Human stem cell-derived neurons and astrocytes to detect novel auto-reactive IgG response in immune-mediated neurological diseases.

BACKGROUND AND OBJECTIVES: Up to 46% of patients with presumed autoimmune limbic encephalitis are seronegative for all currently known central nervous system (CNS) antigens. We developed a cell-based assay (CBA) to screen for novel neural antibodies in serum and cerebrospinal fluid (CSF) using neurons and astrocytes derived from human-induced pluripotent stem cells (hiPSCs). METHODS: Human iPSC-derived astrocytes or neurons were incubated with serum/CSF from 99 patients [42 with inflammatory neurological diseases (IND) and 57 with non-IND (NIND)]. The IND group included 11 patients with previ

Persistence of mechanoreceptors in ACL remnants: a histological and immunohistochemical analysis of 128 patients.

BACKGROUND: Anterior cruciate ligament (ACL) remnants may retain sensory neural elements after rupture, potentially contributing to proprioceptive function and graft reinnervation following reconstruction. However, the persistence of mechanoreceptors in remnant tissues and the factors influencing their preservation remain incompletely understood. This study aimed to evaluate the presence of neural structures in ACL remnants using histological and immunohistochemical methods and to determine the association between mechanoreceptor preservation, injury duration, and remnant attachment pattern.

[Progress in diagnosis and management of syncope in children].

Syncope is a common emergency of children and adolescents, which has serious influence on the quality of life. Neurally-mediated syncope, including postural tachycardia syndrome, vasovagal syncope, orthostatic hypotension and orthostatic hypertension, is the main cause of syncope in children and adolescents. The main manifestations of neurally-mediated syncope are diverse, such as dizziness, headache, chest tightness, chest pain, pale complexion, fatigue, pre-syncope and syncope. Although the clinical manifestations are similar, each subtype of syncope has its hemodynamic feature and optimal t

Studying spinal feedback loops post tendon transfer surgery using the pincer grasp task: A Neuro-Musculoskeletal study using NEUROiD.

The aim of the study is to create an in-silico test bed, which allows to replicate clinical scenarios using a virtual wrist model that encompasses both neural and musculoskeletal components along with implants. We used four wrist models: damaged FPL muscle; healthy FPL muscle; tendon-transfer surgery model with FPL latched to BRD muscle; and pulley-based surgery model with an implanted pulley over tendon-transfer model. These models were used for analyzing surgical cases and musculoskeletal biomechanics. This was developed on NEUROiD platform, simulating neural and musculoskeletal systems usin

A Digital Architecture for the Real-Time Tracking of Wearing off Phenomenon in Parkinson's Disease.

Levodopa administration is currently the most common treatment to alleviate Parkinson's Disease (PD) symptoms. Nevertheless, prolonged use of Levodopa leads to a wearing-off (WO) phenomenon, causing symptoms to reappear. To build a personalized treatment plan aiming to manage PD and its symptoms effectively, there is a need for a technological system able to continuously and objectively assess the WO phenomenon during daily life. In this context, this paper proposes a WO tracker able to exploit neuromuscular data acquired by a dedicated wireless sensor network to discriminate between a Levodop

Orientation and disorientation: lessons from patients with epilepsy.

Orientation in time, space, and person is a fundamental cognitive faculty and the bedrock of neurological and psychiatric mental status examination. Nevertheless, research in orientation and disorientation is neglected in both cognitive science and neuropsychiatry. Specifically, it is still unclear whether disorientations in time, space, and person represent a failure of the same system or merely share a common nomenclature and whether these three domains of orientation depend on different psychological and neural systems. Here, we analyzed descriptions of patients with specific orientation fa

Intestinal plexuses in Crohn's disease and ulcerative colitis in children: pathologic and microdissection studies.

The relative fraction of neural tissue in the plane of the myenteric (Auerbach) plexus (MEP) of ileum and colon of patients with Crohn's disease and ulcerative colitis was analyzed by point-count morphometric analysis of microdissected flat-mount ("circuit diagram") preparations of the plexus. Increase in the relative fraction of neural tissue (myenteric plexus hyperplasia) was seen in the MEP of some patients in both ileum and colon in both diseases. The frequency of ulceration or inflammation extending into the bowel wall deep enough to injure the various levels of the intestinal plexus syst

A Statistical Evaluation of Fetal Heart Rate Derived Parameters Between Healthy and IUGR Fetuses in Active and Quiet States.

The evaluation of fetal behavioral states, specifically the active and quiet phases, plays a crucial role in understanding fetal development during pregnancy. Pathological conditions such as intrauterine growth restriction (IUGR) can disrupt these states, affecting fetal heart rate (FHR) variability and other physiological parameters. This study utilizes a large dataset, comprising 13,652 healthy pregnancies and 7,047 IUGR cases, to analyze the differences in FHR parameters between active and quiet states across both healthy and IUGR pregnancies. A neural network-based approach, based on a pre

Non-pharmacological treatment affects neuropeptide expression in neuropathic pain model.

Chronic constriction injury (CCI) of the sciatic nerve elicits changes in neuropeptide expression on the dorsal root ganglia (DRG). The neural mobilization (NM) technique is a noninvasive method that has been proven clinically effective in reducing pain. The aim of this study was to analyze the expression of substance P, transient receptor potential vanilloid 1 (TRPV1) and opioid receptors in the DRG of rats with chronic constriction injury and to compare it to animals that received NM treatment. CCI was performed on adult male rats. Each animal was submitted to 10 sessions of neural mobilizat

Single-Trial Representations of Decision-Related Variables by Decomposed Frontal Corticostriatal Ensemble Activity.

The frontal cortex-striatum circuit plays a pivotal role in adaptive goal-directed behaviors. However, it remains unclear how decision-related signals are mediated through cross-regional transmission between the medial frontal cortex and the striatum by neuronal ensembles in making decision based on outcomes of past action. Here, we analyzed neuronal ensemble activity obtained through simultaneous multiunit recordings in the secondary motor cortex (M2) and dorsal striatum (DS) in rats performing an outcome-based left-or-right choice task. By adopting tensor component analysis (TCA), a single-t

Human Cytomegalovirus IE2 Disrupts Neural Progenitor Development and Induces Microcephaly in Transgenic Mouse.

Human cytomegalovirus (HCMV) is a significant contributor to congenital birth defects. Limited by the lack of animal models, the pathogenesis of neurological damage in vivo caused by HCMV infection and the role of individual viral genes remain to be elucidated. Immediate early (IE2) protein may play a function in neurodevelopmental problems caused by HCMV infection. Here, this study intended to investigate IE2's long-term effects on development of the brain in IE2-expressing transgenic mice (Rosa26-LSL-IE2+/-, Camk2α-Cre) aimed to observe the phenotype of postnatal mice. The expression of IE2

MRI-related anxiety can induce slow BOLD oscillations coupled with cardiac oscillations.

OBJECTIVE: Although about 1-2% of MRI examinations must be aborted due to anxiety, there is little research on how MRI-related anxiety affects BOLD signals in resting states. METHODS: We re-analyzed cardiac beat-to beat interval (RRI) and BOLD signals of 23 healthy fMRI participants in four resting states by calculation of phase-coupling in the 0.07-0.13 Hz band and determination of positive time delays (pTDs; RRI leading neural BOLD oscillations) and negative time delays (nTDs; RRI lagging behind vascular BOLD oscillations). State anxiety of each subject was assigned to either a low anxiety

Molecular mechanisms of hearing loss in Nager syndrome.

Nager syndrome is a rare human developmental disorder characterized by hypoplastic neural crest-derived craniofacial bones and limb defects. Mutations in SF3B4 gene, which encodes a component of the spliceosome, are a major cause for Nager. A review of the literature indicates that 45% of confirmed cases are also affected by conductive, sensorineural or mixed hearing loss. Conductive hearing loss is due to defective middle ear ossicles, which are neural crest derived, while sensorineural hearing loss typically results from defective inner ear or vestibulocochlear nerve, which are both derived

Acute phase response in amputated tail stumps and neural tissue-preferential expression in tail bud embryos of the Xenopus neuronal pentraxin I gene.

Regeneration of lost organs involves complex processes, including host defense from infection and rebuilding of lost tissues. We previously reported that Xenopus neuronal pentraxin I (xNP1) is expressed preferentially in regenerating Xenopus laevis tadpole tails. To evaluate xNP1 function in tail regeneration, and also in tail development, we analyzed xNP1 expression in tailbud embryos and regenerating/healing tails following tail amputation in the 'regeneration' period, as well as in the 'refractory' period, when tadpoles lose their tail regenerative ability. Within 10 h after tail amputation

Neural correlates of treatment response in depressed bipolar adolescents during emotion processing.

Depressive mood in adolescents with bipolar disorder (BDd) is associated with significant morbidity and mortality, but we have limited information about neural correlates of depression and treatment response in BDd. Ten adolescents with BDd (8 females, mean age = 15.6 ± 0.9) completed two (fearful and happy) face gender labeling fMRI experiments at baseline and after 6-weeks of open treatment. Whole-brain analysis was used at baseline to compare their neural activity with those of 10 age and sex-matched healthy controls (HC). For comparisons of the neural activity at baseline and after treatme
