Exploring Ecological Alternatives for Crop Protection Using Coriandrum sativum Essential Oil.

Essential oils (EOs) are a natural source of active compounds with antifungal, antimycotoxigenic, and herbicidal potential, and have been successfully used in organic agriculture, instead of chemical compounds obtained by synthesis, due to their high bioactivity and the absence of toxicity. The aim of this study was to highlight the importance of Coriandrum sativum essential oil (CEO) as a potential source of bioactive constituents and its applications as an antifungal and bioherbicidal agent. The CEO was obtained by steam distillation of coriander seeds and GC-MS technique was used to determi

Biosynthesis of Monascus pigments by resting cell submerged culture in nonionic surfactant micelle aqueous solution.

Growing cell submerged culture is usually applied for fermentative production of intracellular orange Monascus pigments, in which accumulation of Monascus pigments is at least partially associated to cell growth. In the present work, extractive fermentation in a nonionic surfactant micelle aqueous solution was utilized as a strategy for releasing of intracellular Monascus pigments. Those mycelia with low content of intracellular Monascus pigments were utilized as biocatalyst in resting cell submerged culture. By this means, resting cell submerged culture for production of orange Monascus pigme

Assessment of the microbody luminal pH in the filamentous fungus Penicillium chrysogenum.

The enzymes of the penicillin biosynthetic pathway in Penicillium chrysogenum are located in different subcellular compartments. Consequently, penicillin pathway precursors and the biologically active penicillins have to cross one or more membranes. The final enzymatic step that is mediated by acyltransferase takes place in a microbody. The pH of the microbody lumen in penicillin producing cells has been determined with fluorescent probes and mutants of the green fluorescent protein and found to be slightly alkaline.

A simple and cost-effective method for quantifying cyanide in the fruiting bodies and cultured mycelia of mushrooms using alkaline-SDS extraction and Conway diffusion.

This study describes a simple, cost-effective method for cyanide quantification in mushrooms. This involves an alkaline/SDS extraction producer to disrupt the cell wall while stabilizing volatile hydrogen cyanide as the cyanide ion (CN-) under alkaline conditions, which is subsequently recovered using Conway diffusion and quantified via pyridine-pyrazolone colorimetric analysis. This method demonstrated high sensitivity, with a limit of detection of 1.36 μg/g and limit of quantification of 4.82 μg/g. The spike recovery tests yielded an average recovery of 66-84 % (n = 9) at 10 μg/g CN- and a h

Antifungal effects of volatile organic compounds produced by Trichoderma hamatum against Neocosmospora solani.

Neocosmospora solani causes Fusarium wilt disease and root rot, which are serious problems worldwide. To determine the growth inhibition of Neocosmospora solani by Trichoderma hamatum volatile organic compounds (VOCs), the major chemical components of Trichoderma hamatum VOCs and the differences in their contents at different times were analysed, and the activity of these components was evaluated. The antifungal activity of Trichoderma hamatum was measured by a screening test, as Trichoderma hamatum exhibited strong antagonism against Neocosmospora solani in vitro. The double plate technique w

Life Cycle Dominates the Volatilome Character of Dimorphic Fungus Coccidioides spp.

Valley fever (coccidioidomycosis) is an endemic fungal pneumonia of the North and South American deserts. The causative agents of Valley fever are the dimorphic fungi Coccidioides immitis and C. posadasii, which grow as mycelia in the environment and as spherules within the lungs of vulnerable hosts. Current diagnostics for Valley fever are severely lacking due to poor sensitivity and invasiveness, contributing to a 23-day median time to diagnosis, and therefore, new diagnostic tools are needed. We are working toward the development of a breath-based diagnostic for coccidioidomycosis, and in t

Conversion of dried Aspergillus candidus mycelia grown on waste whey to biodiesel by in situ acid transesterification.

This study reports optimization of the transesterification reaction step on dried biomass of an oleaginous fungus Aspergillus candidus grown on agro-dairy waste, whey. Acid catalyzed transesterification was performed and variables affecting esterification, viz., catalyst methanol and chloroform concentrations, temperature, time, and biomass were investigated. Statistical optimization of the transesterification reaction using Plackett-Burman Design showed biomass to be the predominant factor with a 12.5-fold increase in total FAME from 25.6 to 320mg. Studies indicate that the transesterificatio

[Effects of cadmium stress on the antioxidative system in Ganoderma lucidum mycelia].

The study on the effects of different concentration (0, 10, 50, 100, 200 and 400 micromol x L(-1)) cadmium (Cd) on the antioxidative system in Ganoderma lucidum mycelia indicated that with increasing concentration of Cd, the fresh mass and the proline, total polysaccharides, and reduced polysaccharides contents of G. lucidum mycelia decreased, but non-protein thiol (NPT) content increased. At 400 micromol x L(-1) of Cd, the NPT content increased dramatically, being 5.7 times higher than control. Within the range of test Cd concentrations, the activities of CAT, GR and POD increased first and d

Fungal-sponge composite carriers coupled with denitrification and biomineralization bacteria to remove nitrate, calcium, and cadmium in a bioreactor.

The coexistence of nitrate (NO3--N) and heavy metals in the aquatic environment causes harm to both the aquatic ecosystem and human health. Here, fungal-sponge composite carriers (FSC) were assembled and immobilized with strain WZ39 in a bioreactor to remove NO3--N, Ca2+, and Cd2+. Stable bioreactor performance under heavy metal pressure was achieved. The highest removal efficiencies of NO3--N, Ca2+, and Cd2+ reached 100, 71.81, and 92.50%, respectively. Bacteria and precipitates were found in fungal mycelium and sponge. The precipitates composed of Ca3.9(Ca4.7Cd0.7)(PO4)6(OH)1.8, CaCO3, and C…

Chemical investigations and cytotoxic effects of metabolites from Antrodia camphorata against human hepatocellular carcinoma cells.

Antrodia camphorata is used as a medicinal fungus in Taiwan to treat fatigue, food intoxication, and enhance liver function. Here we identified fermented metabolic components from the mycelium of A. camphorata KH37 and explored their anti-hepatoma potentials with study models of human hepatoblastoma cell lines. Bioassay-guided fractionation of the solid fermentation powder of A. camphorata KH37 led to the isolation of one new quinonol, antroquinonol Z (1), and nine known compounds (2-10). Treatment with 10 μM antrocamols LT1 (2) or LT3 (3) reduced cell viability of HepG2 and Huh-7 cells to abo…

Co-regulation occurred upon heat shock from 36 to 42 degrees C and, only in Pb3, also during mycelium to yeast transformation (26-36 degrees C). In Pb18, PbMDJ1 seemed to be preferentially expressed in yeast. Our study might help understand regulation of genes involved in fungal adaptation to the host.
