Interhemispheric transfer of olfactory information in homing pigeon.

The role of the anterior commissure in discrimination of and habituation to olfactory stimuli was studied in pigeons by measuring changes in the heart rate. In order to exclude any trigeminal response, we presented odorous stimuli to groups of pigeons with one olfactory nerve cut and either the ipsilateral or the contralateral nostril plugged. When the nostril involved in the experiments was on the same side of the cut olfactory nerve, the pigeons did not show any response, whereas they displayed changes in heart rate when the nostril tested was on the opposite side. In habituation transfer ex

Vestibular, olfactory, and vibratory responses of nucleus basalis prosencephali neurons in pigeons.

Evoked multiple unit responses were recorded through chronically implanted electrodes from the nucleus basalis prosencephali of the pigeon (Columba livia) upon vibratory stimulation of the beak-tip and airborne auditory stimulation, thus confirming earlier anatomical and physiological findings. Electrical stimulation of the olfactory nerve led to similar short latency responses. A specific directional sensitivity to rotatory vestibular stimulation was observed. Pitch motions of the head in the downward direction evoked the most pronounced multi-unit responses. These results support the suggest

Morphology and odor sensitivity of regenerated snail tentacles.

This study examined certain structural and functional aspects of the olfactory system in regenerated posterior tentacles of the terrestrial snail Achatina fulica. Regeneration of the epithelial sensory pad occurs with accurate size regulation. All five neuronal cell types which are normally revealed by horseradish peroxidase backfilling are also regenerated. The sensory cells attain normal numbers at about 20 weeks postlesion. The organization of neuronal elements within the tentacle is chaotic, however, at early stages of regeneration. Even later, the digitlike extensions of the ganglion, whi

The distribution of axon collaterals from the olfactory bulb and the nucleus of the horizontal limb of the diagonal band to the olfactory cortex, demonstrated by double retrograde labeling techniques.

Three different pairs of double retrograde axonal tracers have been used to study the distribution of axon collaterals from individual cells in the olfactory bulb and the nucleus of the horizontal limb of the diagonal band: (1) horseradish peroxidase (HRP) and tritiated apo-HRP (3H-HRP), (2) HRP and 125I-wheat germ agglutinin (I-WGA), and (3) the fluorochromes true blue (TB) and bisbenzimide (BB) or nuclear yellow (NY). With each combination of tracers, paired injections were made into different parts of the olfactory system, and the olfactory bulb and the nucleus of the diagonal band were exa

Inhibitory effects of centrally and peripherally induced anosmia on mounting behavior in the female rat.

Adult ovariectomized female rats received a testosterone-propionate (TP)-filled silastic implant and were tested for mounting behavior seven days later. Exposure to estrous female urine but not to urine from ovariectomized females increased the number of TP-treated females that displayed mounting behavior. Olfactory bulb removal on its own and peripheral anosmia induced by intranasal ZnSO4 application impaired mounting behavior. These results are discussed with respect to the effects of anosmia on copulatory behavior in the male.

Cell migration from the transplanted olfactory placode in Xenopus.

The eye vesicle of Xenopus borealis has been replaced with the transplanted olfactory primordium from Xenopus laevis in an attempt to determine whether cells from the transplant could migrate along the regrowing olfactory nerve and become incorporated into the CNS of the host. The use of X. laevis and X. borealis pairs allowed us to distinguish the cells of the host from those of the donor at the cellular level by means of the characteristic fluorescent nuclear spots (Q bands) of X. borealis. Transplantation was performed on pairs of animals at stages 23/24. The olfactory anlage was readily in

Olfactory GnRH3 crypt sensory neurons transduce sex pheromone signals to induce male courtship behavior in zebrafish.

Olfactory activation of neuroendocrine pathways plays vital roles in many organisms for reproduction and survival. The importance of gonadotropin-releasing hormone (GnRH) neurons for reproduction is well-established but little is known about whether they can directly sense and transmit sex pheromone signals. We have uncovered the migration path and distribution pattern of a new GnRH neuronal population that fulfills this role. GnRH3 neurons arise from the region located beneath olfactory placode, undergo bidirectional migration along the olfactory nerve, and cell bodies lie within the olfactor

Implantation of a scaffold following bulbectomy induces laminar organization of regenerating olfactory axons.

Primary olfactory axons expressing different odorant receptors are interspersed within the olfactory nerve. However, upon reaching the outer nerve fiber layer of the olfactory bulb they defasciculate, sort out, and refasciculate prior to targeting glomeruli in fixed topographic positions. While odorant receptors are crucial for the final targeting of axons to glomeruli, it is unclear what directs the formation of the nerve fiber and glomerular layers of the olfactory bulb. While the olfactory bulb itself may provide instructive cues for the development of these layers, it is also possible that

Olfactory Evaluation in Clinical Medical Practice.

The subjective and demanding nature of olfactory testing means that it is often neglected in clinic despite loss of smell leading to significant limitations in everyday life. The list of diseases associated with loss of olfaction far exceeds the field of otorhinolaryngology and can also be seen in neurodegenerative disorders. Knowledge of possible clinical testing is essential to determine a proper differential diagnosis for the loss of olfactory sense. Causes of olfactory impairment can be divided into either failure in transferring odour to the organ of perception or damage to the olfactory

Organization of connections of the basal and accessory basal nuclei in the monkey amygdala.

The present study investigated the intrinsic connections of the basal and accessory basal nuclei of the Macaca fascicularis monkey by means of the anterograde tracers Phaseolus vulgaris-leucoagglutinin (PHA-L) and biotinylated dextran amine (BDA). Analysis of the intranuclear connections of the basal nucleus indicates that there are five modules: dorsal, intermediate, ventral lateral, ventral medial and periamygdaloid sulcal cortex. The dorsal division projects to the intermediate division. Laterally, the intermediate division projects to the ventral lateral division and dorsal parts of the ve

Second-order input to the medial amygdala from olfactory sensory neurons expressing the transduction channel TRPM5.

Recent anatomical tracing experiments in rodents have established that a subset of mitral cells in the main olfactory bulb (MOB) projects directly to the medial amygdala (MeA), traditionally considered a target of the accessory olfactory bulb. Neurons that project from the MOB to the MeA also show activation in response to conspecific (opposite sex) volatile urine exposure, establishing a direct role of the MOB in semiochemical processing. In addition, olfactory sensory neurons (OSNs) that express the transient receptor potential M5 (TRPM5) channel innervate a subset of glomeruli that respond

The SRC homology 2 domain protein Shep1 plays an important role in the penetration of olfactory sensory axons into the forebrain.

Shep1 is a multidomain signaling protein that forms a complex with Cas, a key scaffolding component of integrin signaling pathways, to promote the migration of non-neuronal cells. However, the physiological function of Shep1 in the nervous system remains unknown. Interestingly, we found that Shep1 and Cas are both concentrated in the axons of developing olfactory sensory neurons (OSNs). These neurons extend their axons from the olfactory epithelium to the olfactory bulb located at the anterior tip of the forebrain. However, in developing Shep1 knock-out mice, we did not detect penetration of O

Synchronized rhythmic discharges of the secondary olfactory neurons in carp.

(1) Rhythmic and synchronized multiunit bursts of spike discharges, which were superimposed on the peaks of the oscillatory potentials of 4--10 Hz (olfactory tract waves), were induced in the olfactory tract fibers of the carp in response to olfactory stimulation. Suppression of the spike discharges was observed during the interburst periods of discharges. (2) Single-unit analysis revealed three types of discharge patterns in the olfactory tract fibers in response to olfactory stimulation: rhythmic excitation (Type 1), suppression (Type 2) and no response (Type 3). A synchronization of spike d

Mechanisms of Neuronal Differentiation and Notch Signaling as a Potential Therapeutic Target in Olfactory Dysfunction of Allergic Rhinitis.

PURPOSE: Olfactory dysfunction (OD) in allergic rhinitis (AR) significantly diminishes quality of life, yet its pathophysiology remains unclear. The Notch signaling pathway is known to regulate olfactory epithelium proliferation and differentiation in murine models, but its role in AR with OD (AR+OD) is yet to be elucidated. This study aims to explore neuronal expression patterns and the involvement of Notch signaling in AR+OD. PATIENTS AND METHODS: Symptom severity of 111 AR patients, 65 non-AR patients, and controls was evaluated according to the SNOT-22 criteria. Olfactory dysfunction in a

Structural and functional connectomics of the olfactory system in Parkinson's disease: a systematic review.

Olfactory dysfunction, affecting 75-90 % of Parkinson's disease (PD) patients, holds significant predictive value for PD development. Advanced imaging techniques, such as diffusion MRI (dMRI) and functional MRI (fMRI), offer insights into structural and functional changes within olfactory pathways. This review summarizes a decade of research on MRI-based connectivity of the olfactory system in PD, focusing on structural and functional alterations in olfactory brain areas and their links to early olfactory processing changes. Fifteen dMRI and eighteen fMRI studies met inclusion criteria and wer…

Spatial and temporal mapping of early alphaherpesvirus invasion routes into the mouse central nervous system.

Alphaherpesviruses such as Herpes Simplex Virus 1 (HSV-1) and Pseudorabies virus (PrV) invade the central nervous system (CNS) via peripheral nerves. While olfactory and trigeminal pathways are well-known, additional cranial routes remain underexplored. Using a PrV-∆UL21gfp/US3∆kin mutant in CD1 mice, we mapped early neuroinvasion (4-96 hpi) by immunofluorescence and RNA in situ hybridization. Viral antigen and lytic viral gene expression (UL19 RNA) were detected in the olfactory epithelium, vomeronasal organ, incisors, palate, olfactory bulb, and brainstem. These results indicate multineural
