Orientia tsutsugamushi Ank9 is a multifunctional effector that utilizes a novel GRIP-like Golgi localization domain for Golgi-to-endoplasmic reticulum trafficking and interacts with host COPB2.

Orientia tsutsugamushi causes scrub typhus, a potentially fatal infection that afflicts 1 million people annually. This obligate intracellular bacterium boasts one of the largest microbial arsenals of ankyrin repeat-containing protein (Ank) effectors, most of which target the endoplasmic reticulum (ER) by undefined mechanisms. Ank9 is the only one proven to function during infection. Here, we demonstrate that Ank9 bears a motif that mimics the GRIP domain of eukaryotic golgins and is necessary and sufficient for its Golgi localization. Ank9 reaches the ER exclusively by retrograde trafficking …

Development of an Orientia tsutsugamushi Lc-1 Murine Intraperitoneal Challenge Model for Scrub Typhus: Determination of Murine Lethal Dose (MuLD50), Tissue Bacterial Loads, and Clinical Outcomes.

Currently, no vaccine has been developed to protect humans from naturally acquired heterologous Orientia tsutsugamushi infections. To enhance the validity of vaccine candidates, we are developing a murine chigger challenge model with the O. tsutsugamushi Lc-1-infected Leptotrombidium chiangraiensis Line-1. To this end, an intraperitoneal (i.p.) murine challenge model using an O. tsutsugamushi Lc-1 isolate was developed for eventual validation of the chigger challenge model. We have determined that the murine lethal dose that kills 50% of the challenged mice (MuLD50) of a liver/spleen homogenat…

Evaluation of a broad-ranging and convenient enzyme-linked immunosorbent assay using the lysate of infected cells with five serotypes of Orientia tsutsugamushi, a causative agent of scrub typhus.

BACKGROUND: Scrub typhus is a mite-borne rickettsiosis caused by infection of Orientia tsutsugamushi, which is endemic to several Asia-Pacific Rim countries, including Japan. Although micro-indirect immunofluorescent assay (micro-IFA) is the standard method for the serological diagnosis of scrub typhus, enzyme-linked immunosorbent assay (ELISA) is considered to be more objective, by providing digitized results as opposed to being subject to the judgment of the evaluator as in micro-IFA. Therefore, the aim of this study was to develop a broad-ranging ELISA using the five major prevalent serotyp…

Development of a recombinant protein-based enzyme-linked immunosorbent assay and its applications in field surveillance of rodent mice for presence of immunoglobulin G against Orientia tsutsugamushi.

A recombinant protein containing the immunodominant conserved epitope region of the 56-kDa outer membrane protein of the Karp strain of Orientia tsutsugamushi was purified to near homogeneity using recombinant DNA techniques. The purified protein was used to immunize rabbits and produced an antibody that could recognize different strains of O. tsutsugamushi, as demonstrated both by Western blotting and immunofluorescence assay. An enzyme-linked immunosorbent assay (ELISA) based on this recombinant protein was developed to detect antibody (immunoglobulin G [IgG]) against O. tsutsugamushi in mic…

The High Prevalence of Orientia tsutsugamushi Among Wild Rodents and the Identification of Its Major Serotypes in Yamaguchi Prefecture, Japan, Where Scrub Typhus Patients Are Rarely Identified.

Orientia tsutsugamushi (OT) is an obligate intracellular bacterium transmitted by larval trombiculid mites, responsible for scrub typhus in humans. In Japan, approximately 500 human cases are reported annually, with six major serotypes identified as Kato, Karp, Gilliam, Irie/Kawasaki, Shimokoshi, and Hirano/Kuroki. However, the prevalence of OT in Yamaguchi prefecture remains largely unknown. This study aimed to investigate the prevalence and serotypes of OT in wild rodents collected from eight locations within the prefecture. DNA was extracted from spleen and liver samples of 135 wild rodents…

Evaluation of Recombinant Type-Specific Antigens of Orientia tsutsugamushi Expressed by a Baculovirus-Insect Cell System as Antigens for Indirect Immunofluorescence Assay in the Serological Diagnosis of Scrub Typhus.

Scrub typhus (ST) is a mite-borne rickettsiosis caused by the intracellular bacterium Orientia tsutsugamushi (OTS), which is classified as a biosafety level-3 (BSL-3) pathogen. For serological tests of ST, mouse fibroblast cells infected with the five prevalent serotypes of OTS in Japan are generally used as antigens for indirect immunofluorescence assay (IFA). In this study, Spodoptera frugiperda derived insect cell line (Sf9) cells infected with recombinant type-specific antigen (rTSA)-expressing baculovirus were used for IFA. The paired serum samples of 15 ST patients, 10 rickettsiosis pati…

Acarological investigation and detection of Orientia tsutsugamushi (Rickettsiales: Rickettsiaceae) in field-collected chiggers (Acari: Trombiculidae) and small mammals from scrub typhus positive localities in Pahang, West Malaysia.

Scrub typhus (ST) is an acute febrile illness caused by Orientia tsutsugamushi (Hayashi, 1920) that is transmitted to humans through the bite of infected chiggers. In recent years, ST has re-emerged as a public health concern in Malaysia. In 2024, more than 50 laboratory-confirmed human cases of ST were reported in Pahang. A collaborative investigation was undertaken on a subset of 26 cases to identify potential vector sources and animal hosts associated with O. tsutsugamushi transmission. Chigger-based sampling was conducted by trapping small mammals in suspected infection hotspots, including…
