Environmentally enriched pigs have transcriptional profiles consistent with neuroprotective effects and reduced microglial activity.

Environmental enrichment (EE) is widely used to study the effects of external factors on brain development, function and health in rodent models, but very little is known of the effects of EE on the brain in a large animal model such as the pig. Twenty-four young pigs (aged 5 weeks at start of study, 1:1 male: female ratio) were housed in environmentally enriched (EE) pens and provided with additional enrichment stimulation (a bag filled with straw) once daily. Litter, weight and sex matched controls n= (24) were housed in barren (B) conditions. Behaviour was recorded on alternate days from st

Preplaque ('preclinical') Aβ-induced inflammation and nerve growth factor deregulation in transgenic models of Alzheimer's disease-like amyloid pathology.

BACKGROUND: Alzheimer's disease (AD) neuropathology likely begins decades before clinical symptoms are manifested. Investigations on the early stages of the amyloid pathology are crucial for the discovery of diagnostic biomarkers or new therapeutic targets. Our transgenic (tg) animal models are most suitable to study early AD pathological events, as the pathology evolves in a well-staged manner, starting with intracellular Aβ accumulation and ending with plaque deposition. OBJECTIVE: To determine the occurrence of key inflammatory markers and to look for signs of nerve growth factor (NGF) dys

Drosophila dMRP4 regulates responsiveness to O2 deprivation and development under hypoxia.

For most vertebrates, oxygen is a prerequisite for survival. Although we have previously shown that Drosophila melanogaster is hypoxia tolerant, how this species senses O(2) deprivation and how it survives oxygen-limiting conditions are as yet poorly understood. We began to address this question by testing for anoxic responsiveness in Drosophila adult flies following overexpression of existing EP lines. In this screen, we identified Drosophila CG14709 gene as a homolog of the human multidrug resistance protein 4 (MRP4/ABCC4) that is tightly regulated to oxygen. Ubiquitous expression of dMRP4 i

Quantitative 1H magnetic resonance spectroscopic imaging determines therapeutic immunization efficacy in an animal model of Parkinson's disease.

Nigrostriatal degeneration, the pathological hallmark of Parkinson's disease (PD), is mirrored by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) intoxication. MPTP-treated animals show the common behavioral, motor, and pathological features of human disease. We demonstrated previously that adoptive transfer of Copaxone (Cop-1) immune cells protected the nigrostriatal dopaminergic pathway in MPTP-intoxicated mice. Herein, we evaluated this protection by quantitative proton magnetic resonance spectroscopic imaging (1H MRSI). 1H MRSI performed in MPTP-treated mice demonstrated that N-acetyl

DJ-1 deficiency in SH-SY5Y cells reveals dysregulated networks of genes and pathways involved in neuronal function and disease.

Parkinson's disease (PD) is the second most common neurodegenerative disorder. Although the etiology of idiopathic PD is unclear, recessive loss-of-function mutations in PARK7/DJ-1 cause familial early-onset PD, which mirrors key features of the idiopathic form. In this study, we ablate PARK7/DJ-1 via CRISPR-Cas9 from the human neuronal cell line, SH-SY5Y. Subsequently, RNA sequencing and the DESeq2 toolkit were utilized to identify 5468 differentially expressed genes (DEG) between PARK7/DJ-1 knockouts and control SH-SY5Y cells. Three genes from each of the top 10 upregulated and downregulated

α2δ-1 gene deletion affects somatosensory neuron function and delays mechanical hypersensitivity in response to peripheral nerve damage.

The α2δ-1 subunit of voltage-gated calcium channels is upregulated after sensory nerve injury and is also the therapeutic target of gabapentinoid drugs. It is therefore likely to play a key role in the development of neuropathic pain. In this study, we have examined mice in which α2δ-1 gene expression is disrupted, to determine whether α2δ-1 is involved in various modalities of nociception, and for the development of behavioral hypersensitivity after partial sciatic nerve ligation (PSNL). We find that naive α2δ-1(-/-) mice show a marked behavioral deficit in mechanical and cold sensitivity, bu

Spinal cord neuropathology in human West Nile virus infection.

CONTEXT: During the 1999 New York City West Nile virus (WNV) outbreak, 4 patients with profound muscle weakness, attributed to Guillain-Barré syndrome, were autopsied. These cases were the first deaths caused by WNV, a flavivirus, to be reported in the United States. The patients' brains had signs of mild viral encephalitis; spinal cords were not examined. During the 2002 national epidemic, several patients in Mississippi had acute flaccid paralysis. Electrophysiologic studies localized the lesions to the anterior horn cells in the spinal gray matter. Four of 193 infected patients in Mississip

The effects of self-generated synchronous and asynchronous visual speech feedback on overt stuttering frequency.

PURPOSE: Relatively recent research documents that visual choral speech, which represents an externally generated form of synchronous visual speech feedback, significantly enhanced fluency in those who stutter. As a consequence, it was hypothesized that self-generated synchronous and asynchronous visual speech feedback would likewise enhance fluency. Therefore, the purpose of this study was to investigate the effects of self-generated visual feedback (i.e., synchronous speech feedback with a mirror and asynchronous speech feedback via delayed visual feedback) on overt stuttering frequency in t

[Novel advances in neuropsychology--forward to the "deconstruction" of psychiatry].

Neuropsychology has recently become a science which deals not only with instrumental disorders (e.g., aphasia, apraxia, and agnosia), but also with impairments of interpersonal relationships (e.g., emotional cognitions, social decision making, and understanding others), and many important paradigms are already provided. We could enumerate several representative instances: (1)application of the "theory of mind" to autistic or related disorders (-->"reasoning of psychological state of others"), (2) neuropsychological studies on the "social brain" concerning emotional recognition or social recogn

Post-COVID-19 Hyposmia Does Not Exhibit Main Neurodegeneration Markers in the Olfactory Pathway.

The biological substrate of persistent post-COVID-19 hyposmia is still unclear. However, as many neurodegenerative diseases present with smell impairment at onset, it may theoretically reflect degeneration within the central olfactory circuits. However, no data still exist regarding the post-COVID-19 patients. As the olfactory neurons (ONs) mirror pathological changes in the brain, allowing for tracking the underlying molecular events, here, we performed a broad analysis of ONs from patients with persistent post-COVID-19 OD to identify traces of potential neurodegeneration. ONs were collected

Action Observation Therapy for Arm Recovery after Stroke: A Preliminary Investigation on a Novel Protocol with EEG Monitoring.

This preliminary study introduces a novel action observation therapy (AOT) protocol associated with electroencephalographic (EEG) monitoring … at AOT might be more effective in this subgroup of patients, perhaps because the integrity of their mirror neurons system (MNS) was more preserved, as indexed by EEG recording from central electrodes during action observation. In conclusion, AOT may reveal an effective rehabilitative tool in patients with subacute stroke; the EEG evaluation of MNS integrity may …

Neurophysiological pathways to obesity: below awareness and beyond individual control.

A global obesity epidemic is occurring simultaneously with ongoing increases in the availability and salience of food in the environment. Osteopontin Attenuates Secondary Neurodegeneration in the Thalamus after Experimental Stroke.

Cortical cerebral ischemia elicits neuroinflammation as well as secondary neuronal degeneration in remote areas. Locally distinct and specific secondary neurodegeneration affecting thalamic nuclei connected to cortical areas highlights such processes. Osteopontin (OPN) is a cytokine-like glycoprotein that is excreted in high amounts after cerebral ischemia and exerts various immunomodulatory functions. We here examined putative protective effects of OPN in secondary thalamic degeneration. We subjected male Wistar rats to photothrombosis and subsequently injected OPN or placebo intracerebrovent

Rhodamine-B induces amyotrophic lateral sclerosis symptoms like -behaviours in zebrafish.

Rhodamine B (RhB), often misused as a food adulterant, accumulates in the brain, causing oxidative stress and neurodegeneration. However, its detrimental effects on molecular mechanisms still require further investigation. The zebrafish is a model organism for studying neurodegeneration and neuropathology. We aimed to investigate the impacts of RhB on neuro-molecular and behaviours in Adult male zebrafish. Zebrafish were exposed to varying RhB concentrations of 0.25, 0.5, 1.0 μM and Rotenone (RoT) of 0.5 μM for 21 days. Behavioural assessments, including the mirror test, shoal preference test,

How Do We Motorically Resonate in Aging? A Compensatory Role of Prefrontal Cortex.

Aging is the major risk factor for chronic age-related neurological diseases such as neurodegenerative disorders and neurovascular injuries. Exploiting the multimodal nature of the Mirror Neuron System (MNS), rehabilitative interventions have been proposed based on motor-resonance mechanisms in recent years. Despite the considerable evidence of the MNS' functionality in young adults, further investigation of the action-observation matching system is required in aging, where well-known structural and functional brain changes occur. Twenty-one healthy young adults (mean age 26.66y) and 19 health

Decreased interhemispheric functional connectivity rather than corpus callosum volume as a potential biomarker for autism spectrum disorder.

Previous studies have implicated both structural and interhemispheric functional connectivity alterations in autism spectrum disorder (ASD) although findings are inconsistent. There is evidence that connectivity between corresponding regions in each hemisphere (homotopic) may be of particular importance and therefore the present study used the Autism Brain Imaging Data Exchange data to investigate ASD-related resting-state and structural alterations as well as associations with symptom severity (Autism Diagnostic Observation Schedule - ADOS). We employed a voxel-mirrored homotopic connectivity

Direction-selective laterodorsal thalamic nucleus encodes optic flow and turning in spatial navigation.

Vision is crucial for efficient navigation, but how the brain transforms visual signals into navigational cues remains unclear. The laterodorsal thalamic nucleus (LD), with strong connections to visual and navigation circuits, is well positioned for this function, yet its role has been largely unexplored. In mice, we found that LD neurons exhibit strong direction selectivity, with a dominant directional preference for contralateral-to-ipsilateral visual motion, mirroring horizontal optic flow during contraversive turning. In freely moving animals, LD ensembles were preferentially activated dur

Can we simulate an action that we temporarily cannot perform?

AIMS OF THE STUDY: The scope of individuals' motor repertoire and expertise influences the way they perceive the actions of others. When observing skilled actions, experts recruit the cortical network subserving action perception (action observation network, AON) to a greater extent than non-experts. However, it remains unknown whether and how a temporary motor injury affects activation within the AON. MATERIALS AND METHODS: To investigate this issue, brain hemodynamic activity was recorded twice in thirteen national female gymnasts suffering from a lower extremity injury at the onset of the

Evidence for a sensorimotor prediction in action observation.

The mirror neuron network is activated during both execution and observation of the same action, enabling visual-to-motor transformation. Two main hypotheses account for this process: the direct matching hypothesis proposes a direct link between visual cues and the observer's motor representations, whereas the predictive hypothesis suggests that motor predictions are generated and compared with visual information. We tested these hypotheses during a locomotion task. Motor prediction was assessed by comparing actual and imagined walking (forward or backward) over a 9 m path. Action observation
