infused bilaterally into the striatum of Wistar rats immediately after training of inhibitory avoidance, and retention of the ta…

Functional Interactions Between Sleep and Circadian Rhythms in Learning and Learning Disabilities.

The propensity for sleep is timed by the circadian system. Many studies have shown that learning and memory performance is affected by circadian phase. And, of course it is well established that critical processes of memory consolidation occur during and depend on sleep. This chapter presents evidence that sleep and circadian rhythms do not just have separate influences on learning and memory that happen to coincide because of the circadian timing of sleep, but rather sleep and circadian systems have a critical functional interaction in the processes of memory consolidation. The evidence comes…

GHSR1A blockade by JMV2959 following memory retrieval disrupts reconsolidation and suppresses heroin-seeking behavior.

Opioid addiction persists as a critical public health crisis, where drug-seeking behaviors frequently re-emerge despite extended periods of abstinence. Current therapeutic approaches often fail to effectively address the substantial risk of relapse triggered by drug-related cues. Existing pharmacotherapies primarily focus on alleviating withdrawal symptoms or cravings, but they do not directly intervene in the memory processes that underlie relapse. Here, we assessed the promise of JMV2959, a specific GHSR1A antagonist, in disrupting the reconsolidation of heroin-associated memories by a rat a

Octopamine signaling from clock neurons plays dual roles in Drosophila long-term memory.

Circadian clock genes are best known for regulating circadian rhythms, but they also play crucial roles in memory processes. This suggests that memory is modulated by neural networks containing clock neurons, although the underlying mechanisms remain unclear. In Drosophila melanogaster, approximately 240 clock neurons are grouped into at least eight distinct clusters. Among them, the dorsal-lateral neurons (LNds) are required for maintaining long-term memory (LTM). In contrast, the neuropeptide Pigment-dispersing factor (Pdf), expressed in both small and large ventral-lateral neurons (s-LNvs a

The influence of sleep on fear extinction in trauma-related disorders.

In Posttraumatic Stress Disorder (PTSD), fear and anxiety become dysregulated following psychologically traumatic events. Regulation of fear and anxiety involves both high-level cognitive processes such as cognitive reattribution and low-level, partially automatic memory processes such as fear extinction, safety learning and habituation. These latter processes are believed to be deficient in PTSD. While insomnia and nightmares are characteristic symptoms of existing PTSD, abundant recent evidence suggests that sleep disruption prior to and acute sleep disturbance following traumatic events bot

Apelin receptor antagonist (ML221) facilitates memory reconsolidation in novel object recognition task.

The apelin receptor (APJ), a G protein-coupled receptor, has recently attracted attention as a potential modulator of memory processes, largely through the activity of its endogenous ligand, apelin. However, the effects of APJ blockade on memory, particularly in the process of reconsolidation, remain poorly understood. In this study, we investigated the role of the selective APJ antagonist 5-[(4-Nitrobenzoyl)oxy]-2-[(2-pyrimidinylthio)methyl]-4H-pyran-4-one (ML221) in modulating memory reconsolidation using the novel object recognition (NOR) paradigm. The NOR reconsolidation procedure consiste

Better later: evening practice is advantageous for motor skill consolidation in the elderly.

How does the time of day of a practice session affect learning of a new motor sequence in the elderly? Participants practiced a given finger tapping sequence either during morning or evening hours. All participants robustly improved performance speed within the session concurrent with a reorganization of the tapping pattern of the sequence. However, evening-trained participants showed additional gains overnight and at 1 wk posttraining; moreover, evening training led to a further reorganization of the tapping pattern offline. A learning experience preceding nocturnal sleep can lead to a task-s

Resting-state connectivity after visuo-motor skill learning is inversely associated with offline consolidation in Parkinson's disease and healthy controls.

Procedural memory refers to skills acquired through practice and depends on cortico-striatal and cortico-cerebellar circuits. These circuits are typically affected in Parkinson's disease (PD), leading to impaired skill learning, including defective offline consolidation, early in the course of the disease. Evidence points to a role of slow oscillations (<4 Hz) during sleep for offline consolidation. However recent studies showed consolidation over the course of the day, suggesting that consolidation may arise during wakefulness, too. Here we investigate whether functional connectivity (FC) at

Inhibition of protein synthesis but not β-adrenergic receptors blocks reconsolidation of a cocaine-associated cue memory.

Previously consolidated memories have the potential to enter a state of lability upon memory recall, during which time the memory can be altered before undergoing an additional consolidation-like process and being stored again as a long-term memory. Blocking reconsolidation of aberrant memories has been proposed as a potential treatment for psychiatric disorders including addiction. Here we investigated of the effect of systemically administering the protein synthesis inhibitor cycloheximide or the β-adrenergic antagonist propranolol on reconsolidation. Rats were trained to self-administer coc

Theta Activity During Encoding Interacts With NREM Sleep Oscillations to Predict Memory Generalization.

Relatively little is known regarding the interaction between encoding-related neural activity and sleep-based memory consolidation. One suggestion is that a function of encoding-related theta power may be to "tag" memories for subsequent processing during sleep. This study aimed to extend previous work on the relationships between sleep spindles, slow oscillation-spindle coupling, and task-related theta activity with a combined Deese-Roediger-McDermott (DRM) and nap paradigm. This allowed us to examine the influence of task- and sleep-related oscillatory activity on the recognition of both enc…

Disrupted hippocampal replay is associated with reduced offline map stabilization in an Alzheimer's mouse model.

Alzheimer's disease is characterized by progressive memory decline associated with hippocampal degeneration. However, the specific physiological mechanisms underlying hippocampal dysfunction in the disease remain poorly understood-improved knowledge may aid diagnosis and identify new avenues for therapeutic intervention. We investigated how disruptions in hippocampal reactivations relate to place cell stability and spatial memory deficits in an Alzheimer's mouse model. Using the App knockin mouse model NL-G-F, we conducted simultaneous behavioral and electrophysiological recordings in a radial

Having the landscape in the background: What can metabolic constraints explain about memory?

Haueis and Colaço claim that metabolic considerations function as explanatory constraints. I argue the explanatory contribution of metabolic constraints for synaptic models of memory is not well delineated. While metabolic constraints determine which synaptic architectures are biologically possible, they do not structure the outcome of synaptic consolidation. Metabolic constraints are explanatory since they account for the background conditions of memory formation.

Differential involvement of glutamatergic and catecholaminergic activity within the amygdala during taste aversion retrieval on memory expression and updating.

During memory retrieval, consolidated memories are expressed and destabilized in order to maintain or update information through a memory reconsolidation process. Despite the key role of the amygdala during memory acquistion and consolidation, the participation of neurotransmitter signals in memory retrieval is poorly understood. Hence, we used conditioned taste aversion and in vivo microdialysis to evaluate changes in glutamate, norepinephrine and dopamine concentrations within the amygdala during memory retrieval. We observed that exposure to an aversive-conditioned stimulus induced an augme

Cannabidiol effects on fear processing and implications for PTSD: Evidence from rodent and human studies.

Cannabidiol (CBD) modulates aversive memory and its extinction, with potential implications for treating anxiety- and stress-related disorders. Here, we summarize and discuss scientific evidence showing that CBD administered after the acquisition (consolidation) and retrieval (reconsolidation) of fear memory attenuates it persistently in rats and mice. CBD also reduces fear expression and enhances fear extinction. These effects involve the activation of cannabinoid type-1 (CB1) receptors in the dorsal hippocampus, bed nucleus of stria terminalis, and medial prefrontal cortex, comprising the an
