Histopathology of the teeth in segmental odontomaxillary dysplasia: new findings.

Histological examination of the deciduous teeth in two cases of segmental odontomaxillary dysplasia (SOMD) showed fibrous enlargement of the pulps, an irregular pulp/dentine interface displaying many pseudoinclusions and pulp stones. There were tubular defects in the coronal dentine from pulp horn to cusp tip, an irregular tubular structure to the circumpulpal dentine of the apical half, a focally deficient odontoblast layer and widespread external resorption. Together with the clinical features of unilateral maxillary enlargement, upper alveolar expansion in the distal segment, increased spac

Healing of horizontal root fractures.

The roots of six anterior maxillary and mandibular teeth were intentionally fractured in three monkeys. The teeth were then rigidly splinted for 12 wk. The healing of the fractured roots was examined clinically, radiographically, and histologically for a period of 6 months. Five of six fracture sites healed by either dense connective tissue or mineralized hard tissue which appeared to enter the fracture sites from the periodontal ligament. The canal spaces in these teeth were reduced substantially by calcific deposits. The remaining pulp tissue appeared vital histologically, but with areas of

Cellular Dynamics and Dentine Bridge Formation Following Direct Pulp Capping With MTA and Biodentine in a Murine Model.

AIM: Formation of a mineralized dentine bridge is the hallmark of successful direct pulp capping (DPC). Mineral trioxide aggregate (MTA) and Biodentine are widely used capping materials, but in vivo evidence comparing their regenerative potential remains inconclusive. This study used a controlled murine model to compare dentine bridge formation and pulp response to MTA and Biodentine. METHODS: DPC was performed on maxillary first molars of mice using either MTA or Biodentine. Reparative dentine bridge formation was assessed by micro-computed tomography (bridge volume, mineral content, density

Differential Effects of Escherichia coli- Versus Porphyromonas gingivalis-derived Lipopolysaccharides on Dental Pulp Stem Cell Differentiation in Scaffold-free Engineered Tissues.

INTRODUCTION: To leverage the therapeutic capabilities of dental pulp stem cells (DPSCs) for regenerative endodontic applications, a better understanding of their innate defense and reparative processes is needed. Lipopolysaccharide (LPS) is a major virulent factor of gram-negative bacteria and contributor to endodontic infections. We have developed 3-dimensional scaffold-free DPSC tissues that self-organize into dentin-pulp organoids comprising a mineralized dentin-like tissue on the periphery and an unmineralized pulp-like core. In this study, scaffold-free DPSC constructs were used as contr

Activation and dynamic expression of Notch signalling in dental pulp cells after injury in vitro and in vivo.

AIM: To investigate the expression pattern of Notch signalling in odontoblast-like cells stimulated by lipopolysaccharide (LPS) in vitro, and in injured rat dental pulp in vivo. METHODOLOGY: Mouse odontoblast-like cells (MDPC-23) were exposed to LPS. Expression of Notch-related genes was detected by real-time PCR. A rat pulpitis model was established by mechanical injury and LPS plus mechanical injury was followed by the analysis of expression of Notch2 by immunohistochemical staining. One-way analysis of variance (anova) was performed to examine the effect of differing concentrations of LPS

In vivo study of the pulp reaction to Fuji IX, a glass ionomer cement.

OBJECTIVE: Our aims were to investigate the pulp biocompatibility of Fuji IX, a glass ionomer cement (GIC) used as a restorative material in cavities prepared in rat's upper molars, and to assess the value of this in vivo model for testing dental biomaterials. METHOD: Half-moon class V-like cavities were drilled on the mesial aspect of 26 rat upper first molars. Half of the experimental rats whose molars were restored with the GIC were killed after 8days and the second half after 30days. They were compared with two control groups, also submitted to cavity preparation, but with cavities left u

RvE1 Promotes Axin2+ Cell Regeneration and Reduces Bacterial Invasion.

Vital pulp therapy and root canal therapy (RCT) are the dominant treatment for irreversible pulpitis. While the success rate of these procedures is favorable, they have some limitations. For instance, RCT leads to removing significant dentin in the coronal third of the tooth that increases root-fracture risk, which forces tooth removal. The ideal therapeutic goal is dental pulp regeneration, which is not achievable with RCT. Specialized proresolving mediators (SPMs) are well known for inflammatory resolution. The resolution of inflammation and tissue restoration or regeneration is a dynamic an

Histologic evaluation of apical pulp of immature apex following extraction, surface treatment, and replantation in different storage media in dogs.

AIM: This study was designed to evaluate the apical pulp of open apex teeth following extraction, surface treatment, and preservation in different storage media in dogs. MATERIALS AND METHODS: Four healthy male dogs were included in this prospective animal study. In the first phase of extraction, 30 teeth were extracted atraumatically from dogs (incisors and premolars) and then randomly divided into three groups based on different types of storage media: (i) milk, (ii) HBSS, and (iii) dry storage. In milk and HBSS groups, teeth were contaminated by dog's saliva and then kept in milk and HBSS

Action of nitric oxide on healthy and inflamed human dental pulp tissue.

Irreversible pulpitis has been associated with pain and an increase in the number of pulp inflammatory cells. Based on the action of nitric oxide (NO) elsewhere, NO may possibly participate in the sensory and autonomic innervation of the dental pulp, and may influence local inflammatory responses. The purpose of this study was to analyze normal and inflamed human dental pulp for the presence of NADPH-diaphorase (NADPH-d), as an index of NO system activity. Six non-carious second premolar pulp tissue samples were obtained from young patients who required extractions for orthodontic reasons and

Pulpal response to different pulp capping methods after pulp exposure by air abrasion.

Air abrasion is regaining popularity especially in the area of pediatric dentistry due to its ease of use and its advantages. Due to the lost of tactile information, while using this technique, there is an increased risk for pulpal exposure. On the other hand, Ca(OH)2 medicament has been proven to induce dentin bridge formation, but an adequate sealing seems to be even more important that the capping material used. The purpose of this study was two fold: to assess the pulpal response after pulpal exposure by air abrasion and to evaluate the healing potential after using Ca(OH)2 medicament or L

C-CAM expression in odontogenesis and tooth eruption.

The distribution of the cell adhesion molecule C-CAM was analyzed during tooth development, eruption and formation of the junctional epithelium in rat molars by immunohistochemistry and in situ hybridization. C-CAM could not be detected during odontogenesis until the late bell stage; then only the mRNA was demonstrated in the odontoblasts and ameloblasts. Prior to eruption, a local increase in C-CAM (mRNA and protein) was observed in the reduced enamel epithelium. During eruption, high C-CAM levels were seen in the fusion zone between the oral epithelium and the reduced enamel epithelium. In t…

Odontoclastic resorption of the superficial nonmineralized layer of predentine in the shedding of human deciduous teeth.

Resorption by odontoclasts of a superficial nonmineralized layer of predentine that occurs in prior to the shedding of human deciduous teeth was studied by light and electron microscopy. As resorption of the tooth roots neared completion, multinucleate cells appeared on the predentine surface of the coronal dentine between the degenerated odontoblasts, excavated characteristic resorption lacunae in the nonmineralized predentine. These multinucleate cells had the same ultrastructural characteristics as odontoclasts and histochemical demonstration of tartrate-resistant acid phosphatase activity …

[The effect of doxorubicin on the unimpeded eruption rates of rat mandibular incisors].

We examined the effects of daily injections of doxorubicin, an anti-tumor drug, on unimpeded eruption rates, pulp cells, odontoblasts, periodontal ligament and dentine formation in the rat mandibular incisor. The experimental animals were intraperitoneally injected with 5 mg of doxorubicin/kg body weight/day for a period of 7 days. The eruption rates decreased gradually; and on the seventh day, the mean eruption rate had decreased to 14% of the control value. In addition, we noticed marked destruction of pulp cells and periodontal cells at the basal region of the incisor. The amount of dentine…

Influence of doxorubicin on rat incisor mesenchymal cells.

Three groups of female Wistar rats were intravenously injected with the antineoplastic drug doxorubicin in a single dose of 5 mg/kg, 10 mg/kg and 20 mg/kg, respectively. The animals were killed 1 and 5 days after injection. Paraffin and Vestopal W embedded sections were prepared of the maxillary right and left incisors, respectively, and evaluated by light microscopy. Doxorubicin produced necrosis of single cells, of small groups of cells in the basal pulp and destroyed the preodontoblasts as evaluated 1 day after injection. On the 5th day of observation, the late preodontoblasts and basal pul…

Effect of selenium on developing mouse teeth in culture in vitro.

First molars from 2-day-old mice were cultured for 7 days in medium supplemented with 0.04, 0.4, 4.0 parts/10(6) selenium (Se) or 10 per cent calf serum. Collagen synthesis, measured after labelling with [3H]-proline for the last 24 h of culture, was most active at 0.4 parts/10(6) Se. Supplementation with 300 parts/10(6) Se for the first 24 h of culture caused ultrastructural changes in newly-deposited enamel and dentine matrices.
