Detection of Clonorchis sinensis in stool samples using real-time PCR.

Human clonorchiasis, caused by infection with the trematode Clonorchis sinensis, is a common health problem in East Asia. In an attempt to develop a new, sensitive method for the diagnosis of the disease, the use of a real-time PCR (targeting the internal-transcribed-spacer-2 sequence of the parasite) to detect C. sinensis-specific DNA in faecal samples has recently been evaluated. The PCR-based assay, which included an internal control to detect any inhibition of the amplification by faecal constituents in the sample, was performed on stool samples and on DNA controls representing a wide rang

Fecal egg output in relation to worm burden and clinical features in human opisthorchiasis.

A study of the relation of fecal egg excretion to worm burden and clinical features was carried out in 45 opisthorchiasis patients who had no signs of biliary obstruction. The fecal egg excretion was consistent and correlated with the worm burden. Although there was no definite association between clinical signs and intensity of infection, mild hepatomegaly and thickened wall or dilatation of the gallbladder were found more commonly in heavily infected patients. Eosinophilia was observed more often than previous reports. Concomitant parasitic infections were found in 82% of the patients. After

Epidemiological study on factors influencing the occurrence of helminth eggs in horses in Germany based on sent-in diagnostic samples.

Gastrointestinal nematodes are ubiquitous parasites of grazing equines with Parascaris spp., and strongyles being the most relevant ones regarding the prevalence and potential disease severity. Despite their importance, epidemiological data regarding the presence and egg-shedding intensities of these parasites are scarce. Data from 1067 horse samples collected on German horse farms initially to compare diagnostic methods were used for epidemiological analyses. Due to its higher sensitivity, presence/absence data were based on a combined sedimentation/flotation technique while faecal egg counts

Impacts of targeted selective treatment on performance, nematode faecal egg counts and anthelmintic use in sheep: A meta-analysis.

Gastrointestinal nematode (GIN) infections of ruminant livestock have traditionally been controlled through regular, "suppressive", treatment with anthelmintic drugs, but this has resulted in the evolution of anthelmintic resistance and reduced efficacy. There is now a critical need to reduce anthelmintic use to maintain anthelmintic efficacy by creating refugia where susceptible GIN reside. Targeted selective treatment (TST) strategies only treat animals that need it, ensuring animals affected by GIN maintain their performance, while animals unaffected by GIN are not treated unnecessarily. Low efficacy of levamisole for Ascaridia galli control in experimentally infected poultry: first report in Cajamarca (Peru).

Chickens play an essential role in the global economy and household nutrition. The farming system, particularly extensive or backyard production, affects their productivity due to intestinal parasites, with Ascaridia galli being the most prevalent. Various active compounds are utilized to mitigate its impact; however, improper administration frequently leads to resistant parasites. In some instances, efficacy assessments are not conducted, leaving the effectiveness of different drugs uncertain. This study aimed to evaluate the clinical and controlled efficacy of levamisole in poultry experimen

ASCARIS SUUM EGG RECOVERY FROM SLUDGE SAMPLES AFTER PHASE EXTRACTION.

Some helminth test methods for sanitation samples include a phase extraction step to reduce lipid content and final pellet size before microscopy. Hydrophilic and lipophilic solutions are used to create 2 phases, with a plug of organic material or debris in between, whilst eggs are supposedly compacted at the bottom of the test tube. We tested 10% formalin, acetoacetic buffer, and acid alcohol as the hydrophilic solutions, and ethyl acetate and diethyl ether as the lipophilic solvents for egg recoverability from water, primary sludge, and fatty sludge. Normally, the supernatant and debris plug

Improvement of the Diagnostic Method for the Detection of Parasite Eggs Using the Formalin Ethyl Acetate Concentration Technique.

PURPOSE: The formalin-ethyl acetate (FEA) concentration method is commonly used in routine clinical practice to detect parasite eggs in feces. This procedure involves extraction of oil with the organic solvent ethyl acetate (EA), which reduces fecal sediment and provides a cleaner background for microscopic analysis. However, clinically, some sediment failed to float after EA treatment. METHODS: Hexane, commonly used in the food oil extraction from oilseeds did not float the feces. Gas chromatography-mass spectrometry (GC-MS) analysis showed that neither the amount of the oil nor the classes

Comparison of the efficacy of macrocyclic lactone anthelmintics, either singly or in combination with other anthelmintic(s), in nine beef herds in southern NSW.

Anthelmintic resistance (AR) is a well-recognized challenge in farmed ruminants. The use of anthelmintics in combination is one of the strategies recommended to slow the rate of AR development. Two studies were undertaken in 2017 and 2019 to assess the efficacy of single-dose macrocyclic lactone (ML) anthelmintic and ML combination drenches. In total, 11 Faecal Egg Count Reduction Trials (FECRTs) were set up in 10 different beef herds, with results available from 10 of those FECRTs (9 herds). AR to a single ML anthelmintic was detected in all 9 herds, with resistance to Cooperia and Haemonchus

Efficacy of levamisole, ivermectin and moxidectin against Capillaria spp. in European hedgehogs (Erinaceus europaeus).

This randomised study aimed to assess and compare the efficacy of treatment protocols containing levamisole, ivermectin, or moxidectin against Capillaria spp. in naturally infected European hedgehogs (Erinaceus europaeus) presented to a British wildlife rehabilitation centre. Faecal analysis, consisting of wet mount and flotation, was performed for 229 hedgehogs weighing ≥200g. Animals testing positive for Capillaria spp. (81%), excluding pregnant females, were randomly allocated a treatment protocol. Initially, hedgehogs (n = 50) received one of six 'pilot' protocols, whereas the remaining an

Composite Fasciola hepatica faecal egg sedimentation test for cattle.

Options for diagnosing Fasciola hepatica infection in groups of cattle are limited. Increasing the opportunities for herd-level diagnosis is important for disease monitoring, making informed treatment decisions and for flukicide efficacy testing. The sensitivity of a simple sedimentation method based on composite faecal samples for the detection of fluke eggs in cattle was assessed through a combination of experimental and statistical modelling techniques. Initially, a composite sample method previously developed for sheep was used to investigate the sensitivity of composite sample testing com

Development of a sensitive method to extract and detect low numbers of Cryptosporidium oocysts from adult cattle faecal samples.

Cryptosporidium transmission studies to date have concluded that adult cattle are not a significant source of oocysts contributing to clinical cryptosporidiosis in calves on farm. However current methods of sample processing have been optimised for calf faecal samples and may be less sensitive when used on adult samples due to lower numbers of oocysts and larger size of samples. A modified and novel method of oocyst extraction and concentration was developed and applied in an experiment involving spiking adult cattle faecal samples with known concentrations of Cryptosporidium oocysts. The resu

The impact of baseline faecal egg counts on the efficacy of single-dose albendazole against Trichuris trichiura.

There is considerable variation in the efficacy of single-dose albendazole (400mg) against Trichuris trichiura across human trials. Factors contributing to this variation have not yet been identified. We assessed the impact of mean baseline faecal egg counts (FEC) on the efficacy of single-dose albendazole against T. trichiura in five previously conducted trials. Our results suggest that efficacy measured by reduction in mean FECs decreased significantly (p<0.0001) when mean baseline FECs increased, highlighting that this parameter should be considered as an important confounding factor for dr

A new miracidia hatching device for diagnosing schistosomiasis.

It is still imperative to develop a parasitological technique highly sensitive for diagnosing schistosomiasis in epidemiological and individual surveys. A simple and cheap hatching device with a collecting container was manufactured and tested under experimental conditions. Twelve Kato-Katz slides were performed as golden standard for comparison. Quantitative results can be carried out by counting miracidia in a plate and parasite load can be calculated (miracidia/gram of feces). Statistically significant values were higher in the hatching test. More sensitive results, with statistical signifi

Variability of egg excretion in Schistosoma mansoni infection in Ethiopia: a case report.

An Ethiopian boy, aged 18 years, and heavily infected with S. mansoni (1250 eggs per gram of faeces), gave stool sample for microscopy three times a day (at 9:00 am, 1:00 pm and 4:00 pm) for 5 consecutive days. Each time two slides were prepared by the modified Kato's thick-smear technique. The maximum egg count at each examination was converted to eggs per gram of faeces (EPG). There was no significant variability (at 10% level, F-value = 0.04) in in egg counts made at different times of the day. However, the coefficient of variations between the egg counts made on different days were fairly

Infection pattern of Cryptosporidium and Giardia in calves.

Faecal samples were collected weekly over a 3 month period from 0- to 20-week-old calves on an Ohio dairy farm, and examined for Cryptosporidium oocysts and Giardia cysts by a quantitative direct immunofluorescence assay. Oocysts and cysts were detected as early as 4 days of age. Shedding of Cryptosporidium oocysts peaked at 1 week and was low by 3 weeks. Some calves, however, continued to pass low numbers of oocysts. Shedding of Giardia cysts peaked at 2 weeks of age with high levels maintained until 7 weeks of age. Most calves continued to pass low numbers of Giardia cysts after weaning. Cum
