Biomagnification of mercury in selected species from an Arctic marine food web in Svalbard.

Concentrations and biomagnification of total mercury (TotHg) and methyl mercury (MeHg) were studied in selected species from the pelagic food web in Kongsfjorden, Svalbard. Twelve species of zooplankton, fish and seabirds, were sampled representing a gradient of trophic positions in the Svalbard marine food web. TotHg and MeHg were analysed in liver, muscle and/or whole specimens. The present study is the first to provide MeHg levels in seabirds from the Svalbard area. The relative MeHg levels decreased with increasing levels of TotHg in seabird tissues. Stable isotopes of nitrogen (delta(15)N

Carbon and Acyl Chain Flux during Stress-induced Triglyceride Accumulation by Stable Isotopic Labeling of the Polar Microalga Coccomyxa subellipsoidea C169.

Deriving biofuels and other lipoid products from algae is a promising future technology directly addressing global issues of atmospheric CO2 balance. To better understand the metabolism of triglyceride synthesis in algae, we examined their metabolic origins in the model species, Coccomyxa subellipsoidea C169, using stable isotopic labeling. Labeling patterns arising from [U-13C]glucose, 13CO2, or D2O supplementation were analyzed by GC-MS and/or LC-MS over time courses during nitrogen starvation to address the roles of catabolic carbon recycling, acyl chain redistribution, and de novo fatty ac

Proxy Measures and Novel Strategies for Estimating Nitrogen Utilisation Efficiency in Dairy Cattle.

The efficiency with which dairy cows convert dietary nitrogen (N) to milk N is generally low (typically 25%). As a result, much of the N consumed is excreted in manure, from which N can be lost to the environment. Therefore there is increasing pressure to reduce N excretion and improve N use efficiency (NUE) on dairy farms. However, assessing N excretion and NUE on farms is difficult, thus the need to develop proximate measures that can provide accurate estimates of nitrogen utilisation. This review examines a number of these proximate measures. While a strong relationship exists between blood

Effect of raw humus under two adult Scots pine stands on ectomycorrhization, nutritional status, nitrogen uptake, phosphorus uptake and growth of Pinus sylvestris seedlings.

Ectomycorrhiza (EM) formation improves tree growth and nutrient acquisition, particularly that of nitrogen (N). Few studies have coupled the effects of naturally occurring EM morphotypes to the nutrition of host trees. To investigate this, pine seedlings were grown on raw humus substrates collected at two forest sites, R2 and R3. Ectomycorrhiza morphotypes were identified, and their respective N uptake rates from organic (2-(13)C, (15)N-glycine) and inorganic ((15)NH(4)Cl, Na(15)NO(3), (15)NH(4)NO(3), NH(4)(15)NO(3)) sources as well as their phosphate uptake rates were determined. Subsequently

Cryotherapy-induced milia en plaque: case report and literature review.

BACKGROUND: Cryotherapy-induced milia is a rarely described cutaneous reaction that may occur in patients who have received cryotherapy with liquid nitrogen. Cryotherapy-induced milia is characterized by 1-2 millimeter white dermal cysts that develop at the healed cryotherapy site. Milia en plaque, an erythematous plaque containing numerous milia, has not previously been described following treatment of a skin lesion with liquid nitrogen cryotherapy. PURPOSE: We describe a man who developed cryotherapy-induced milia en plaque after receiving cryotherapy to his dorsal hand for the treatment of

Beyond emissions: unravelling the effects of ecosystem change on contaminant concentrations in herring from the Baltic Sea.

The effects of environmental changes on contaminant fate in the ecosystem are poorly understood, even in the otherwise well-studied Baltic Sea. This area is considered one of the most polluted in the world and is currently undergoing rapid shifts related to climate change and eutrophication. In this study, we focus on the effects of an altered productivity base and changes in food web structure on contaminant concentrations in the commercially important Baltic herring, which is also a key-species in the ecosystem. In herring of known size and age, collected within the Swedish National Monitori

Contaminants in Atlantic walruses in Svalbard part 1: Relationships between exposure, diet and pathogen prevalence.

This study investigated relationships between organohalogen compound (OHC) exposure, feeding habits, and pathogen exposure in a recovering population of Atlantic walruses (Odobenus rosmarus rosmarus) from the Svalbard Archipelago, Norway. Various samples were collected from 39 free-living, apparently healthy, adult male walruses immobilised at three sampling locations during the summers of 2014 and 2015. Concentrations of lipophilic compounds (polychlorinated biphenyls, organochlorine pesticides and polybrominated diphenyl ethers) were analysed in blubber samples, and concentrations of perfluo

Cribra orbitalia as a potential indicator of childhood stress: Evidence from paleopathology, stable C, N, and O isotopes, and trace element concentrations in children from a 17th-18th century cemetery in Jēkabpils, Latvia.

Cribra orbitalia (CO), or porotic hyperostosis (PH) of the orbital roof, is one of the most common pathological conditions found in archaeological subadult skeletal remains. Reaching frequencies higher than 50% in many prehistoric samples, CO has been generally attributed to a variety of factors including malnutrition (e.g., megaloblastic anemia) and parasitism. In this study, we tested the relationship between CO, trace element concentrations, and stable isotope values (δ13C, δ15N, δ18O) in subadult skeletons from a 17th to 18th century cemetery in the historic town of Jēkabpils, Latvia. Combined use of stable isotopes and heavy metal analysis of Microlophus atacamensis reveals anthropogenic impacts in the coastal upwelling zone, Northern Chile.

This study evaluates the utility of the tail of the lizard Microlophus atacamensis as a biomonitor for assessing variation in heavy metals contamination (lead, copper, nickel, zinc and cadmium) using non- or minimally invasive methods that do not require sacrificing animals. Sampling was conducted at three coastal sites in Antofagasta (Atacama Desert) that differ in the degree of human impact. Three matrices were analyzed: soil, lizard tails (biological tissue) and the lizard putative prey. Lizard trophic ecology was characterized using stomach contents and the longer-term stable isotope ratio

Evaluating cleansing effects on trace elements and stable isotope values in feathers of oiled birds.

Feathers of seabirds are widely used as a nondestructive tissue for pollution monitoring of trace elements, as well as convenient samples for trophic ecology studies by means of stable isotope analysis (SIA). Nevertheless, feathers can be occasionally impregnated with oil from deliberate ship discharges and from massive oil spill accidents. The feather structure makes them effective traps for particles and are subject to external contamination. It is unknown to what extent the oil adhered to feathers can change trace element concentrations or stable isotope signatures. This study has two prima

Linking nutritional biochemistry and trophic ecology to health of adult female California sea lions in the Gulf of California.

Our planet is experiencing sudden and unpredictable changes that affect most land and marine environments. We investigated blood analytes relevant to nutritional biochemistry and isotopic signatures of adult female California sea lions (CSL) from the Gulf of California, an area that has suffered changes in sea surface temperature in the past decades. During the 2016 and 2020 breeding seasons we collected fur, plasma and serum samples from apparently healthy adult female CSL (2016, n = 43; 2020, n = 12). We determined packed cell volume (PCV) and quantified 11 blood analytes directly or indirec

Nonlinear Mixed Effects Modeling of Glucagon Kinetics Assessed Using [13C15N]-Glucagon in Individuals With and Without Type 1 Diabetes.

To date, few studies have quantified glucagon kinetics in humans with and without diabetes, with results often varying due to differences in experimental designs and glucagon assay methodologies. This has limited the ability to study glucagon secretion in vivo using methods such as deconvolution. To overcome these limitations, a novel stable glucagon tracer, [13C15N]-glucagon, along with high-resolution mass spectrometry, has been used to study glucagon kinetics and turnover in humans. In this work, we present a nonlinear mixed effects modeling approach to describe glucagon kinetics in healthy

1H, 13C and 15N resonance assignments and structure prediction of translation initiation factor 1 from Clostridium difficile.

Clostridium difficile is a gram-positive, toxin-producing, anaerobic bacterium whose virulence factors and mechanisms of pathogenesis require further investigation. C. difficile infections (CDI) result in the severe and potentially fatal gastrointestinal diseases pseudomembranous colitis and toxic megacolon following extensive broad spectrum antibiotic treatment. The increasing C. difficile fatalities are a result of the bacteria's growing antibiotic resistance and consequential CDI recurrence, which led to the unmet need for new CDI treatment. Bacterial protein synthesis is an essential metab

Seasonal and individual variation in the use of rail-associated food attractants by grizzly bears (Ursus arctos) in a national park.

Similar to vehicles on roadways, trains frequently kill wildlife via collisions along railways. Despite the prevalence of this mortality worldwide, little is known about the relative importance of wildlife attractants associated with railways, including spilled agricultural products, enhanced vegetation, invertebrates, and carcasses of rail-killed ungulates. We assessed the relative importance of several railway attractants to a provincially-threatened population of grizzly bears (Ursus arctos) in Banff and Yoho National Parks, Canada, for which rail-caused mortality has increased in recent de

Molecular-level secondary structure, polymorphism, and dynamics of full-length alpha-synuclein fibrils studied by solid-state NMR.

The 140-residue protein alpha-synuclein (AS) is able to form amyloid fibrils and as such is the main component of protein inclusions involved in Parkinson's disease. We have investigated the structure and dynamics of full-length AS fibrils by high-resolution solid-state NMR spectroscopy. Homonuclear and heteronuclear 2D and 3D spectra of fibrils grown from uniformly (13)C/(15)N-labeled AS and AS reverse-labeled for two of the most abundant amino acids, K and V, were analyzed. (13)C and (15)N signals exhibited linewidths of <0.7 ppm. Sequential assignments were obtained for 48 residues in the h

Resonance Raman study of cyanide-ligated horseradish peroxidase. Detection of two binding geometries and direct evidence for the "push-pull" effect.

Resonance Raman spectroscopy has been employed to investigate the structure of cyanide adducts of the basic isoenzymes of horseradish peroxidase (HRP) in the pH range 5.5-12.5. Evidence for the binding of cyanide in two forms, characterized by the reversal of ordering of the Fe-CN stretching and Fe-C-N bending vibrations, is observed. Moreover, it is shown that both conformers exhibit an acid-alkaline transition in the pH range employed. In the first conformer, the Fe-C-N linkage is essentially linear, exhibiting axial Fe-CN stretching and Fe-C-N bending frequencies at 453 and 405 cm-1, respec

An overview of nitrate sources and operating processes in arid and semiarid aquifer systems.

Nitrate concentration in most aquifers in arid and semi-arid areas has increased in the past several decades as a result of human activities. Under the predominantly oxic conditions of these aquifers, denitrification is inhibited, allowing nitrate, a soluble and stable form of nitrogen (N), to accumulate. Because of its close association with municipal and agricultural wastes, nitrate is commonly used as an indicator of anthropogenic contamination. Aquifers affected by agricultural waste may contain salts from irrigation returns and herbicides in addition to nitrates. Preventing leakage from s

Effects of oxygen-prebreathing on tissue nitrogenation in normobaric and hyperbaric conditions.

BACKGROUND: Breathing pure oxygen causes nitrogen washout from tissues, a method commonly deployed to prevent decompression sickness from hypobaric exposure. Theoretically prebreathing oxygen increases the capacity for nitrogen uptake and potentially limits supersaturation during dives of short duration. We aimed to use 13N2, a radioactive nitrogen isotope, to quantify tissue nitrogen following normobaric and hyperbaric exposures. METHODS: Twenty Sprague Dawley rats were divided in 4 conditions; normobaric prebreathe, normobaric control, hyperbaric prebreathe, hyperbaric control. Prebreathed
