Hematological adaptations in diploid and triploid Salvelinus fontinalis and diploid Oncorhynchus mykiss (Salmonidae, Teleostei) in response to long-term exposure to elevated temperature.

Hematology is a simple but reliable method to determine the influence of environmental stressors on the physiology and performance of teleost fish. Understanding the detailed impacts of elevated temperature on the hematology of fish can help us to evaluate fish responses to climate change. Therefore, cellular and biochemical changes in peripheral blood were investigated in diploid (2n) and triploid (3n) brook trout and in 2n rainbow trout exposed to 20 °C for 32 days in comparison to fish acclimated to 9 °C. Additionally, survival and growth rates and the relative mRNA expression of heat-shock

Hypoxic responses in Oncorhynchus mykiss involve angiogenesis, lipid, and lactate metabolism, which may be triggered by the cortisol stress response and epigenetic methylation.

The incidence of hypoxia in water bodies is increasing more rapidly than aquatic life can adapt. This study aimed to determine the effects of hypoxia on fish physiology, as well as protein expression through proteomics. To do this, 40 rainbow trout were divided into normoxic control (11.5 mg/L dissolved oxygen) and hypoxic treatment (5 mg/L dissolved oxygen) tanks for a period of 7 days. Fish were then anesthetized and blood was sampled. Fish were then euthanized and heart and liver samples were taken. Blood glucose, cortisol and lipid, body and liver mass, fork length, hematocrit and, blood c

The role of the kidney in compensating the alkaline tide, electrolyte load, and fluid balance disturbance associated with feeding in the freshwater rainbow trout, Oncorhynchus mykiss.

The effect in freshwater rainbow trout of digesting a commercial pellet meal on the renal handling of water, ions and acid-base equivalents was investigated through urine collection over a 48 h period following meal ingestion. The glomerular filtration rate (GFR) and urine flow rate (UFR) were reduced in fed fish between 12 and 24h following the meal, likely reflecting a loss of endogenous water across the gastric epithelium as a result of ingesting dry, ion-rich food pellets. The kidney was also responsible for the excretion of some excess dietary Ca(2+), and, to a much lesser extent, Na(+) a

Isolation and characterization of a cDNA encoding for preprosomatostatin containing [Tyr7, Gly10]-somatostatin-14 from the endocrine pancreas of rainbow trout, Oncorhynchus mykiss.

Somatostatins are a diverse group of peptides known to influence various aspects of growth and metabolism of vertebrates. In order to further our understanding of the physiological roles of somatostatins in fish, we initiated an analysis of somatostatin gene structure and expression in rainbow trout pancreas. Using rapid amplification of cDNA ends polymerase chain reaction, we have isolated, cloned, and sequenced a novel cDNA derived from pancreatic total RNA. Sequence analysis revealed a 624-bp cDNA containing the complete 5'-untranslated region with a single initiation site 107 bases from th

Contrasted modifications of IgM and IgT repertoires induced by high- and low-virulent infectious pancreatic necrosis virus strains in rainbow trout (Oncorhynchus mykiss).

INTRODUCTION: Infectious pancreatic necrosis virus (IPNV) is a significant pathogen in salmonid aquaculture, with virulence determined mainly by amino acid residues in the VP2 capsid protein. METHODS: This study investigates the modifications of rainbow trout (Oncorhynchus mykiss) IgM and IgT repertoire induced by two IPNV strains that differ at VP2 positions 217 and 221, specifically IPNV-TA (T217A221) and IPNV-PT (P217T221). For these studies, doubled-haploid isogenic trout were immunised with IPNV-TA or IPNV-PT, and IgHμ and IgHτ repertoires were analysed by high-throughput sequencing. RE…

Development of anti-CD4-1/CD8β monoclonal antibodies and spatiotemporal immune profiling of response to live attenuated IHNV in rainbow trout (Oncorhynchus mykiss).

Understanding the cellular mechanisms of live attenuated vaccines against infectious hematopoietic necrosis virus (IHNV) has long been hindered by the lack of species-specific immunological reagents in rainbow trout (Oncorhynchus mykiss). To bridge this gap and elucidate the protective basis of a high-efficacy mutant candidate (rIHNVMut), this study first established a core technical platform by developing and validating monoclonal antibodies (mAbs) targeting the T-cell co-receptors CD4-1 and CD8β. Validation via Western blot and immunohistochemistry confirmed the high specificity of these mAb…

Immunogenicity and seasonal effect of formalin-killed bivalent vaccines against Streptococcus iniae and Yersinia ruckeri in rainbow trout (Oncorhynchus mykiss) under farm conditions.

Streptococcosis and Yersiniosis are prominent diseases affecting rainbow trout farming, frequently leading to large-scale fish deaths and substantial financial setbacks for producers. This study aimed to evaluate the immunogenicity of formalin-killed vaccines (injection form) and assess the seasonal influences on streptococcosis and yersiniosis in rainbow trout. In this study, 1500 fish were divided into two groups: a vaccinated group (VG, vaccinated against both streptococcosis and yersiniosis) and an unvaccinated control group, with three replicates per group in each season. The average fish

Oral immunization with attenuated Salmonella Typhimurium as a carrier of DNA vaccine against infectious hematopoietic necrosis virus (IHNV) in rainbow trout (Oncorhynchus mykiss).

Infectious hematopoietic necrosis virus (IHNV) is a serious pathogen in the salmonid aquaculture industry and leads to economic losses in the world. This study aimed to develop a new oral DNA vaccine designed to protect rainbow trout against infection by IHNV. Fish were administered via the oral route by the attenuated Salmonella enterica serovar Typhimurium as a carrier of pcDNA3.1-IHNG (glycoprotein (G)) plasmid for 7 days and finally, fish were challenged by 105 pfu ml-1 IHNV. The results revealed that the antigen gene was identified in different tissues of rainbow trout at 15-, 30-, and 45

Bioretention filtration prevents acute mortality and reduces chronic toxicity for early life stage coho salmon (Oncorhynchus kisutch) episodically exposed to urban stormwater runoff.

As the human population of western North America continues to expand, widespread patterns of urban growth pose increasingly existential threats to certain wild stocks of Pacific salmon and steelhead (Oncorhynchus sp.). Rainfall previously absorbed into the soils of forests and grasslands falls instead on pavement and other hardened surfaces. This creates stormwater runoff that carries toxic metals, oil, and many other contaminants into salmon-bearing habitats. These include freshwater streams where coho salmon (O. kisutch) spawn in gravel beds. Coho salmon embryos develop within a thick eggshe…

Studying the non-thermal effects of microwave on amino acids in sterilized rainbow trout (Oncorhynchus mykiss) fillets using a double side approximating method.

The aim of this study was to investigate the impact of microwave non-thermal effects on thermal sensitive amino acids in rainbow trout (Oncorhynchus mykiss) fillets. To distinguish non-thermal effects from thermal effects occurring simultaneously, a Double Side Approximating Method (DSAM) derived from computational mathematics was developed. Two corresponding water bath treatments were designed for each microwave processing to approximate the time-temperature profiles at the hot and cold spots of the microwave processed samples while maintaining a comparable thermal intensity (F0). The microwa…

Active feeding of downstream migrating juvenile pink salmon (Oncorhynchus gorbuscha) revealed in a large Barents Sea river using diet and stable isotope analysis.

The recent, rapid spreading of non-native pink salmon Oncorhynchus gorbuscha in the North Atlantic area has raised concerns about their possible negative impacts on native salmonid species. Potential interactions include competition for food resources during the short freshwater phase of juvenile O. gorbuscha, but little is known about their feeding behavior in the newly occupied North Atlantic rivers. Using stable isotope and stomach content analyses, patterns of freshwater feeding of non-native O. gorbuscha fry were studied in a large Fennoscandian river, the Teno, that discharges to the Bar…

CRISPR/Cas9-Mediated Transgenesis of the Masu Salmon (Oncorhynchus masou) elovl2 Gene Improves n-3 Fatty Acid Content in Channel Catfish (Ictalurus punctatus).

Omega-3 polyunsaturated fatty acids (n-3 PUFAs), particularly eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), play a very important role in human health. Channel catfish (Ictalurus punctatus) is one of the leading freshwater aquaculture species in the USA, but has low levels of EPA and DHA compared to some fish such as salmon. To improve EPA and DHA content, a modification of the n-3 PUFA biosynthetic pathway was achieved through the insertion of an elovl2 transgene isolated from masu salmon (Oncorhynchus masou) driven by a carp β-actin promoter using a two-hit by…

Inhibition of RNA synthesis differentially affects in vitro melatonin release from the pineal organs of ayu (Plecoglossus altivelis) and rainbow trout (Oncorhynchus mykiss).

The effects of actinomycin D (RNA synthesis inhibitor) and cycloheximide (protein synthesis inhibitor) on melatonin release from the cultured pineal organ of two teleosts with or without the circadian regulation of melatonin production (ayu Plecoglossus altivelis and rainbow trout Oncorynchus mykiss, respectively) were investigated. Actinomycin D decreased melatonin release from the pineal organ during the dark phase but there was a significant difference between the two species (22.2% for ayu and 59.1% for trout as compared with the respective control). This difference might be due to whether
