Coix lacryma-jobi root extract exhibits self-toxicity, with butyl palmitate being the primary toxic substance.

Coix lacryma-jobi growth is often inhibited by continuous cropping due to autotoxic effects. The autotoxic effects and decomposition of aqueous extracts from different parts of C. lacryma-jobi plants and impact of their decomposed matter are unclear. The present study tested aqueous extracts from shoots, roots, and decomposed residues at four concentrations to assess their autotoxic properties. The extracts and decomposition products inhibited seed germination and seedling growth in a concentration-dependent manner. At concentrations of ≥ 0.1 g mL-1, the extracts significantly suppressed seed

Fatty acid composition, total phenolic and total flavonoid contents, and antioxidant activity of Niger seed (Guizotia abyssinica) accessions collected from major producer areas of Ethiopia.

BACKGROUND: Oils from various sources are vital nutritional components with a variety of roles in our body. Niger seed (Guzoita abyssinica) is endemic to Ethiopia and is among the major oil seed crops grown in the country. The fatty acid composition and the concentration of other bioactive phytochemicals in it vary with species type, geographical origin, cultivation season, and varietal types. The present work investigated the fatty acid profile and the total phenolic (TPC), total flavonoid content (TFC) and antioxidant activity (AA) of Niger seed samples obtained from five different zones in

Inhibitory effect of FO-1561 (S-adenosyl-L-methionine sulfate tosylate) on phospholipase A2.

The mechanism of the protective effect of FO-1561 (S-adenosyl-L-methionine sulfate tosylate) on mitochondria was investigated using rat heart muscle. FO-1561 stimulated CO2 production from palmitic acid in the mitochondria, indicating an increase in energy production. FO-1561 inhibited mitochondrial swelling induced by Ca2+ added exogenously or by addition of snake venom phospholipase A2. Moreover, FO-1561 inhibited phospholipase A2 activity directly. These results suggested that FO-1561 blocked mitochondrial swelling by inhibiting endogenous phospholipase A2 activity and so preventing decreas

The effect of retinol deficiency on the metabolism of all-trans-retinyl acetate in rat testes.

Following intratesticular injection, the metabolism of all-trans-[11-3H] retinyl acetate was studied in rats fed a vitamin A-deficient diet supplemented either with retinyl palmitate (-A + RP) or retinoic acid (-A + RA). Analysis of testicular metabolites by HPLC at 6 h and 24 h demonstrated the hydrolysis of retinyl acetate to retinol, esterification of retinol to retinyl palmitate and the formation of trace amounts of retinoic acid and other metabolites in both groups of rats. Eight and nine metabolite peaks were present in the -A + RP and -A + RA groups, respectively. The HPLC profile was s

Sn-2 palmitate-enriched structured lipids attenuate early aging phenotypes in senescence-accelerated mice.

Aging is characterized by progressive cognitive decline and metabolic dysregulation, contributing to increased vulnerability to age-related disorders. β-position palmitic acid-containing structured lipids (sn-2 palmitate), naturally enriched in human milk fat, have been reported to modulate inflammation and lipid metabolism. However, whether sn-2 palmitate-rich lipids exert beneficial effects beyond early development, particularly in the context of aging-related decline, remains unclear; further, their effects in aged populations or senescence models remain largely unknown. Therefore, we inves

Palmitate enhances MSC immunomodulation of human macrophages via the ceramide/CCL2 axis in vitro.

BACKGROUND: The immunomodulatory function of human mesenchymal stromal cells (MSCs) strongly depends on external factors; such as cytokines and other signalling molecules encountered in the disease microenvironment. An insufficiently inflammatory environment can fail to activate MSCs, and certain signals can impair their function. Obesity is on the rise worldwide, making it an additional factor to be considered prior to MSC therapy, as the microenvironment presents its own challenges. Elevated levels of serum free fatty acids, specifically palmitate, have the potential to affect MSC therapy. P

An Investigation of Volatile Flavor Compounds and Lipolysis-Oxidation in Coppa as Affected by the Inoculation of Coagulase-Negative Staphylococcus during the Air-Drying Stage.

This study aimed to explore the effects of coagulase-negative Staphylococcus inoculation on flavor generation and lipolysis-oxidation in Coppa. Acid lipase, neutral lipase, phospholipase, and lipoxygenase (LOX) activities, as well as free fatty acids, volatile compounds, and sensory evaluation, were determined during the fermentation and air-drying processes of Coppa over 40 days. Staphylococcus carnosus and Staphylococcus xylosus or a combination of both strains were selected for this study, and natural fermentation was treated as a control. The results showed that Staphylococcus inoculation

Fatty acid metabolism changes in association with neurobehavioral deficits in animal models of fetal alcohol spectrum disorders.

Fetal alcohol spectrum disorders (FASD) show behavioral problems due to prenatal alcohol exposure (PAE). A previous study reports changes in gene expressions linked to fatty acid (FA) metabolism in the cerebral cortex of the PAE mouse model. We find an increase of palmitic acid and arachidonic acid in phospholipid in the cerebral cortex of PAE at postnatal day 30. The increase of palmitic acid is consistent with increase of the producing enzyme, Fasn (fatty acid synthase). Decrease of 26:6 FA is also consistent with the increase of the enzyme which uses 26:6 as a substrate for making very long

Proton leak induced by reactive oxygen species produced during in vitro anoxia/reoxygenation in rat skeletal muscle mitochondria.

Superoxide anion generation and the impairment of oxidative phosphorylation yield were studied in rat skeletal muscle mitochondria submitted to anoxia/reoxygenation in vitro. Production of superoxide anion was detected after several cycles of anoxia/reoxygenation. Concomitantly, a decrease of state 3 respiration and phosphorylation yield (ADP/O) were observed. The latter resulted from a proton leak. The presence of palmitic acid during anoxia/reoxygenation cycles led to a dose-dependent inhibition of superoxide anion production together with a partial protection of the ADP/O ratio measured aft

Palmitate inhibits insulin gene expression by altering PDX-1 nuclear localization and reducing MafA expression in isolated rat islets of Langerhans.

Abnormalities in lipid metabolism have been proposed as contributing factors to both defective insulin secretion from the pancreatic beta cell and peripheral insulin resistance in type 2 diabetes. Previously, we have shown that prolonged exposure of isolated rat islets of Langerhans to excessive fatty acid levels impairs insulin gene transcription. This study was designed to assess whether palmitate alters the expression and binding activity of the key regulatory factors pancreas-duodenum homeobox-1 (PDX-1), MafA, and Beta2, which respectively bind to the A3, C1, and E1 elements in the proxima

Targeting Insulin Resistance, Reactive Oxygen Species, Inflammation, Programmed Cell Death, ER Stress, and Mitochondrial Dysfunction for the Therapeutic Prevention of Free Fatty Acid-Induced Vascular Endothelial Lipotoxicity.

Excessive intake of free fatty acids (FFAs), especially saturated fatty acids, can lead to atherosclerosis and increase the incidence of cardiovascular diseases. FFAs also contribute to obesity, hyperlipidemia, and nonalcoholic fatty liver disease. Palmitic acid (PA) is human plasma's most abundant saturated fatty acid. It is often used to study the toxicity caused by free fatty acids in different organs, including vascular lipotoxicity. Fatty acid overload induces endothelial dysfunction through various molecular mechanisms. Endothelial dysfunction alters vascular homeostasis by reducing vaso

Identification by GC-FID and GC-MS of amino acids, fatty and bile acids in binding media used in works of art.

GC-FID was used as single methodology for the identification and differentiation of proteins, lipids and ox bile from binders used in artistic paintings. The samples were hydrolyzed by HCl. Subsequently, the simultaneous formation of volatile derivatives of the amino, fatty and bile acids with ethyl chloroformate was performed quickly and safely in an aqueous medium. The derivatives were separated by capillary GC and characterized by GC-MS. The ageing of drying oils was studied, identifying pelargonic acid among other degradation products. Proteinaceous and lipoid binding media were characteri

Study on the antiinflammatory activity of methanol extract from seagrass Zostera japonica.

Methanolic extracts from the seagrass Zostera japonica were extracted successively using n-hexane (n-C(6)H(14)), dichloromethane (CH(2)Cl(2)), ethyl acetate (EtOAc), and water to give the n-C(6)H(14) (16.8%), CH(2)Cl(2) (40.6%), EtOAc (34.1%), and H(2)O (8.5%) soluble fractions, respectively. We have demonstrated that the hexane fraction has the highest capacity to inhibit proIL-1beta expression as compared to other fractions in lipopolysaccharides (LPS)-stimulated J774A.1 murine macrophages. Further analysis of the composition and antiinflammatory activity of the subfraction H5 from hexane fr

β cells keep bad epigenetic memories of palmitate.

Palmitic acid, or hexadecanoic acid, a 16-carbon saturated fatty acid (FA), accounts for approximately 38% of the total circulating FA in lean or obese humans. In an article published in BMC Medicine, Hall et al. report that cultured islets from healthy donors, when exposed to palmitate, undergo changes in CpG methylation that are associated with modifications of expression in 290 genes. Their results provide a first look at the mechanisms used by the endocrine pancreas of humans to keep a durable genomic imprint from their exposure to FA that can influence gene expression and possibly cell ph
