Weight-Loss Strategies for Prevention and Treatment of Hypertension: A Scientific Statement From the American Heart Association.

Hypertension is a major risk factor for cardiovascular and renal diseases in the United States and worldwide. Obesity accounts for much of the risk for primary hypertension through several mechanisms, including neurohormonal activation, inflammation, and kidney dysfunction. As the prevalence of obesity continues to increase, hypertension and associated cardiorenal diseases will also increase unless more effective strategies to prevent and treat obesity are developed. Lifestyle modification, including diet, reduced sedentariness, and increased physical activity, is usually recommended for patie

Obesity Epidemic: Pharmaceutical Weight Loss.

Obesity is a chronic disease universally defined as an excess of adipose tissue resulting in body mass index (BMI) > 30.0 kg/m2. Over the past few years, the concept of prevention has gained increased awareness, thus leading to the development of additional pharmaceutical options for the treatment of obesity since 2012. Treating obesity revolves around an individualized, multi-disciplinary approach with additional focus on a healthy and supportive lifestyle to maintain the weight loss. Targeting oncogenic activation of FLT3/SREBP/FASN promotes the therapeutic effect of quizartinib involving disruption of mitochondrial phospholipids.

FMS-like tyrosine kinase 3-internal tandem duplication (FLT3/ITD) is a common driver mutation that presents with a high leukemic burden and … ukemia cells to quizartinib. Quizartinib combined with SREBP inhibitor fatostatin or FASN inhibitor orlistat provided substantial therapeutic benefit over monotherapies in the murine FLT3/ITD leukemia model. Our results indicated the mechanistic link between FLT3/ITD and SREBP degradation and suggested the combination therapy via targeting FLT3/SREBP/FASN …

Effects of Octacosanol Isolated from Moringa oleifera Leaves on Inhibiting the Activity of Pancreatic Lipase.

Moringa oleifera Lam., a perennial species of the Moringaceae family, is esteemed for its multifaceted nutritional, medicinal, and economic … ecular docking analyses revealed that OCT binds to pancreatic lipase with an affinity comparable to orlistat and stronger than that of 4-nitrophenyl palmitate. The binding of OCT to key active sites (Ser152, His263, and Asp176) likely disrupts the enzyme's conformation, decreasing its substrate affinity. Additionally, OCT significantly attenuated lipid abs …

Identification of effective synthetic molecules against viral-induced cytokine release syndrome using in silico and in vitro approaches.

Acute respiratory distress syndrome (ARDS) is the leading cause of mortality in pathogen-mediated lung inflammation. Viral-induced cytokine release syndrome (CRS) has emerged as a global pandemic, characterized by a hyperactive immune response and excessive cytokine production causing irreversible lung injury. This study aimed to evaluate FDA-approved drugs for their potential to target hyperactive immune response and SARS-CoV-2 viral replication simultaneously. Six potential 3-CLpro inhibitors were identified by molecular docking using MOE software, including ebastine (1), orlistat (2), atrac…

Weight loss significantly reduces serum lipocalin-2 levels in overweight and obese women with polycystic ovary syndrome.

Serum lipocalin-2 levels are elevated in obese patients. We assessed serum lipocalin-2 levels in polycystic ovary syndrome (PCOS) and the effects of weight loss or metformin on these levels. Forty-seven overweight/obese patients with PCOS [body mass index (BMI) >27 kg/m(2)] were instructed to follow a low-calorie diet, to exercise and were given orlistat or sibutramine for 6 months. Twenty-five normal weight patients with PCOS (BMI <25 kg/m(2)) were treated with metformin for 6 months. Twenty-five normal weight and 25 overweight/obese healthy female volunteers comprised the control groups. Ser…

Pharmacological management of obesity: Current landscape and emerging therapies.

Obesity is now recognized as a chronic, multifactorial, and progressive disease as opposed to mere excess of body weight. It significantly increases the risk of type 2 diabetes, cardiovascular disease, metabolic-associated fatty liver disease, and mental health disorders. 2022 data reveal that nearly 3 billion individuals worldwide were living with either obesity or overweight. Pharmacological therapy has evolved remarkably. Earlier medications targeted primarily the brain's monoaminergic pathways but they were withdrawn due to safety concerns. Currently, six medications (Orlistat, Phentermine…

Effects of Passiflora incarnata Leaf Polyphenol Extract on High-Fat-Diet-Induced Metabolic Syndrome in Mice and Associated Changes in the Gut-Liver-Adipose Axis.

Metabolic syndrome, driven by obesity, poses a significant global health burden. P. incarnata leaf polyphenol extract (PP), a crude 70% ethanol extract, exhibits potential bioactive properties, yet its efficacy and underlying mechanisms against metabolic disorders remain unclear. This study investigated effects of this extract (PP) on high-fat diet (HFD)-induced metabolic syndrome in mice and explored possible involvement of the gut-liver-adipose axis. Thirty mice were divided into five groups: ND (normal diet), HFD, PC-HFD (HFD + orlistat), PP1 (HFD + 75 mg/kg/day PP), and PP2 (HFD + 150 mg/k…
