Some of the most promising novel tuberculosis vaccine strategies currently under development are based on respiratory vaccination, mimicking the natural route of infection. In this work, we have compared pulmonary and subcutaneous delivery of BCG vaccine in the tuberculosis-susceptible DBA/2 mouse strain, a model in which parenterally administered BCG vaccine does not protect against tuberculosis. In contrast to most mammalian species, the ciliary processes in the mouse eye form an irregular pattern. Different strains were studied using scanning electron microscopy. The ciliary processes of C57BL/6J animals showed quadrant-specific characteristics: in the superior quadrant, large and radial oriented processes were present. Circadian rhythms in body temperature, locomotor activity, and the circadian changes of plasma and pineal melatonin content were investigated in B6D2F(1) mice synchronized by 12 h of light and 12 h of darkness. During 8 wk continuous recording, activity and temperature displayed a marked stable and reproducible circadian rhythm, with both peaks occurring near the middle of darkness. The generation of cytotoxic T cells to alloantigens in mixed lymphocyte culture was suppressed by the addition of spleen cells from adult mice acutely infected with lymphocytic choriomeningitis virus. T cells of surface phenotype Lyt 1+2+ were required for the suppression. Suppressive cells appeared also in lymph nodes, but not in the thymus. In the spleen their number was maximal 6 to 8 days after infection. Infectious virus could not be detected in the suppressive spleen cells. Therefore the virus itself does not seem to cause the effect.

The inbred mouse strains C3H/HeJ and DBA/1J have been shown to represent the extremes of susceptibility and resistance, respectively, to pulmonary blastomycosis. This pattern was completely reversed when challenge was performed by the intraperitoneal route, whether a virulent or an attenuated strain of Blastomyces dermatitidis was utilized. By a third route (subcutaneous), the differences were insignificant. Inhibition of replication of blastomyces in vitro by macrophages from both strains, before or after activation by subcutaneous infection, was similar.
