The nude mouse does not reject xenografts of malignant and nonmalignant tissues of mammalian or avian origin, due to a deficiency of functional T-lymphocytes. In this study, tissue from a cold-blooded vertebrate, a teleost fish, was for the first time successfully transplanted to Swiss albino nu/nu mice. Malignant melanotic melanoma of Xiphophorus transplanted to nude mice showed progressive growth and could be serially passaged. Four lines of evidence (clinical findings, epidemiology, inhibition of tumor growth together with prolongation of survival time, and mechanism of action) which suggest that certain phenothiazines, particularly chlorpromazine, possess a range of anti-neoplastic activities in man and in rodents which has not been generally recognised are reviewed and discussed. Chlorpromazine interferes with energy production in sensitive tumours; the mode of action appears to correspond to autooxidative cellular injury, a common form of cell death.

The possible promoting effect of vagotomy and pyloroplasty on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced carcinogenesis in the gastrointestinal (GI) tract was studied in Syrian golden hamsters on high fat diet (10% corn oil). After truncal vagotomy and pyloroplasty or sham operation, 50 micrograms/ml of MNNG was given for 4 months in drinking water. Tetramethylthiourea (TMTU) was added to the diet of male and female ChR-CD rats for up to 2 years at levels of 0, 30 and 300 ppm. Fifty rats of each sex were used at each dietary level. The cytostatic TS-160 (trichloromethine hydrochloride, tris-/2-chloroethyl/amine hydrochloride) was injected subcutaneously into SPF Wistar rats of both sexes. A total 39 kidney tumors were induced by a continuous oral administration of N-butylnitrosourea (BNU) in 33 out of 204 rats of W/Fu strain and of 41 (W/Fu X ACI/N)F1 rats. No spontaneous renal tumors were observed among 66 males and 109 females of W/Fu rats which survived beyond the age of 19 months. Because of reports on tumorigenic activity in different animal species exposed to DDT a decision was made in 1969 to evaluate the long-term effects of DDT on 24 cynomolgus and rhesus monkeys.
