Recurrent Pregnancy Loss: A Couple-Based Framework for Integrating Paternal Assessment.

Background/Objectives: Recurrent pregnancy loss (RPL) has traditionally been investigated predominantly through maternal factors, while the clinical role of paternal assessment remains inconsistently defined. Current guidelines differ substantially regarding semen analysis, sperm DNA fragmentation (SDF), genetic testing, and referral for andrological evaluation. This review aimed to compare contemporary guideline recommendations, critically appraise the directness, prognostic value, and clinical utility of the supporting evidence, and classify paternal assessment strategies as routine, selecti

From sperm to pregnancy: insights into paternal regulation of fertility divergence in cattle.

Embryo development and pregnancy establishment require successful interaction between the oocyte and sperm and proper activation of developmental signaling pathways. In the bovine, the successful execution of this biological equation requires, on one hand, maternal determinants such as the intrinsic quality of the oocyte and molecules secreted by the endometrium epithelium critical to sustain embryo development. In contrast, paternal contributions encompass both visible sperm characteristics assessed by routine semen evaluation and hidden traits that escape conventional laboratory assessment o

Paternal folate deficiency induces hepatic insulin resistance in offspring mice.

BACKGROUND: Folate (vitamin B9) is a water-soluble vitamin necessary for one-carbon metabolism, supporting the synthesis, repair, and methylation of DNA. While maternal folate status is well-studied for its role in fetal development and metabolic programming, the impact of inadequate folate intake in males on offspring development and metabolic diseases remains poorly understood. This study investigates the effects of folate deficiency in male parents on developing hepatic insulin resistance in offspring, focusing on molecular and metabolic disruptions within the liver. METHODS: Three-week-ol

Association of Paternal Diabetes With Hemorrhagic Stroke in Bangladeshi Women: The MAGPIE Study.

BACKGROUND: Hemorrhagic stroke (HS) accounts for ~10% of all first-time strokes, with an increasing incidence in Asia and a scarcity of information available about sex-related differences. This study investigates the independent predictors of HS among Bangladeshi women. METHODS: The Multidimensional Approach of Genotype and Phenotype in Stroke Etiology (MAGPIE) study is a Bangladeshi observational study that recruited nationwide HS patients between 2022 and 2024. We utilized univariate analysis to identify risk patterns in the data sets, whereas a multivariate logistic regression (LR) analysi

Effects of paternal 5G RFR exposure on health of male offspring mice.

With the widespread application of 5G communication technology, the potential health risks of radiofrequency radiation (RFR) have been paid much attention. Prior studies have demonstrated that the testes are highly sensitive to RFR, and notably, paternal epigenetic modifications can be transmitted to offspring, impacting their reproductive and neurobehavioral phenotypes. To investigate the intergenerational effects of paternal exposure to 5 G RFR on male offspring health, 7-8-week-old male C57BL/6 mice were randomly divided into Sham group and 4.9 GHz RFR group (16 mice per group). The mice in

A paternal lactate dehydrogenase critically enhances male gametogenesis and malaria transmission.

Malaria blood stage parasite development relies on glycolysis to generate ATP, which requires pyruvate to lactate conversion by an essential lactate dehydrogenase enzyme (LDH1). Conversely, parasites developing in the mosquito employ mitochondrial chemiosmosis for ATP production. The source of ATP during transition from vertebrate to insect is less clear; gametes form in the mosquito midgut lumen within minutes of gametocyte ingestion, and while female gametes possess a mitochondrion, this organelle is absent from male gametes (microgametes). Here, we investigate a second LDH enzyme (LDH2) fou

Assisted reproduction with advancing paternal and maternal age: an Ethics Committee opinion.

Ethical considerations for developing policies regarding providing assisted reproductive care to individuals of advanced reproductive age are complex; considerations include both reproductive autonomy and the well-being of the potential offspring. This document replaces the previously published document titled "Oocyte or embryo donation to women of advanced reproductive age: an Ethics Committee opinion" (Fertil Steril 2016;106:e3-7).

Clinical and Molecular Presentation of a Patient with Paternal Uniparental Isodisomy of Chromosome 16.

Uniparental disomies (UPDs) are among the causes of imprinting disorders. Specific phenotypes of most causative UPDs have been described. Here, we describe the case of a 2-year-old female patient who presented a syndromic phenotype. Chromosomal microarray analysis revealed UPD of the whole chromosome 16. Microsatellite analysis demonstrated paternal origin of the UPD and its isodisomic pattern (UPiD (16) pat). Mosaic trisomy 16 was not detected using the FISH method. Whole-exome sequencing revealed no pathogenetic genetic variants sufficient to explain the syndromic phenotype nor unmasked path

Paternal valproate use and impact of shared genetic susceptibility on child neurodevelopment.

Paternal use of valproate during spermatogenesis has been associated with increased risk of neurodevelopmental disorders (NDDs) in offspring, yet the role of genetic confounding is unclear. Using data from the Norwegian Mother, Father, and Child Cohort Study (MoBa), we assessed genetic susceptibility to epilepsy, ADHD and autism spectrum disorders (ASD) in fathers with epilepsy treated with valproate (n = 41), lamotrigine or levetiracetam (n = 37), other anti-seizure medications (ASMs; n = 80), and healthy controls (n = 54,752). Fathers using valproate had significantly higher polygenic risk s

Shared Death Anxiety: Obsessive-Compulsive Disorder and Advanced Paternal Age.

Obsessive-compulsive disorder (OCD) often emerges during childhood, presenting as distressing obsessions and compulsions that interfere with daily functioning. Advanced paternal age (APA) introduces unique psychosocial dynamics within families, potentially exacerbating stress, anxiety, and intergenerational emotional challenges. This case report examines the presentation of OCD in a 10-year-old boy, characterized by pervasive death anxiety shared with his 83-year-old father, whose advanced age intensifies existential concerns. The report delves into the complex interplay between APA, the child

Maternal androgen exposure induces intergenerational effects via paternal inheritance.

IN BRIEF: Activation of TLR2/TLR1 alters in vitro formation of capillary-like structures and induces inflammatory processes in ovine luteal endothelial (OLENDO) cells. ABSTRACT: Postpartum bacterial infections of the uterus affect uterine physiology and ovarian activity, causing fertility problems. The outer membrane component of Gram-negative bacteria, lipopolysaccharide, is involved in the initiation of the local inflammatory processes, and other bacterial toxins, particularly lipopeptides, have also been shown to be potent cytokine inducers, acting via Toll-like receptor-2 (TLR2). However,

Anthropogenic noise disrupts early-life development in a fish with paternal care.

Anthropogenic noise is a global pollutant but its potential impacts on early life-stages in fishes are largely unknown. Here, using controlled laboratory experiments, we tested for impacts of continuous or intermittent exposure to low-frequency broadband noise on early life-stages of the common goby (Pomatoschistus microps), a marine fish with exclusive paternal care. Neither continuous nor intermittent noise exposure had an effect on filial cannibalism, showing that males were capable and willing to care for their broods. However, broods reared in continuous noise covered a smaller area and c

High paternal homocysteine causes ventricular septal defects in mouse offspring.

Maternal hyperhomocysteinemia is widely considered as an independent risk of congenital heart disease (CHD). However, whether high paternal homocysteine causes CHD remains unknown. Here, we showed that increased homocysteine levels of male mice caused decreased sperm count, sperm motility defect and ventricular septal defect of the offspring. Moreover, high levels of paternal homocysteine decrease sperm DNMT3A/3B, accompanied with changes in DNA methylation levels in the promoter regions of CHD-related genes. Folic acid supplement could decrease the occurrence of VSD in high homocysteine male

Paternal thrombophilia and recurrent implantation failure: an exploratory case-control study.

OBJECTIVE: Many pieces of literature have reported that inherited and acquired thrombophilia might be a risk factor for recurrent implantation failure (RIF), however, most studies have only focused on RIF patients and not their male partners. We studied the possible association of paternal thrombophilia with RIF risk. METHODS: Forty-two male partners aged 20-45 suffered from RIF compared with 42 males from couples with at least one successful pregnancy. All participants were investigated for thrombophilia markers. RESULTS: The prevalence of coagulation Factor V activity was significantly hig

The Dutch Pregnancy Drug Register: Suitable to Study Paternal Drug Exposures?

Paternal medication use around the time of conception is common, but information about its effects on pregnancy outcome and the health of the child is generally limited. The aim of this study is to examine the feasibility of studying paternal exposure in the Dutch Pregnancy Drug Register by using immunosuppressants as a proof of concept. In 113 of 15,959 pregnancies, long-term paternal immunosuppressant use was reported 3 months before conception. In total, 134 immunosuppressants were used. Pregnancy outcome was known for 54 cases and was in accordance with previous findings. Two spontaneous a
